Effect of Long-Term Treatment with Alendronate on Bone Repair and Mineralization Around Implants in Rats.

A total of 160 female Wistar rats were randomly assigned to two groups: the control group (CTL) and the alendronate group (ALD). The ALD group received a subcutaneous injection of sodium alendronate (1 mg/kg/week), while the CTL group received weekly injections of saline solution. After 120 days of treatment, a bilateral implant was placed in the tibia of each rat. Ten rats from each group were euthanized at 5, 10, 15, 20, 25, 30, 45, or 60 days post-surgery. Picro-sirius red staining was utilized to assess the distribution and arrangement of collagen fibers near the implant threads. Bone mineralization mapping of the native bone adjacent to the implant was performed using images obtained through scanning electron microscopy (SEM) across all follow-up periods. SEM-based mineralization mapping revealed an increase in both the degree and homogeneity of bone mineralization in the ALD group compared to the CTL group. Alendronate administration affected collagen arrangement and distribution, leading to a connective tissue with reduced organization and thinner collagen fiber bundles. Treatments are usually mutilating, palliative, and expensive. This study aimed to evaluate the prevalence of lip diseases and factors associated with its development in peddlers on two beaches of the Guarujá/Brazil coast. Data were collected by clinical examination and use of a questionnaire of 182 peddlers on the beaches. They also received an explanatory folder with information about oral care, and the importance of the use of sunscreen for cancer prevention. All workers diagnosed with lip injuries were referred for dental treatment. The prevalence of lip alterations was 40.7% (n = 70), furthermore, smokers included 27.5%, and alcohol consumption was 20.3%. Regarding selfie protection, 72% of workers reported wearing a hat or cap frequently, 48.4% reported using body sunscreen, and only 11.5% reported using lip balm. There were no significant differences in factors such as alcoholism and smoking related to lip lesions diagnosis. However, when characterizing these factors, 48.6% of those who drink, or smoke have lip alterations. Reference casts (maxillary Kennedy class III and IV) were produced by computer assisted design and manufacture, and linearly measured at occlusocervical, interarch, and edentulous space dimensions. Intraoral scanner (IOS), extraoral scanner (EOS) and cone beam computed tomography (CBCT) digitized the reference casts. Each digital file was 3D-printed using stereolithography technology, totalizing sixty experimental casts (n=10 per group). The same measurements taken from the reference casts were performed on experimental casts. Two-way ANOVA and Bonferroni post-test were used for trueness (distortion between the experimental and reference casts). Distortion was significantly greater for class IV when compared with class III and increased after the 3D-printing step. Among digitizing methods, IOS and EOS had a similar performance and casts from CBCT showed higher distortion, reaching -1.0 and -1.4 mm in the edentulous spaces of digital and 3D-printed cast, respectively. The aim of this study was to evaluate the effect of the FGP removal protocol on the time for FGP removal (min), the root dentin removed (mm3), the angle alteration of the root canal alignment (o), and the root perforation occurrence using cone-beam computed tomography (CBCT) analysis. Thirty extracted maxillary molars were randomly assigned to 3 groups (n = 10): Mic-Ult, FGP removed using ultrasonic inserts under microscopic magnification; Mic-DiB, FGP removed using diamond burs under microscopic magnification, and Endo-G, FGP removed using Endo-G. CBCTs were made after access opening and after FGP removal. The volume of root dentin, the root canal alignment and root perforation were performed using InVesalius and Mimics softwares. Root canal angle alteration after FGP removal was performed using ImageJ software. The time for FGP removal was calculated in min. Data were analysed by one-way analysis of variance was followed by Tukey HSD test (α = 0.05). Endo-G resulted in significantly lower root dentin removal (p< .001), and significantly less time than Mic-Ult and Mic-DiB (p< .001). Mic-DiB had 1 and Mic-Ult had 2 root dentin perforations. FGP post removal using Mic-Ult and Mic-DiB exhibited significantly greater alteration in the root canal alignment than that using Endo-G (p< .001). The 200 central incisors were endodontically treated using different materials (Sealapex, Bio-C Sealer, AH-Plus, Endofill, and Portland cement) and distributed according to the presence or absence of dentin hybridization. The teeth underwent colorimetric analysis using the Easyshade® spectrophotometer at four different times: determining the color of the dental substrate before root canal filling (RCF), 7 days, 60 days, and 180 days after RCF. The data were evaluated for normality and homogeneity by the Shapiro-Wilk and Levene tests. A two-way analysis of variance was performed, with Sidak's post-test for multiple comparisons. The significance level was 5%. Dentin hybridization influenced the coronary chromatic alteration of Sealapex® after 7 days and in Bio-C Sealer® and AH Plus® after 60 days. The AH Plus after 7 days obtained the highest ΔE* when hybridized, and the Endofill® obtained the highest ΔE* when not hybridized. The AH Plus after 60 days got the highest ΔE* when hybridized and non-hybridized. The Sealapex® after 180 days obtained the highest ΔE* when hybridized, and the AH Plus obtained the highest ΔE* when not hybridized. The studied root canal sealers behaved differently regarding coronary chromatic alteration, making it difficult to predict a particular behavior. The high polyphenol content in cocoa pod husk (Theobroma cacao L) has the potential to influence bone metabolism and contribute to the inhibition of bone resorption. The aim of this study was to analyze the anti-osteoclastogenesis potential of cocoa pod husk (Theobroma cacao L.) in both in silico and in vivo study. An analysis of the anti-osteoclastogenesis potential of T. cacao bioactive compounds was conducted using molecular docking simulations. Thirty male Wistar rats (Rattus novergicus) were randomly assigned to control negative groups (placebo gel), control positive groups (2% doxycycline gel), and treatment groups (10% cocoa pod husk (CPH) extract gel), with measurements taken on days 7 and 14. Wistar rats were induced with 0.05 ml of P. gingivalis at a concentration of 2x109 CFU/ml intrasulcularly in the maxillary molar to achieved in periodontitis. The number of osteoclasts was observed by hematoxylin and eosin staining, the level of TNF-α was assessed by enzyme-linked immunosorbent assay, and the expression of RANKL was evaluated by immunohistochemical staining. Data were analyzed using One-way ANOVA to examine the differences between the groups. The in silico study showed that the catechin, epicatechin, quercetin, and procyanidin B2 had a strong binding affinity for TNF-α and RANKL. Administration of 10% CPH reduced the number of osteoclasts (p<0.05), TNF-α level on days 7 and 14 (p<0.05), and RANKL expression on day 7 (p<0.05) in experimental rats with periodontitis. Two hundred and seventy dentin specimens from human molars were prepared. The specimens were randomly allocated into 10 experimental groups (n=10): 4 corresponding to desensitizing mouth rinses, 4 to conventional mouth rinses, a negative control group (C-: distilled water), and a positive control group (C+: 500 ppm fluoride plus 800 ppm tin mouth rinse). Specimens were subjected to an erosion-abrasion cycling model: 5 min immersion in 0.3% citric acid and 60 min exposure to artificial saliva. This procedure was repeated 4x/day for 5 days. Immediately after the first and last erosive challenges, the specimens were brushed with a slurry of fluoride toothpaste for 15 s, with a total of 2 min exposure to the slurry. Afterward, the specimens were exposed to the mouth rinses. Dentin surface loss (SL, in µm) was determined by optical profilometry. Data were statistically analyzed by using Kruskal-Wallis and Dunn's tests (α=0.05). The only mouth rinses that presented significantly lower dentin SL than the C- was a desensitizing one containing sodium fluoride (220 ppm F-) with dipotassium oxalate (1.4%) and the C+ (p<0.001 and p=0.013, respectively), without significant differences between them (p>0.05). Dentin SL of the other groups did not significantly differ from the C- (p>0.05). This study evaluated the quality of communication between professors and students during the process of learning Dentistry. A questionnaire was developed and applied to evaluate their perceptions about their communications in an undergraduate Dentistry course. The questionnaire had ten items to analyze professors' self-perception and ten for students' perceptions, using a Likert-like scale and a final open question. During its construction, it was sent to five specialists to analyze content validity. The open question gathered suggestions to improve and intensify communications effectively and to identify vulnerabilities. Total scores ranged from 10 to 50, with 10 as the worst perception, and 50, as the best. The scores were calculated by adding all answers and then dividing that sum by the total number of items. Cronbach's alpha was calculated to evaluate the instrument's internal consistency. The level of significance was set at 0.05%. The Student t-test was used to determine differences between groups. Professors' self-perceptions and students' perceptions of the quality of communication of the university teaching staff during the student's learning process had statistically significant differences. Professors classified their ability to communicate when emitting and receiving information as satisfactory. Students, however, did not fully agree with them, particularly on the items about receiving information. Five electronic databases and grey literature were systematically screened to identify studies comparing the tooth discoloration potential of endodontic sealers with different chemical bases. Studies that did not evaluate human teeth or did not employ spectrophotometry as a method for color measurement were excluded. The risk of bias in the included studies was assessed using a modified version of the JBI Critical Appraisal Checklist for Experimental Studies. Data were grouped according to the chemical composition of the sealers and analyzed qualitatively. Fourteen studies were included. None of the studies met all domains of the risk of bias checklist. Qualitative synthesis suggested that zinc oxide and eugenol-based sealers have a greater potential for tooth discoloration. Silicone-based and calcium hydroxide-based sealers demonstrated less potential for tooth discoloration than resin-based sealers and salicylate resin-based sealers containing calcium silicate. All investigated endodontic sealers induced tooth discoloration, which is chemical and time dependent. Positioning guides (n=10) in different colors (translucent, purple, yellow, green and, blue) were made for each surface (ceramic, resin composite and bovine tooth). The CIEL*a*b* color coordinates were measured with the positioning guides and with no positioning guide (control group). The ΔL*, Δa*, Δb*, ΔE*ab, and ΔE00 were calculated between the control group and the different groups. The CIEL*a*b* color coordinates were analyzed by one-way ANOVA followed by the post-hoc Tukey test (α=0.05). Color differences were interpreted using the 50:50% perceptibility and acceptability visual thresholds. Positioning guides with different colors presented significant differences for all surfaces and color coordinates. ΔE*ab, and ΔE00 mean values for all surfaces and positioning guide colors exceeded the acceptability visual thresholds, except for the bovine tooth surface measured with the green positioning guide.
