Trimethylation of histone H3 at lysine 27 (H3K27me3) is a key regulator in chromatin remodeling-controlled transcription. Focusing on the epigenetic mechanism, we aimed to investigate the expression of H3K27me3 in PCMC by immunohistochemistry. A retrospective cohort of PCMC patients from a tertiary hospital in Taiwan was enrolled to evaluate the clinicopathologic features, treatment outcome, and protein expression. Immunohistochemistry for H3K27me3 was performed on all PCMCs and a comparison group of colonic mucinous adenocarcinoma and pure mucinous carcinoma of the breast. The percentage of H3K27me3-negative tumor cells was calculated and analyzed. Three patients with PCMC were recruited. All PCMCs were solitary and slow growing, arising from the head-and-neck region. All PCMCs had tumor excision without local recurrence or metastasis. The loss of H3K27me3 expression was significant in PCMCs (mean ± standard deviation [SD]: 21.0% ± 6.6%) compared to other mucinous carcinomas (mean ± SD: 3.8% ± 1.7%) (P = 0.019). In conclusion, we report a reduction in H3K27me3 expression in PCMC. In contrast, H3K27me3 expression is retained or mildly reduced in other mucinous carcinomas. The effect of H2S on allergic contact dermatitis (ACD), an inflammatory skin disease that negatively affects the quality of life, is unknown. Objectives: We planned to investigate the antipruritic and anti-inflammatory effects of the H2S donor sodium sulfide (Na2S) in the experimental mouse model of contact hypersensitivity (CHS), which is widely used for ACD research. Methods: CHS was created in Balb/c mice using 1-fluoro-2, 4-dinitrobenzene. Na2S was administered systemically (0.2-2-20 mg/kg/i.p.) and locally (1-3-10 nmol/both ear/i.d.) at 3 h and 25 h after the challenge. Ear thickness and the number of scratches were determined at 24 h and 48 h following the challenge. Ear tissue and serum interferon-gamma, interleukin (IL)-2, IL-4, and IL-5 cytokine levels were evaluated by enzyme-linked immunosorbent assay (ELISA). H and E staining was performed for histopathological studies. CD4+ and CD8+ T cells located in the skin were examined by immunohistochemical staining. Results: Locally (1-3-10 nmol/ear/i.d., P < 0.001, P < 0.0001, P < 0.0001, respectively) and systemically (2–20 mg/kg/i.p., P < 0.01, P < 0.0001, respectively), Na2S administration decreased ear thickness dose dependently. Local (1-3-10 nmol/ear/i.d.) Na2S treatment decreased serum IL-2 levels (P < 0.01, P < 0.05, and P < 0.01, respectively). Na2S administered locally (3–10 nmol/ear/i.d., P < 0.05) and systemically (20 mg/kg/i. p., P < 0.05) decreased the number of CD4+ T lymphocytes. Despite recent advances in treatment, pemphigus is associated with high patient mortality and has a significantly negative impact on patients' quality of life and mental health. Validated scoring systems to accurately measure pemphigus severity are vital for the monitoring of disease activity, evaluation of treatment efficacy and trialing, comparison and introduction of new treatments. Sex hormones, a systemic factor, paradoxically promote and suppress hair growth in different sites of the body, leading to diseases such as hirsutism, androgenetic alopecia, and female pattern hair loss. The past decades have seen progress in the treatment of sex hormone-related hair diseases, but the pathogenesis of some of these diseases remains obscure and even controversial. We review the biological effects of major sex hormones on hair growth and summarize their known impacts. We discuss the different responses of animal and human HFs to sex hormones, summarize the pathogenesis of sex hormone-related hair diseases, and highlight the benefits of and controversies in the current management of these diseases. Objectives: The objective is to provide a novel direction for improving overall survival (OS) for CM. Methods: We obtained the gene sets related to ultraviolet (UV) reaction from MsigDB database and CM HTSeq-FPKM data from The Cancer Genome Atlas (TCGA). Gene set variation analysis was used to calculate the enrichment scores in each sample. DAVID and Gene Set Enrichment Analysis (GSEA) were used to explore the function of differentially expressed genes (DEGs) between cluster 1 and cluster 2. The ssGSEA was used to analyze the degree of immune infiltration in samples. Weighted gene co-expression network analysis (WGCNA), protein–protein interaction (PPI) network, and mutation analysis were performed to screen the DEGs related to UV response. Results: The samples were divided into the high activity of UV response (cluster 1) and low activity of UV response (cluster 2). We found that cluster 2 was related to poorer OS and had a higher reaction to UV response. Function analysis indicated that the DEGs are involved in angiogenesis, epidermal development, and inflammatory reaction. Furthermore, the cluster 2 had a higher degree of immune infiltration. The results of WGCNA indicated that the genes in the MEyellow module were highly related to UV response, which is involved in the process of angiogenesis, cell migration, and skin development. PPI and mutation analysis indicated that COL5A1 was the risk factor for CM.
