COVID-19 infections varied in the clinical aspect from mortality to asymptomatic episodes. The symptoms included fever, cough, difficulty breathing, loss of mobility, and chest pain. On the other hand, the patients who passed away from COVID-19 showed multiple organ failure, respiratory dysfunction, and disseminated coagulations throughout the body. D-dimer is a biomarker implemented in blood clotting and is commonly elevated in patients with thrombotic complications. This review sheds light on the correlation between gene mutations in the coagulopathy cascade among COVID-19 patients with high D-dimer levels. The elevated D-dimer levels are significantly associated with mutations in genes involved in the coagulopathy cascade. The findings suggest that these mutations may play an important role in developing thrombotic problems in COVID-19 patients. As a result, understanding the genetic basis of thrombotic development in COVID-19 could lead to new ways of avoiding and treating thrombotic problems.","PeriodicalId":15126,"journal":{"name":"journal of applied pharmaceutical science","volume":"175 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"76765014","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Agelas sp. , Ircinia sp., and Petrosia weinbergi were collected off the coast of Iligan Bay, Misamis Oriental, Philippines. The antiproliferation, proapoptotic, and antimigration properties of their hexane (H), dichloromethane (DCM), and methanol: water (MW) extracts against Human colorectal carcinoma cell line (HCT116) carcinoma cells were investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), Caspase-Glo ® 3/7, and in vitro scratch wound assay, respectively. The extracts exhibited cytotoxicity against HCT116. The viability of the treated cells was significantly lower ( p < 0.05) than the negative control. Treatment of the HCT116 with MW and DCM extracts of P. weinbergi and DCM extract of Ircinia sp . resulted in a statistically higher ( p < 0.05) luminescence than the negative control. Relative to dimethyl sulfoxide, the extracts increased caspase-3/7 activity more than onefold. This suggests the proapoptotic activity through the activation of the caspase-3/7. Lastly, the MW extract of Ircinia sp . and the H extract of P. weinbergi significantly ( p < 0.05) inhibited the migration of HCT116 after wounding. Thus, Ircinia sp . and P. weinbergi were found to show potential antiproliferative, proapoptotic, and antimigration properties against HCT116. Further investigation of compounds and the mechanism of action involved in their anticancer property are thus recommended.","PeriodicalId":15126,"journal":{"name":"journal of applied pharmaceutical science","volume":"21 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"75211089","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Six fungal endophytes were identified as Penicillium steckii AAB-01, Nemania bipapillata AAB-02, Xylaria feejeensis AAB-03, Hypomontagnella monticulosa AAB-04, Daldinia eschscholtzii AAB-05, and Phyllosticta capitalensis AAB-06. All of the isolated endophytic fungi were subjected to fermentation on rice media, followed by extraction with ethyl acetate. When tested for antibacterial activity, P. steckii AAB-01 extract showed the most potent inhibition against Staphylococcus aureus ATCC 6538 and Staphylococcus epidermidis ATCC 12228. Toxicity screening employing brine shrimp lethality test (BSLT) revealed potential toxicity of P. steckii AAB-01 and X. feejeensis AAB-03 extracts. Further investigation showed P. steckii AAB-01 extract had the highest inhibition toward MCF-7 cells, while D. eschscholtzii AAB-05 extract revealed the strongest cytotoxicity against 4T1 cells. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis tentatively identified five compounds, where indole acetic acid, O -acetylharmol, and oxindole I were suggested as the major metabolites in P. steckii AAB-01 extract, while 6-hydroxymelatonin and 2-methyl-6-pentadecylpyridine were suggested as the major metabolites in D. eschscholtzii AAB-05 extract. This is the first report on LC-MS/MS identification of the main metabolites from bioactive extracts of P. steckii AAB-01 and D. eschscholtzii AAB-05 isolated from the leaves of A. bunius , which might contribute to the antibacterial and cytotoxic properties of the extracts. Thus, a detailed investigation of antibacterial and cytotoxic metabolites from these endophytes merits further studies.","PeriodicalId":15126,"journal":{"name":"journal of applied pharmaceutical science","volume":"37 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"75429989","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Urb. is a potential plant to be developed as a raw pharmaceutical active ingredient. This plant can be utilized as raw materials for herbal medicine production. A green solvent is one of the innovative approaches to developing and utilizing natural products as a source of raw materials. This study aims to design and optimize the microwave-assisted extraction (MAE) of polyphenols from E. bulbosa bulbs using choline chloride- sorbitol-based natural deep eutectic solvent (NADES). The MAE method with choline chloride and sorbitol as the deep eutectic solvent composition was used to extract polyphenols content from E. bulbosa . Experimental design with some independent variables was involved, including NADES (choline chloride-sorbitol) ratio, liquid-solid ratio, microwave power, and extraction time. Response surface methodology with the Box-Behnken Design (using Design-Expert v12 program) was used to optimize the extraction condition based on the total polyphenols content (TPC) as a dependent variable. TPC value was determined using the Folin–Ciocalteu reagent and measured using a UV–Vis spectrophotometer at 761 nm. According to the study results, the optimum MAE condition was obtained at 1:1 g/g NADES (choline chloride-sorbitol) ratio, 5 minutes extraction time, 20%W microwave power, and 12:1 ml/g liquid-solid ratio. A 50 g sample was extracted under the ideal MAE conditions for the scale-up confirmation test, yielding a TPC value of 39.88 mgGAE/g sample. The MAE of polyphenols with choline chloride-sorbitol-based NADES produced a higher TPC as a target secondary metabolite and was conducted efficiently, rapidly, and in an environment-friendly way.","PeriodicalId":15126,"journal":{"name":"journal of applied pharmaceutical science","volume":"196 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"72744565","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In this study, the optimum nutritional and physical growth preferences of P. sanguineus in submerged conditions were established and the biological activities of ethanol extract of its mycelia were determined. The highest yield (100.13 mg/30ml) was produced in rice bran broth with a wide range of pH concentration. Biomass production was favored under 28°C, in lighted and agitated conditions with 147.20 mg/30 ml, 168.47 mg/30 ml, and 413.30 mg/30 ml, respectively. The extract inhibited the growth of Staphylococcus aureus while no antibacterial activity was observed in Escherichia coli . It was found toxic to brine shrimp with an LC 50 value of 190.55 µg/ml. Exposure of zebrafish embryos to the extract caused delayed growth resulting in a lower hatching rate. Growth retardation was the most noticeable teratogenic effect of the extract after 48 hours and high mortality rates were observed in high extract concentrations. This study suggests that the optimum biomass yield of P. sanguineus can be achieved when incubated under the most favorable conditions . Mycelia of P. sanguineus could be a valuable resource of antibacterial and cytotoxic compounds.","PeriodicalId":15126,"journal":{"name":"journal of applied pharmaceutical science","volume":"144 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"77519198","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
