Ketta","doi":"10.17221/91/2022-cjas","DOIUrl":"https://doi.org/10.17221/91/2022-cjas","url":null,"abstract":"The aim of the present study was to compare carcass characteristics and meat quality of fast- (Ross 308), medium- (Hubbard JA757) and slow-growing (ISA Dual) chickens (n = 1 980). When the chickens reached an average live weight of 2 kg, 40 birds (males to females, 1 : 1) of each genotype were randomly selected and slaughtered for carcass analysis. Pectoralis major (PM) muscle samples were taken to determine the physical and chemical parameters of the meat quality. The fast- and medium-growing genotypes had higher (+3.24% and +3.84%, respectively) dressing out percentages than the slow-growing chickens. As expected, the breast percentage significantly decreased in the order of fast growth > medium growth > slow growth. The abdominal fat percentage was the lowest in Ross 308 chickens, but the abdominal fat percentage in JA757 and ISA Dual chickens did not differ. For edible organs, fast- and medium-growing chickens had heavier hearts and livers than slow-growing chickens. In contrast, ISA Dual chickens showed heavier gizzards. Genotype dramatically affected the chemical composition of PM muscle. The ISA Dual chickens exhibited a significantly higher proportion of dry matter and protein and lower ether extract and cholesterol content than Ross 308 chickens, with intermediate values for JA757 chickens. The ISA Dual group had a lower ash content than Ross 308 and JA757 groups. In terms of physical parameters, the ISA Dual chickens had higher lightness and yellowness and lower redness, pH 24, and cooking loss than those of the other genotypes. Concerning all genotypes, the shear force values increased from fast- to slow-growing chickens. In conclusion, this study found a difference between the genotypes. {"title":"A meta-analysis of heat stress in dairy cattle: The increase in temperature humidity index affects both milk yield and some physiological parameters","authors":"Ersin Kulaz, G. Our goal in the meta-analysis was to find the parameter(s) primarily affected under heat stress. A total of 16 studies with the temperature humidity index value higher than 72, which is an important factor in determining the effect of heat stress, were included in the meta-analysis. The variables of interest in the meta-analysis included: milk yield (kg/day), respiratory rate (breaths/min), rectal temperature (°C). In addition to the meta-analysis, principal component analysis (PCA) was also performed. In the meta-analysis, high variation or heterogeneity (I2 > 99%) was determined between the results of the studies. This may depend on many factors (climate, region, number of samples and management etc.). Heterogeneity is desirable in the meta-analysis, because it provides accurate and reliable interpretations of the variances of parameters. Due to high heterogeneity, the results of the studies were combined according to the mixed model. According to the mixed model and PCA results, a linear relationship was determined between the temperature humidity index and these physiological parameters. According to the meta-analysis, at THI > 72, the mean effect size of milk yield was 50%, and the effect sizes of respiratory rate and rectal temperature were approximately 65% and 38%. All three parameters have a significant effect under heat stress (P < 0.000 1). As a result, there is a linear relationship between temperature humidity index, milk yield and physiological parameters. Recent studies revealed that the secretion of leucine into sow milk from the mammary gland is significantly lower than the amount taken up by the mammary gland from the blood, suggesting the potential and fundamental role in udder metabolism. However, little is known about the underlying mechanism. In this study, porcine mammary epithelial cells (PMECs) were employed to investigate the effects of leucine on protein synthesis and mammalian target of rapamycin (mTOR) signalling. Results showed that leucine supplementation significantly increased the proliferation of PMECs in a dose-dependent manner from 0 to 1 mmol/l. Secretions of four casein subunits were significantly increased by addition of leucine. The phosphorylation of proteins related to mTOR signalling, including mTOR, eukaryotic translation initiation factor 4E-binding protein, and p70 ribosomal protein S6 kinase, were accordingly upregulated by leucine supplementation. Casein secretion was not affected by leucine treatment when mTOR signalling was inhibited by rapamycin. This suggests that leucine stimulates protein synthesis through the mTOR signalling pathway. Ma","doi":"10.17221/209/2021-cjas","DOIUrl":"https://doi.org/10.17221/209/2021-cjas","url":null,"abstract":"We investigated the effects of sweet potato vine silage (SPVS) supplementation on the growth performance, apparent digestibility and gut health of finishing pigs. 180 Bali Black pigs (Berkshire × Licha Black, with body weight of 74.54 ± 3.32 kg) were assigned to three groups: basal diet (Ctrl), Ctrl supplemented with 2.5% SPVS (Lspvs) and 5.0% SPVS (Hspvs). Animals were slaughtered after nine weeks of feeding. The results indicated that dietary SPVS supplementation improved average daily food intake and average daily gain. However, SPVS treatment decreased the apparent digestibility. Activities of antioxidant enzymes including total superoxide dismutase, total antioxidant capacity, glutathione peroxidase and catalase in the intestines of the Lspvs group were markedly upregulated. Concentrations of IL-1β and IL-6 were decreased and secretory immunoglobulin A was increased in Lspvs group. A significant increase (P < 0.05) of ileum diamine oxidase in Lspvs group was observed. The ileum villus height/crypt depth in the Hspvs group was significantly reduced. The ratio of Firmicutes to Bacteroidetes in the caecum contents of pigs was reduced, and the abundance of Lactobacillus was significantly increased. Specifically, Hspvs treatment markedly reduced the abundances of Proteobacteria. Candidate genes affecting IMF deposition were screened by comparing the RNA-Seq profiles of test (2% S. alopecuroides) and control (no S. alopecuroides) groups. The results showed that the intramuscular fat content of the diet supplemented with S. alopecuroides was significantly lower than that of the control group, and the intramuscular fat content decreased significantly with the increase of the dietary content of S. alopecuroides (P < 0.05). The highest IMF content of the control group (group A) was 4.21%, which was significantly higher than that of the other four experimental groups (B, C, D, E) (P < 0.05). Moreover, 66 genes were significantly upregulated and 202 genes were significantly downregulated in the test group compared to the control. Gene Ontology functional annotation revealed that the differentially expressed genes were closely related to plasma lipoprotein particle assembly, plasma lipoprotein particle organization, protein-lipid complex subunit organization, and other biological processes. KEGG pathway analysis showed that the differentially expressed genes belonged to 147 metabolic pathways. Finally, four differential genes which may be related to IMF deposition were screened out in PPAR signalling pathway, fatty acid biosynthesis, fatty acid degradation, and fatty acid metabolism. The quantitative analysis of four differential genes including ACSL3, PLIN2, ABCA1 and ANGPTL4 was carried out by RT-qPCR method, and the results were basically consistent with those of RNA-seq. Among them, the quantitative results of PLIN2 gene were slightly different from the sequencing results, but the difference was not significant, and they may be false positive. This research aimed to know the effects of genotype, days of lactation and climatic factors on the fatty acids (FA) profile of milk and particularly the content of CLA in milk fat. Seventeen first-calving milking cows in early lactation were used for the assessment of milk; 12 were Gyr and five were F1 (Holstein/Gyr) crosses. Sampling was carried out every 15 days, from the beginning to the end of lactation (300 days). Fatty acids were analyzed employing gas chromatography. The genotype did not influence the content of the fatty acid groups: saturated fatty acids (SFA), monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA) and CLA. The highest MUFA and PUFA contents were recorded at 100 days of lactation (32.334 and 3.553 g/100 g of FA, respectively), while SFA and CLA had their highest production at 200 days of lactation (63.238 and 1.378 g/100 g of FA, respectively). The topic relates to the variability of the feed value of maize being an important feedstuff in livestock nutrition. The nutritional values determined by pNIRS in comparison with the chemical analysis were higher (P < 0.001) in starch and ash content but lower in dry matter, neutral detergent fibre and crude protein (CP) content. The digestibility levels of neutral detergent fibre and the net energy of lactation as well as the potential milk production per hectare in relation to each tonne of dry matter were also lower. According to this result, it would be necessary to calibrate all tested indicators for a given spectrometer. However, the pNIRS results are useful for evaluating nutrient variability; the standard deviation of the values found in pNIRS was mostly lower than that determined chemically. There were established three groups of chickens for fattening with 80 individuals each (control group C and experimental groups). After the 34-day fattening period chicken breast and thigh muscles were analysed to find out the fatty acid composition. Lupin protein-based diets had a positive effect on the muscle quality of fattened chickens due to changes in fatty acid composition, compared to soya protein-based diets. The feeding of lupin-based diets to broiler chickens resulted in the reduction of saturated fatty acids (P ≤ 0.05) by 14% in LS50 group and 17% in LS100 group and increase of unsaturated fatty acids (P ≤ 0.05) by 58% in LS50 group and 90% in LS100 group in muscle fat. However, little attention is given to the excellent characteristics of this breed’s meat. We used label-free proteomics to analyse the total protein content in five different muscles, and 1 227 proteins were detected. Through sequential windowed acquisition of all theoretical fragment ions (SWATH), 16, 33, 49, 39, and 31 differentially expressed proteins were successfully detected in the five muscles. Protein–protein interaction network analysis of differentially expressed proteins revealed many strong interactions related to fatty acid beta oxidation and muscle development. Based on SWATH in five muscles, 25 differentially expressed proteins related to muscle development were detected, including seven muscle fibre structural proteins (ACTG2, ACTN4, TAGLN, MYL3, MYL1, MYL6B and MYH4). We used online databases to analyze the gene structure, chemical characteristics, and evolutionary relationship of Sox family genes through bioinformatics, and we studied the expression profiles and regulatory mechanisms of Sox family genes. A total of 29 rainbow trout Sox genes were identified. The phylogenetic analysis found that Sox genes of rainbow trout were clustered in seven subfamilies (B1, B2, C, D, E, F and H), and the gene structure of each subfamily was relatively conserved. Furthermore, Sox1, Sox4, Sox6, Sox8, Sox9, Sox11, Sox17, Sox18, and Sox19 developed into two copies, which might be the result of teleost fish-specific genome replication. Multiple HMG box domain alignments indicated that the motifs for all Sox sequences are conserved. Gene expression studies reveal that Sox expression is tissue-specific and that multiple Sox genes are involved in rainbow trout gonad and central nervous system development.
