This research aimed to obtain basic data on DNA polymorphism, genetic relationship, genetic similarity level, and molecular accession of Bali’s unique local mango. Sampling was conducted in four regencies in the Province of Bali. DNA preparation, PCR, and microsatellite analysis were carried out at the Laboratory of Genetics and Plant Breeding, Faculty of Agriculture, Gadjah Mada University. It found 44 accessions of mangoes in Bali. The test results with 10 pairs of SSR markers showed that all primers produced polymorphic loci. There were 825 amplified DNA bands. Primer of AY31 produced the highest number of loci, which were 14 loci, while AY21 produced the fewest loci, i.e., 3 loci. Forty-four mango accessions showed a genetic similarity coefficient of 0.27 to 0.97. At a coefficient of 0.27, accessions were divided into 2 major groups: group A and group B. Group A consisted of two accessions, namely, KRA-005 and BDG-006 (Mangifera foetida Lour.), which had a similar coefficient of 0.657. Group B was divided into two smaller groups, namely, groups B1 and B2, at a similarity coefficient of 0.342. Group B1 consisted of 39 accessions, while group B2 consisted of 3 accessions. The application of indigenous endomycorrhizal biofertilizer combined with Rhizobium is expected to be able to improve nitrogen and phosporus uptake so as to increase the productivity of soybeans in dry land. The experiment used two-factor factorial randomized block design. The first factor was dose of endomycorrhizal spores of Glomus that consisted of four levels, namely, treatment without endomycorrhiza inoculation, dose of 50 endomycorrhizal spores, dose of 100 endomycorrhizal spores, and dose of 150 endomycorrhizal spores. The second factor was inoculation of Rhizobium inoculum that consisted of two levels, namely, treatment without Rhizobium inoculation and Rhizobium inoculation. The T-DNA harbored the SoSPS1 gene under the control of the CaMV 35S promoter from the cauliflower mosaic virus and contained the NPTII gene (kanamycin resistance gene) as a selectable marker for transformant selection. Generally, gene transformation in plants is carried out by tissue culture. However, tissue culture has several disadvantages such as its being time-consuming, its sometimes resulting in somatic mutations and somaclonal variations, and the requirement of sterile conditions in the procedure of gene transfer. In planta transformation is a useful system for those plants that lack tissue culture and regeneration system. The main function of in planta transformation is to recover the advantages of tissue culture as an efficient, quick method, including its ability to produce a large number of transgenic plants and to accumulate a high concentration of total soluble protein in short time. There are two procedures of in planta transformation for the seeds of citrus plants, namely “prick and coat” and “seed tip-cutting and imbibition”. In the prick and coat method, seeds are pricked on their entire surfaces and smeared with a suspension of Agrobacterium tumefaciens. In the seed tip-cutting and imbibition method, on the other hand, seeds are cut at the tip and soaked in a suspension of Agrobacterium tumefaciens. The leaves derived from seeds treatment were taken as samples for DNA extraction and PCR using primers of the NPTII gene (Forward: 5’-GTCATCTCACCTTCCTCCTGCC-3’; Reverse: 5’GTCGCTTGGTCGGTCATTTCG-3’). This research found that only the seed tip-cutting and imbibition plants amplified along the 550-bp band, while those of the prick and coat method did not. In this study, an innovation was carried out by trying to do colchicine treatment in hydrogen peroxide. In this in vivo induced mutagenesis research, colchicine in hydrogen peroxide was administered as mutagen on seeds three species of crops i.e. mutan M2 cowpea, yardlong bean, and common bean by soaking method. The results of this research showed that three times multistep mutant (M3) of cowpea was having mutant characteristics that the shape of its leaflet changed from rhombus to ovate. Besides, both yardlong bean varieties used in this research (BLR and KTR) showed indication of a mutant crop that having compound leaves with more than three leaflets in a stalk. Common bean of PTW variety was not attacked by any aphid but it was attacked by leafminer. The purpose of this study was to analyze limonene and other antioxidants content of the essential oils in the market compared to homemade virgin coconut oil. The method used was Gas Chromatography Mass Spectrometry (GC-MS) and Fourier-transform infrared (FTIR) Spectroscopy. The infrared spectrogram showed the presence of alkanes and esters respectively at wave numbers of 3000-2850 and 1750-1730 cm. A batch of 30 samples of Mongal cultivar tomato purchased at two markets in Ouagadougou, Burkina Faso was used as a matrix for the collection of LAB isolates. From a preliminary biochemical characterization, 97 isolates were collected based on their antimicrobial activity. Seventeen isolates (17.52 % of the isolates collected) displayed antimicrobial activities with an inhibitory zone higher than or equal to 12 mm of diameter, against one to three out to 16 pathogenic indicator strains used including bacteria, moulds and yeasts. Based on their phenotypic characteristics, the 17 isolates were regrouped and 12 representative isolates were used for the identifiation by 16S rDNA sequencing. The results showed that the 17 LAB isolates displaying antimicrobial activity, were capable of fermenting various carbohydrates, and three of them were capable of producing bacteriocin-like substances (BLS) against Salmonella infantis SKN 557 (2 isolates) and Pseudomonas aeruginosa ATCC (1 isolate). Fourtheen isolates showed inhibitory activity against both A. flavus and A. fumugatus. The isolates identified belong to the species Lactobacillus plantarum (50% of the isolates) , Lactobacillus fermentum (25%), Lactobacillus pentosus (8.33%), Lactobacillus plantarum/pentosus (8.33%) and Pediococcus acidilactici (8.33%) with a dominance of Lactobacillus genus which represents 91% of the identified isolates against 9% of Pediococus genus. The isolates revealed interesting properties and shoud be potential candidates to be used as starter cultures for food fermentation and preservation including fruits and vegetables products. The first year of study was conducted in Marga Tabanan Village. The field research was Randomized Block Designed (3 groups) with single factor of treatment. The tested treatment were 6 formulation of compound fertilizers and a control (unfertilized treatment). The compound fertlizers consisted of urea, SP-36, KCl, local compost, and dolomite in varying doses. The fertilizer formulations were applied on 21 agarwood trees that previously inoculated with mixed inoculant of Fusarium solani and Rhisopus sp. Three months after inoculation, the data from the following parameters were measured and statistically analyzed: plant height, stem circumference, sapwood weight, resin content and soil chemical properties.The results showed that the tested fertilizer compound had significant effect on plant height, sapwood weight, and resin rendement. No significant effect of fertilizer compound measured on the stem circumference. The highest sapwood weights was obtained on C treatment (14.39 g). The highest resin yield was obtained on B treatment (3.91%) which was relatively the same as that on C treatment (3.85%). The research was taken using purposive sampling technique and complete random program. Virus symptoms were observed on two potato fields. Serological test employed DAS-ELISA with PVX, PVS, PVY and PLRV antiserum. In vivo elimination virus were done by thermoteraphy in 37°C±1°C (4 days), 34°C±1°C (3 days) on 3 weeks and 33°C±1°C (4 days), 30°C±1°C (3 days) on 2 weeks. Results of the study at Pancasari Village Bali shown varied symptoms on potato leaves, such as mottle, mosaic, malformation, leafroll, witchesbroom. Through serological test, had identified viruses were 11.11% PVX, 55.56% PVS, 80% PVY and 11.11% PLRV which were infected farmer’s potato. Wirawan","doi":"10.24843/ijbb.2018.v05.i02.p04","DOIUrl":"https://doi.org/10.24843/ijbb.2018.v05.i02.p04","url":null,"abstract":"In the previous research, we obtained three catagories of M1 mutant cowpea namely 1) purple pod mutant cowpea, 2) green pod mutant cowpea which has three pods in a stalk, and 3) green pod mutant cowpea which has two pods in a stalk. All those three catagories of M1 mutant cowpeas were treated again with colchicine in a multistep mutagenesis proses in vivo. Then, we found changes in number of leaflet in a petiole to 4 leaflets (quadrifoliate) and to five leaflets in a petiole (pentafoliate). These changes are indications that beside as a chromosome multipying alkaloid, colchicine causes other genetic changes as well. Through this mutagenesis research, we created gigas mutant cowpea that having longer leaves, longer pods and heavier seeds.
