Major concerns with the use of herbal preparations and their application as functional ingredients are about their stability, solubility, and bioaccessibility. Crocus sativus is one of the crucial nootropic traditional herbs, rich in potential phytochemicals used for the treatment of long-standing disorders. Many studies have suggested that the bioactive present in C. sativus extract may undergo enzymatic degradation in the intestine and hence less bioavailable at the site of action. A novel drug delivery approach to herbal preparation may combat the associated challenge. The present research focuses on the feasibility of developing w/o nanoemulsion incorporating the carotenoid-enriched C. sativus extract by spontaneous emulsification method. Critical process variables were optimized using 32 factorial design with Design of Expert software. The optimized formulation of w/o nanoemulsion comprising 9 mg/ml of C. sativus extracts with 12% internal phase, 21% S-mix, and 67% oil phase produced a globule size of 94.34 nm. The total content present in the nanoemulsion was 83% bioactive determined by the High Performance Liquid Chromatography- Photo Diode Array (HPLC-PDA) method. Further characterization, the Transmission Electron Microscopy (TEM) images showed the overall nanoemulsion system morphology with uniform distribution of globules of the internal phase. The in vitro release of bioactives from developed nanoemulsion showed an initial burst release of 60% at 1 hour following slow drug release. This study aims to examine the extraction and characterization of collagen from common carp (Cyprinus carpio L.) scales, as an alternative source in dentistry. The samples used in this study consisted of common carp (C. carpio L.) scales. In the first step, common carp scales were acid-extracted and spray-dried to isolate collagen. After the collagen had been isolated, it was tested for moisture content, ash, acidity, and functional groups, with Indonesian National Standard (SNI) 8076:2014 as the reference standard. The characterization parameters of the common carp scale collagen powder revealed an 11% yield, 3.66% moisture content, 0.32% ash content, and a pH of 7.38. Furthermore, fourier transform infrared spectroscopy spectra confirmed the presence of a triple helix structure. The extraction of collagen from common carp scales using the acetic acid method and spray drying technique successfully adhered to the composition standards specified by the SNI 8076:2014. In this study, we aimed to prepare a novel recombinant protein polyprotein (P1) and 3D polymerase (3D p ° l ) protein of EV-A71B5 and determine mice immunogenicity and neutralization activity against EV-A71 of the B5 subgenotype commonly found in Thailand. Using a dual promoter system and investigating its expression in Sf9 cells, we constructed a novel recombinant protein containing P1 and 3D p ° l protein. The purified P1-3D p ° l was observed by western blotting and transmission electron microscopy (TEM) to determine the particle size. Furthermore, we determined the immunogenicity and neutralization activity against EV-A71 of the B5 subgenotype using concatenation of Bagg and Albino (BALB/c) mice. The results revealed that P1-3D p ° l was expressed in Sf9 cells. We used TEM to visualize the particle size of P1-3D p ° l to be approximately 33 nm. P1-3D p ° l had the potential to elicit the production of immunoglobulin G and immunoglobulin M antibodies and the T helper type 1-dominant cytokine interferon-γ after immunizing BALB/c mice and inducing neutralization antibodies against EV-A71B5. A prospective cross-sectional study was conducted with 165 patients to identify the health-related QOL in CLD, for 6 months. The QOL of CLD patients was assessed using the World Health Organization quality of life-BREF questionnaire and the severity of the disease was assessed using Child-Pugh Model for End-Stage Liver Disease (MELD) score. The incidence of CLD was found to be high in males (89.09%). While assessing, the overall QOL was identified as 50.96 ± 15.77. The highest score was observed in the social relationships domain (58.33 ± 19.34) followed by environment (54.74 ± 16.74), psychological (46.70 ± 19.81), and physical health (44.08 ± 18.64). The socio-demographic factors associated with QOL include body mass index, education, alcohol consumption, smoking, and tobacco. In the Child-Pugh MELD Score, the QOL was higher in class A (Good hepatic function) with a MELD score of 6–9 (1.9% mortality), the mean score being 60 ± 21 in contrast with the other classes. From the study, it is evident that CLD has a significant impact on the QOL. The extract of wampee peel (WP) exhibits several medical properties, especially antioxidant effects. We aimed to explore the antihyperlipidemic, hypoglycemic, and antioxidant effects of WP peel extract in an experimental T2DM rat model. T2DM was induced in the experimental groups by a high-fat diet and streptozotocin injection (HFD/STZ). Rats with a fasting blood glucose level above 200 mg/dl were considered diabetic and were separated into six groups (control, diabetic, diabetic treated with WP peel extract at 200, 400, or 600 mg/kg, and diabetic treated with metformin at 250 mg/kg). After 4 weeks of treatment, blood samples were collected for analysis. HFD/STZ-induced rats exhibited hyperglycemia, dyslipidemia, increased HbA1c, and increased lipid peroxidation, but reduced serum antioxidant enzyme activities. However, these changes were attenuated in T2DM rats with the highest dose of WP peel extract. However, there are limited data about vaccine willingness among children, which is largely confined to parents' opinions. With the emergence of new variants affecting this age group, children's opinions on COVID vaccination are important. This study aimed to explore children's willingness and opinions regarding COVID-19 vaccination. Focus group discussions, including children, were carried out in different Jordanian schools. Purposive sampling was implemented to select students of different socioeconomic statuses. A total of 54 school students aged 6–14 years from three schools participated in nine focus groups. Overall, participating children were willing to take the COVID-19 vaccine, and three overarching themes described their view regarding the vaccine: benefits, motivators, and hesitancy and barriers. Participating children reported that the vaccine could be beneficial as it protects them from being infected, and they are motivated to take it as it has been approved for children in other countries. However, some children were hesitant regarding taking the vaccine as it could be harmful, and they believed that COVID-19 is not severe in children. In conclusion, this study shows a clear willingness among participants from primary and secondary schools to take the COVID-19 vaccine. Owing to the purported pharmaceutical activities of plant-derived secondary metabolites, we assessed the inhibitory potentials of the Philippine native plant Phaeanthus ophthalmicus tetrahydrobisbenzylisoquinoline alkaloidal constituents tetrandrine ( 1 ) and limacusine ( 2 ) against enzymes implicated in type 2 diabetes (T2D) and obesity such as α-glucosidase, dipeptidyl peptidase-IV (DPP-IV), porcine pancreatic lipase (PPL), and human monoacylglycerol lipase (MAGL) using in vitro experiments and molecular docking. Both alkaloids 1 (IC50 = 2.29 μg/ml) and 2 (IC50 = 2.68 μg/ml) showed stronger inhibition against α-glucosidase compared to the drug control acarbose (IC50 = 4.12 μg/ml). Alkaloids 1 (IC50 = 4.92 μg/ml) and 2 (IC50 = 3.80 μg/ml) also exhibited better inhibitory activities against DPP-IV compared to the drug control sitagliptin (IC50 = 6.90 μg/ml). Molecular docking results revealed better binding propensities for both 1 and 2 onto the active pocket of α-glucosidase and DPP-IV compared to their respective control drugs. Meanwhile, alkaloid 2 showed better in vitro (IC 50 = 0.70 μg/ml) and in silico inhibitory activity vs PPL compared to orlistat. Both alkaloids 1 and 2 showed moderate bioactivity against MAGL. Both alkaloids were predicted to possess drug-likeness properties.
