The activity of chymosin in the yeast culture was 174.5 U/mL. Chymosin was concentrated 5.6-fold by cross-flow ultrafiltration with 10 kDa cut-off membrane, and chymosin was purified by ion exchange chromatography. As a result of optimization of virus cultivation conditions, the cytopathic effect of the virus was recorded on the Vero cell culture. Previously, genotyping of the rabies virus circulating in Kazakhstan was based on the nucleoprotein gene sequence (N gene). The protocol describes the methods of sample preparation, whole genome sequencing and genotyping currently used for epidemiological analysis not only of Rabies virus, but also for other viral pathogens such as SARS-CoV-2, Influenza and others. The aim of the work was to develop and test a protocol for whole genome sequencing and genetyping of rabies virus isolates for further use in epidemiological monitoring. As a result of the research, whole genome sequences of 31 isolates of Rabies virus isolated from various infected animals and regions of Kazakhstan were obtained for the first time. All the isolates belong to the major steppe-type clade – Cosmopolitan (30 isolates belong to the minor clade CA1, while 1 isolate belongs to an unassigned minor clade). 2 local outbreaks and 6 separated rabies virus lineages were identified. Estimated dates of divergence from a common ancestor were calculated for both the Cosmopolitan clade and the minor clade CA1.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"1 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-01-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"128735860","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

It was suggested long ago (by Fisher, in 1931) that tight linkage to a sex-determining locus facilitates the accumulation of SA mutations even when their detrimental effect in one sex exceeds its benefit in the other. Due to the difficulties in detecting SA alleles directly, we know very little about the nature of X-linked SA mutations and their role in the evolution of differential gene expression between sexes. To investigate how X-linked SA mutations affect the transcriptome profile in both sexes, we analyzed changes in genome-wide gene expression pattern in Drosophila melanogaster head tissue that had undergone 95 generations of a female-limited X-chromosome experimental evolution. We found an evidence of change in gene expression towards to the female optimum, which we interpret as the result of resolution of sexual conflict over X-linked SA mutations. In addition, we also found a potential unknown gene expression effect of the balancer chromosome on gene expression (that was necessary to use to limit the inheritance of target X chromosome to the female line). All these findings indicate that a highly polymorphic nature of the X chromosome with an important role in genome-wide gene expression profile.","PeriodicalId":101644,"journal":{"name":"Eurasian Journal of Applied Biotechnology","volume":"109 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2022-12-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"116160422","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The lack of moisture remains a limiting factor and leads to a decrease in plant productivity in an arid climate. Therefore, an important direction of modern breeding, and today, is to increase the drought tolerance of plants when creating new varieties. To assess the resistance of plants, various methods are used, including laboratory ones. In physiological assessment, an effective method is to assess drought tolerance at the seedling stage. A deeper analysis is molecular labeling based on the genes that control the trait under study. The article presents the results of studies on the assessment of soft durum wheat samples for resistance to osmotic stress and their molecular screening using marker technology. The purpose of the research is to study and select the selected varieties of spring wheat on the basis of drought tolerance. The quality of the product depends on the strain and is due to differences between strains in the presence and activity of metabolic pathways. The selection of competitive bacterial cultures and the development of bacterial starters are fundamental to establishing and maintaining industrial microorganism collections. The process of creating bacterial starters takes a lot of time and includes the main stages of work: the isolation of microbes from various sources, their development, the selection of promising strains with industrially active properties, and the subsequent isolation of pure cultures as part of a consortium of bacterial starters. Potential probiotics were isolated from samples of fermented milk products - cottage cheese, butter, etc. home-made in the Akmola region, then identified and characterized. In this study, new probiotic bacteria were selected with high antibacterial activity against pathogenic microorganisms for humans and sensitivity to antibiotics. 4 strains of the studied isolates showed resistance to antibiotics and test strains. Because it reduces the risks of many chronic diseases such as diabetes, cancer and digestive disorders. Due to these properties, β-glucan is a natural substance that can be used in pharmaceutical preparations for the prevention and treatment of many chronic diseases. Therefore, an analysis was made for crops rich in sources of β-glucans - 100 varieties of wheat, 50 varieties of barley and 50 varieties of oats, and a collection of genomic DNA was made. Each variety was screened to identify the genes responsible for the synthesis of β-glucans. In the frames of this study, 10 genes were considered. As a result, these genes have been identified in barley, wheat, and oat varieties.\u0000 For the isolation of β-glucan, the methods of acid extraction, microwave, and heating at high temperature were used. A modified method for isolating β-glucan from oatmeal was developed. According to the results, the acid extraction method proved to be more effective. It is easy to obtain pure β-glucan by this method, and the use of citric acid is safer and more affordable than conventional acids.
