Its main function is related to the regulation of mechanisms that control cell growth and division. The RPS6 can be phosphorylated on defined serine and threonine residues by kinases such as S6K1 and S6K2, which in turn are activated by signaling pathways associated with the activation of the mTORC1 molecular complex. The phosphorylation process of protein RPS6 plays a key role in the regulation of cell growth and protein synthesis. Activated RPS6 protein influences translation initiation, which means start the process of protein synthesis on the ribosome. Thus, RPS6 is associated with the regulation of cell size and their ability to divide. In addition, the RPS6 protein can be associated with other proteins and participate in various molecular interactions, which may vary depending on the context of cellular activity. In this study, cloning and site-directed cDNA mutagenesis of the second isoform of the AtRPS6 protein (AtRPS6B) was performed. Further, the obtained phosphomimetic and nonphosphorylated forms of this protein were expressed in E.coli ArcticExpress (DE3) cells, the proteins were purified by metal chelate chromatography (IMAC) and the presence and purity of the obtained proteins was confirmed by immunoblotting","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"37 3","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-05-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"141019844","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This work  presents the results on the expression of the goat  prochymosin gene in the yeast  Pichia pastoris and the study of the milk-clotting  activity of recombinant goat chymosin. The goat prochymosin gene was integrated into the chromosomal DNA of yeast and the secretion of prochymosin into yeast culture was carried out by methanol induction. After activation, chymosin was purified by sequential anion and cation exchange chromatography. A biochemical study showed that a decrease in the milk-clotting activity of the enzyme was observed with an increase in pH. The highest activity of recombinant goat chymosin was manifested at 60 °C and pH 4.5-5.0 and amounted to 7680 ± 0.32 and 8727 ± 0.39 for cow's and goat's milk, respectively. The total proteolytic activity of the enzyme was 7769.2±0.38 U/mg. The results obtained suggest that recombinant goat chymosin can be used in the cheese industry and has good prospects for practical application in the production of cheese from goat's milk.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"18 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-04-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140742224","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In recent years, the role of these cells in the regulation of a number of non-pathological processes, in particular, in the development of vaccine-induced immune response, has been actively discussed. This work was aimed at investigating the role of MDSC in the development of an immune response in old mice immunized with an inactivated whole virion SARS-CoV-2. Immunization of old animals induced an increase in the relative content of B cells and the production of specific antibodies to S1 and N proteins of SARS-CoV-2 in titers comparable to those of young animals. At the same time, elevated MDSC levels in aged animals negatively correlated with the level of CD8+ T cells. Splenic MDSCs in old animals were characterized by increased production of reactive oxygen species compared to young animals. In vitro experiments showed that MDSC depletion increased antigen-specific production of IFNγ  by CD4+ T cells. The data obtained suggest a negative role for MDSC in regulating the T-cell vaccine-induced response to inactivated SARS-CoV-2 and encourage further studies in this direction, which may be a key to developing approaches to increase the efficacy of vaccines against SARS-CoV-2 in at-risk groups.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"3 2","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-04-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140742486","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Transmission occurs through the bites of hard ticks within the genus Ixodes. Cases of fever of unknown origin are recorded in southern Kazakhstan, however, the prevalence of LB in this area remains unstudied. Almaty oblast is recognized as an endemic area for LB. Ixodid tick vectors also inhabit Jambyl oblast, where antibodies to LB agent were previously detected among residents, while it is classified as non-endemic. Turkistan oblast is also considered non-endemic for LB. The purpose of this study was to investigate the seroprevalence of IgG antibodies to B. burgdorferi s.l. among apparently healthy residents of Almaty city (Almaty oblast), Taraz city (Jambyl oblast), and Shymkent city (Turkistan oblast). A total of 465 human serum samples were collected from March 2022 to September 2023. Using the ELISA method, we detected the presence of IgG antibodies to the LB agent in 5.4% of the entire study group (24/465, 95% CI: 3.3-7.6%), and specifically in 9.3% of residents of Almaty city (11/118, 95% CI: 4.7-16.1%, p=0.03), 4.5% – Taraz city (11/242, 95% CI: 2.3-8.0%) and 1.9% – Shymkent city (2/105, 95% CI: 0.2-6.7%). The results of the study confirm the seroprevalence of IgG antibodies to B. burgdorferi s.l. among the population of Almaty city and Jambyl oblast, and also reveal seropositive samples in residents of Turkistan oblast, suggesting possible infection during travel to endemic regions. It is important to continue LB surveillance in endemic and emerging regions, as well as to raise awareness of the infection among healthcare workers and the general public in non-endemic areas.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"21 3","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-04-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140742003","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This has resulted in an unprecedented increase in the rate of biodiversity loss in human history. According to the International Union for Conservation of Nature and Natural Resources, over the past 500 years, as a result of direct or indirect influence of the anthropogenic factor, 774 animal species and 123 plant species have become extinct. To prevent further loss of biodiversity, in the context of the ongoing «crisis of biodiversity», all over the world, including Kazakhstan, there is a need for reliable conservation of the available genetic resources of plants and animals, primarily rare and endangered species. In this regard, specialized biobanks of organized collections of living biomaterials are successfully used worldwide for the conservation and restoration of populations of rare and endangered species of flora and fauna. The article presents information about some of the largest biobanks and their importance in solving this global problem. The experience of scientific centers of Kazakhstan in the field of biobanking of biomaterials of wild flora and fauna, including rare and endangered species, is given, and the results of the National Center for Biotechnology in this area are summarized.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"228 ","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-04-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140769178","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This study was carried out to analyze genetic diversity and identify marker-trait association analysis related to salt tolerance using SSR markers for 215 accessions of G. arboreum. The salt tolerance-related traits like germination rate, fresh weight, stem length, water content, chlorophyll content, electric conductivity, and MDA of the accessions were identified using 150 mM salt for 7 days of seedling growth. \u0000According to the comprehensive index of salt tolerance, 215 accessions were categorized mainly into four groups. Twenty four accessions were classified as highly tolerant to salt treatment (>2.5). The natural population was classified into 3 main groups by phylogenetic analysis. The classifications of phylogenetic analysis were largely congruent with the breeding history and ecological region. \u0000Twenty-two strong marker-trait associations were obtained with strict significant P value i.e. P< 0.01 and four makers including NAU1023, NAU1099, JESPR222, and NAU2783 were significantly related to salt tolerance. The marker NAU2783 was highest associated (P= 1.98E-12) with relative electric conductivity, and with the highest phenotype variation of 20.87%. Some markers are significantly associated with more than two traits. MUSS020 was significantly (P<0.01) associated with relative fresh weight, and relative MDA, NAU1375 was associated with relative fresh weight and relative stem length, while NAU3468 was significantly associated with relative fresh weight, relative germination rate, and relative water content. The aim of this work was to study the phenotypic and genotypic diversity of non-poliomyelitis EVs that circulated from 2003 to 2022 in our country during epidemic upsurges and decreases of enteroviral morbidity. It was established that during this period there was a circulation of 44 different types of EVs belonging to the species  Enterovirus B (97.8%), Enterovirus A (1.88%), Enterovirus C (0.9%), Enterovirus D (0.4 %). Among Enterovirus B viruses, Coxsackievirus B5, ECHO 30, and ECHO11 prevailed. During the years of epidemic rises of morbidity, the ECHO 30, ECHO 9 and ECHO 11 viruses were significantly more often identified (p< 0.001), while in the years of decreases Coxsackievirus B1, B2, B3 and B4 were detected more often (p<  0.001). ECHO 6 and Coxsackievirus B5 viruses were registered both during periods of epidemic ups and downs and were characterized by the maximum intratype genetic diversity, significantly greater than ECHO30, ECHO6 and ECHO 11 (p< 0.05). During the observation period, 9 genetic variants of ECHO 30, 6 – of ECHO 9, 3 – of ECHO 11, 6 – of ECHO 6 and 13 – of  Coxsackievirus B5 were identified. An analysis of the genetic diversity of ECHO 30, 9, 11, 6, Coxsackie B5 viruses showed that the upsurges of enteroviral morbidity was accompanied by the maximum number of detected genetic variants  and some of them were new to the country's population.
