As a result of the autopsy of wild animals, a collection of Alaria spp. was collected, the intensity of invasion varied from 1 to 1489 samples/animal, and the prevalence of invasion ranged from 10 to 36.11%. The primary taxonomic affiliation of the pathogen was carried out using determinants. Genetic identification was carried out by amplification of a section of the internal transcribed spacer 2 (ITS2) marker gene. The resulting 557 bp amplicons. sequenced using the Sanger method. The nucleotide sequences were deposited in the international GenBank database under accession numbers: from the fox - OM630451, ON248128, ON248130, from the corsac - OM630460, ON248043, ON248044, ON248045, from the wolf - ON358106. Bioinformatics analysis to build a phylogeny showed that the samples of Alaria alata studied by us were combined into a separate cluster by identity, which ranged from 90-98%.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"9 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2022-07-09","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"132069355","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Glyoxalase DJ-1 consists of 189 amino acids and has a molecular weight of 20 kDa. The physiological role of DJ-1 is unclear but recent evidence suggest that it can detoxify reactive electrophiles formed in glycolysis. Expression and purification of the recombinant DJ-1 protein in Escherichia coli revealed an abnormally high affinity of rDJ-1 for Ni-NTA, which was accompanied by a color change of Ni-NTA from blue to blue-violet. rDJ-1 formed the strong complex with Ni-NTA, which was destroyed only by 2400 mM imidazole. Experiments with the mutant DJ-1 protein containing the Cys106Ser substitution confirmed the key role of cysteine at position 106 for the formation of this complex. The discovered phenomenon shows the potential of using the DJ-1-tag as a protein tag for obtaining high-purity protein preparations and for bioanalytical applications.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"16 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2022-07-09","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"132830332","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Previously, one of the major approaches for the development of new cultivars was breeding projects based on the evaluation of key morphological traits, including flowering time and plant height (PH). However, with the development of soybean genetics new molecular tools have emerged, including the development of DNA microsatellites, or simple sequence repeats (SSRs). In this study, a collection of hundred accessions grown in the southeast of Kazakhstan in 2020-2021 was assessed using eight agronomic traits, including plant height (PH), the height of lowest pod, number of lateral branches, number of fertile nodes, number of seeds per plant, thousand seeds weight, yield per square meter (YP2), and maturity group. The assessment of field trials showed a wide range of variability in agronomic traits and suggested that PH is significantly correlated with all studied traits, including YP2. The collection was genotyped using eight SSRs associated with PH. The results suggested that three SSRs (Satt244, Satt288, and Satt371) were the most polymorphic, and five out of eight SSRs were associated with six important agronomic traits, including flowering time, PH, and yield per square meters. Obtained results can be efficiently used in soybean breeding based on a marker-assisted selection approach.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"12 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2022-07-09","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"129410834","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Estimates of the incidence of Parkinson's disease worldwide range from 5 to 35 new cases per 100,000 people per year. The disorder is defined by the presence of classic motor signs, including the characteristic presence of bradykinesia in all patients, tremor in most, and rigidity. One of the proteins associated with Parkinson's disease is DJ-1, also known as Parkinson disease protein 7 (PARK7). Studies have shown that DJ-1 is an antioxidant protein that plays a key role in counteracting oxidative stress in dopamine neurons. In the framework of the studies carried out in Escherichia coli BL21(DE3)-RIPL strain, the recombinant rDJ-1 protein was obtained in an amount of 3.5 mg. By immunizing a rabbit, polyclonal anti-rDJ-1 antibodies were isolated, which, in enzyme immunoassay and Western blotting, showed high sensitivity to the DJ-1 protein at a dilution of up to 1: 512,000. The resulting polyclonal antibodies showed the required informativeness when studying the expression of DJ-1 in the cell lines HEK293 (wild type) and HEK293park- (with knocked-out park gene). The resulting antibodies can be used in studies to assess the expression level of DJ-1 in immunological tests to detect and quantify the target protein: flow cytometry, ELISA, Western blotting, immunofluorescence, immunohistochemistry, immunoelectrophoresis.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"460 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2022-06-03","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"132361232","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

A total of 39 wolves (Canis lupus) were examined, and their parasite fauna was assessed. The study identified the following types of helminths Echinococcus granulosus, Taenia krabbei, Dypilidium spp., Mesocestoides spp., Toxascaris leonina, Trichinella nativa, Dirofilaria repens. The extensiveness of the invasion wolves by helminths was relatively high in the western part region (96.5%), and towards the north-central part region, this indicator significantly decreased (65.2%). The average number of helminths was high, with some infected animals carrying several types of parasites. Thus, the intensity of invasion by helminths in wolves was 7.6 specimens per infected individual host. The research aimed to study the endoparasitic fauna of wild wolves using complex classical parasitological and molecular genetic methods. Samples of helminths were differentiated by amplification and sequencing using the marker gene cytochrome oxidase COX1 (GenBank accession number: MT877205, MZ505444, MZ669895, MZ724175).
