The ones of most distributed viruses among cultivated raspberries are Raspberry leaf blotch virus (RLMV) and Raspberry leaf mottle virus (RLBV). The influence of these viruses on the raspberry crop yield is large and losses count in 30-50%. To detect the infected stock planting material or plants carrying infection in orchards, the sets of primer have been developed in this work. Capsid protein gene and nucleocapsid protein gene of RLMV and RLBV, respectively, were considered for design of primers. The specificity analysis confirmed absence of any off-target complementary of primers considering the nucleotide collection of NCBI. Eighty-five samples of raspberry were tested by developed primers. Four cultivars of pomological garden and one raspberry plant of wild population were positive for RLMV and RLBV, respectively. The sequencing analysis of RLMV CP and RLBV NP revealed, that homology with isolates from Bosnia and Herzegovina is higher compared to other isolates in NCBI.

Hemicelluloses are divided into three main types: xylans, mannans and arabinogalactans. Xylan is the second most abundant carbohydrate in nature after cellulose. The specific enzymes that hydrolyze xylan into xylooligosaccharides and D-xylose are xylanases. Prospective xylanases are enzymes derived from bacteria and mycelial fungi. Bacterial xylanases are characterized by high unitary activity, and additional manose chains can significantly increase the stability of the enzyme protein globule. Obtaining a glycosylated variant of bacillary xylanase, which are known for their high specific activity, seems promising.\u0000The Bacillus sonorensis T6 xylanase gene was cloned and expressed in the yeast Pichia pastoris. The recombinant xylanase was isolated and purified. The biochemical characteristics of the glycosylated recombinant xylanase were studied. It was found that the recombinant xylanase had maximum activity at 47-50°C and pH 6.0. The Km, Vmax and Kcat values are 3.037 ± 0.362 (mg/ml), 667.8 ± 31 (units/mg) and 100.3 ± 4.6 (s-1), respectively. The unitary activity of the recombinant enzyme is 873.8 units/mg. The glycosylated recombinant xylanase was found to have high temperature stability and retained 47% activity after a 2-hour incubation at 55°C. In addition to temperature stability, recombinant xylanase showed high pH stability - 10 h incubation in buffers with pH 3-11 did not decrease the activity. The effect of metal ions, detergents, and organic solvents on the activity of recombinant glycosylated xylanase was studied.\u0000The high biochemical parameters of recombinant glycosylated xylanase from Bacillus sonorensis T6 suggest that this enzyme is promising for use in the biotechnology and food industry.

In this regard, the relevance is due to the growing interest in finding ways and means to increase oil recovery, especially to the constant use of microbiological methods.\u0000Oil is an important source of energy and chemical raw materials. Its reserves are limited and irreplaceable. The existing methods allow us to extract up to 40-60% of geological oil reserves in the development of oil fields. One of the most urgent tasks is to create the most effective methods to increase oil production in the developed fields.\u0000Microbiological methods used to increase oil production attract attention with their high efficiency and environmental safety, low capital intensity. In biotechnology, additional oil displacement occurs to a certain extent, these mechanisms provide both physical and chemical methods, but since the metabolites of microorganisms are formed directly in the plate openings, their impact efficiency is higher. It is impossible to master these methods without taking into account the microbiological and physico–chemical conditions in oil plates.

The main protein signature databases are accessible through the integrated InterPro database, which provides the UniProtKB sequence classification. In addition to characterizing proteins through protein families, many researchers are interested in analyzing the complete set of proteins from the genome (i.e., the proteome), and there are databases and resources providing unreduced sets of proteomes and analyzes of proteins from organisms with fully sequenced genomes. The disease is responsible for serious losses to the cattle industry globally. Vaccination is the most effective way of controlling LSDV. The currently available live attenuated vaccines are used in many countries and provide stable immunity. However, use of these vaccines can sometimes cause postvaccinal complications in animals. Attenuation of viruses by the classical method is associated with random mutations in the genome, and some vaccines can contain a mixture of viruses of different genotypes and virulence. Hence, there is a need to develop a safe immunogenic vaccine that will provide improved protection against LSDV. The most promising approach is gene modification to develop an attenuated capripoxvirus strain with high immunogenicity and protectivity. Sequencing and analysis of capripoxvirus DNA genomes has resulted in the identification of a number of genes whose protein products effectively stimulate antibody production in the host organism. These proteins are able to function both inside and outside the cell, neutralizing the complement factors, inhibiting apoptosis and synthesis of interferons, and reducing counter-inflammatory cytokines and chemokines. Here, we review the literature for instances of genetic manipulation of genes encoding the above proteins, resulting in capripoxvirus attenuation.

Of the more than 335 human infectious diseases that have emerged over the past six decades, influenza is one of the most common infection, causing up to 650,000 deaths per year. In addition to the annual seasonal influenza A, the emergence of its new, pandemic, and highly pathogenic variant is even more dangerous. This review describes the classification of influenza pathogens, their structural features, and sources of isolation. The latest published data on the structure, ecology, phylogenesis and pathogenicity of influenza A subtype H10 viruses from various host species are summarized. It is noted that the avifauna is not the only possible reservoir for H10 influenza viruses; avian H10N8 infected dogs in China and mink in Scandinavia, and H10N7 caused widespread mortality of seals in the Baltic Sea in 2014. The first three human cases of avian influenza A H10N8 were reported in 2013-2014 in China, one of which lead to mortality. Information is provided on the genetic variability and evolution of influenza A subtype H10 strains circulating in avifaunal, mammalian, and human populations. Influenza A subtype H10 virus is actively evolving by exchange of genetic information. The potential epidemiological hazard of influenza A virus (H10N8) is as a causative agent of a pandemic, and the need to control its circulation in populations of birds, mammals and among the people is emphasized.

In recent years, recombinant genotypes of noroviruses, which include RNA-polymerase GII[16], have become globally widespread. The aim of the research was to analyze the genetic diversity of noroviruses circulating in 2016–2021 in the Republic of Belarus in order to establish the contribution of the identified genotypes to the formation of morbidity and to study the features of the circulation of their recombinant variants containing RNA polymerase GII [P16]. Sequencing and genetic analysis of a fragment of the ORF1 / ORF2 genome of 242 noroviruses from patients with AGE collected in 2016-2021 was carried out. It was found that 199 norovirus isolates (82.2% of all identified) were recombinant. During this period, genotypes containing GII.P16 polymerase and the VP1 gene of genotypes GII.2, GII.3, GII.4, GII.12, GII.13 prevailed (143 isolates, 71.9% of all recombinant genotypes). The proportion of individual recombinant genotypes was distributed as follows: GII.4 [P16] - 42.0%, GII.2 [P16] - 32.2%, GII.3 [P16] - 16.8%, GII.12 [P16] - 8.4%, GII.13 [P16] - 0.7%. The genotypes GII.4 [P16] and GII.2 [P16] circulated for the longest time - from 2016-2017 to 2021. Their circulation was accompanied by the emergence of outbreaks of AGE: genotype GII.2 [P16] caused outbreaks in 2016, 2018 and 2021, GII.4 [P16] - in 2017 and 2021. All investigated isolates of different recombinant genotypes contained the same variant of the GII.P16 RNA polymerase gene, which became globally distributed in the world in 2015-2017. Comparison of the nucleotide sequences of isolates within genotypes showed that, despite the long circulation period, there were no accumulation of mutations and no selection of genovariants within the genotype.

In Kazakhstan, the rate of drug-resistant tuberculosis among newly diagnosed patients is more than 20%. Moreover, the Republic of Kazakhstan ranks second among the thirty countries with a high disease burden according to the World Health Organization. In this regard, it becomes obvious that it is necessary to study the nature of resistance of locally circulating strains of mycobacteria. The use of genetic typing methods will allow tracking the spread of tuberculosis pathogens of any family, which, in turn, will provide a personalized approach to treatment. The strains of the Mycobacterium tuberculosis complex capable of infecting humans include seven phylogenetic lineages, initially associated with their geographical distribution. In this study, the genomes of three drug-resistant clinical isolates of the Latin American Mediterranean family (LAM) circulating in Kazakhstan were studied. We used whole genome sequencing to study the distribution and drug resistance of isolates. Phylogenetic analysis grouped the genomes described in this study with sequences from Russia, Uzbekistan and Kazakhstan, also belonging to the LAM family. One isolate was identified as having extensive drug resistance to seven anti-TB drugs. Our results suggest that at least two drug-resistant genotypes of the LAM family are circulating in Kazakhstan. Thus, the use of genetic methods is becoming more and more popular and effective in combating the spread of tuberculosis infection.

Certain metabolites of the gut microbiome: certain bile acids, short-chain fatty acids and trimethylamine N-oxide, may contribute to the development and progression of cardiovascular disease. It has been shown that trimethylamine N-oxide can exacerbate cardiovascular disease through the progression of atherosclerosis and thrombotic risks, and has been shown to be correlated with other cardiometabolic diseases such as non-alcoholic fatty liver disease and chronic kidney disease. One of the many factors affecting the level of formation of trimethylamine N-oxide in the human body is nutrition. This article examines the effect of food on the production of trimethylamine N-oxide. The analysis of the data showed that an increase in the synthesis of trimethylamine N-oxide can be influenced by products with an increased content of primary substrates, the composition of the human intestinal microbiome, and the method of cooking products. It should be noted that the concentration level of the oxidized form of trimethylamine can be reduced by some biologically active compounds containing polyphenols. Studies in this area are few and contradictory, which necessitates further research on the effect of food on the metabolism of trimethylamine and its oxidized form.
