And only in this way can focus the integration of CM and WM on solving practical problems and obtaining clinical efficacy. Methods SGC-7901 human gastric cancer cell strain was taken as subjects. The vectors of NF-κB (ReIA/p65)-PECFP and STAT3-PEYFP were constructed and transfected in SGC-7901 human gastric cancer cells. Cells were then intervened by MLR after stimulated by LPS. The crosstalk between NF-κB and STAT3 in cells was detected using fluorescence resonance energy transfer and co-immunoprecipitation. Results After LPS stimulation, the crosstalk between NF-κB and STAT3 was enhanced. But it was significantly weakened after MLR intervention. Methods C57BL/10J mice TLR4 gene (TLR4 +/+) and knockout mice (TLR4 -/-) were randomly divided into 3 groups, the blank control group, the radiation model group, the BJR group. Sampling was respectively performed at day 1 , 14, and 30 after radiation. The general condi- tion of mice was observed. White blood cell (WBC) , red blood cell ( RBC) , and platelet ( PLT) were counted in each group. Thigh bone of mice was collected for bone marrow specimen. Bone marrow slice was prepared. Pathomorphological changes were observed under electron microscope. Results Com- pared with the blank control group, WBC, RBC, and PLT all decreased or showed a decreasing tendency after one day radiation. RBC and PLT significantly decreased after 14 days of radiation (P <0. 01 , P < 0. 05). Counts of WBC and RBC were higher in the TLR4 +/+ BJR group than in the rest 2 groups (P < 0. 01 , P <0. 05). No statistical difference in RBC or PLT between mice after 30 days of radiation and mice in the blank control group (P >0. 05). PLT count in the TLR4 +/+ BJR group was most approximate to the normal value. WBC count obviously increased, but still with statistical difference as compared with the blank control group (P <0.01). WBC count recovered most rapidly in the TLR4 +/+ BJR group. Results under light microscope showed the structure of bone marrow was injured to different degrees in all mice except those in the TLR4 +/+ BJR group. Methods Totally 162 clinical undergraduates were randomly and equally assigned to the observation group (PBL group) and the control group [lecture-based learning (LBL) group], 81 in each group. Clinical practice was conducted in them. At the end of clinical practice the teaching effect was assessed using test and questionnaire. Results Performance records of case analysis and four diagnoses of CM were higher in the PBL group than in the LBL group (P <0. 05). The satisfaction survey for teaching was better in the PBL group than in the LBL group (P <0. 05). {"title":"[Effect of Active Ingredients of Qingxin Kaiqiao Recipe on Spatial Learning and Memory Capacities, and Expressions of Bax, Bcl-2, Caspase-3, and PAPP in Hippocampus of AD Model Rats].","authors":"Yi-Ru Wang, Yi-Hui Zhang, Zhi-Yu Chen, Dong-Mei Xu, Xiang Chen, Xiao-Yan Zhang, Hai-Yan Hu","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Objective To observe the effects of active ingredients of Qingxin Kaiqiao Recipe (QKR) , such as saponins, volatile oils, effective compositions of polysaccharides, on expressions of Bcl-2 associated X protein (Bax) , B-cell lymphoma-2 (Bcl-2) , cysteinyl aspartate specific proteinase-3 (Caspase-3) , and β-amyloid precursor protein (pAPP) in hippocampus of Ap1_40-induced Alzheimer's disease (AD) model rats. Methods Totally 112 male Sprague-Dawley (SD) rats were randomly divided into 7 groups, i.e., the normal control group, the sham-operation group, the model group, the Aricept group, the saponin group, the volatile oil group, the polysaccharide group, 16 in each group. The AD rat model was established by injecting Aβ₁₋₄₀ from bilateral hippocampus. Equal volume of double distilled water was administered to rats by gastrogavage in the normal control group, the sham-operation group, the model group from the 2nd day after modeling, once per day for 2 successive weeks (at 10:00 am). Aricept (Donepezil Hydrochloride Tablet, 1. 67 mg/kg per day) , saponin (9 mL/kg per day) , benzene (3. 33 mL/kg per day) , and polysaccharides (8. 33 mL/kg per day) was administered to rats by gastro- gavage to the Aricept group, the saponin group, the volatile oil group, the polysaccharides group, re- spectively, once per day for 2 successive weeks (at 10:00 am). By the end of gastrogavage spatial learning and memory capacities were detected using Morris water maze (MWZ). Apoptosis in hippocam- pal CAI region was detected using TUNEL staining. Expressions of Bax, Bcl-2, Caspase-3, and PAPP were measured via Real-time fluorescent quantitative PCR, Western blot, and immunohistochemistry, respectively. Results There was no statistical difference in pre-modeling escape latency and times of crossing platforms among groups at the same time point (P >0. 05). Besides, escape latency was gradu- ally shortened as time went by. Compared with the model group, escape latency was shortened, and times of crossing platforms was significantly increased in the Aricept group and the saponin group (P < 0. 05, P <0. 01). Compared with the model group, the amount of apoptotic cells in hippocampal CA1 re- gion was obviously reduced (P <0. 05, P <0. 01) , expressions levels of Bax, Caspase-3, and pAPP were down-regulated, Bcl-2/Bax ratio was obviously elevated in the saponin group, the volatile oil group, the polysaccharide group (P <0. 05, P <0. 01). {"title":"[Treatment of Idiopathic Thrombocytopenic Purpura by Xiaoban Decoction: a Case Observation of 119 Patients].","authors":"Er-Yun Su, Yi Zhao, Li-Hua Huang, An-Na Zhao","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Objective To observe the clinical effect of Xiaoban Decoction (XBD) in treating idio- pathic thrombocytopenic purpura (ITP). Methods Totally 119 ITP patients were treated by XBD. The he- mogram, bleeding symptoms, effective rate, and adverse reactions were observed before and after treatment. Patient condition was observed while taking XBD but with other treatment program withdrawal (including glucocorticoids). Results After 6 -18 months of treatment, WBC count, hemoglobin concen- tration, and platelet count were all increased, as compared before treatment. Complete relief (CR) oc- curred in 57 cases (platelet count≥100 x10⁹/L) , response(R) in 27 cases (platelet count≥50 x10⁹/L) , no relief (NR) in 35 cases (platelet count <50 x10⁹/L). The total effective rate was 70. 6%. Bleeding symptoms were improved after one-month treatment. There were 70 patients using glucocorticoids before taking XBD. No withdrawal crisis occurred after rapid withdrawal. No adverse reaction occurred, except increased times of passing stool. Seven patients were pregnant during follow-up and gave birth safely lat- er. Conclusions XBD, independent of glucocorticoids, could rapidly improve bleeding symptoms and steadily elevate platelet count. Methods Chemotherapy induced myelosuppression model was established in Lewis lung tumor bearing mice by intraperitoneal injection of cyclophosphamide (CTX). And then they were intervened by SHSBG. Routine white blood cell (WBC) count, red blood cell (RBC) count, platelet count, and tumor mass were calculated. Ratios of bone marrow hematopoietic stem cell (Sca, CD34 double positive cells) were detec- ted by flow cytometry. mRNA expression of main genes in Wnt signaling pathway (Wnt, β-catenin, Frizzted, DSH, GSK3) were detected using real time fluorescent quantitative PCR. Results The number of WBC and ratio of hematopoietic stem cells in the treatment group were higher than those in the model group (P<0. 05). Expressions of Wnt, β-catenin, Frizzted, DSH, and GSK3 mRNA in the bone marrow were higher in the treatment group than in the model group (P <0. 05). Expressions of Wnt, β-catenin, Frizzted, and DSH mRNA expression in tumors were lower in the treatment group than in the model group (P <0. 05). There was no statistical difference in counts of RBC and platelet, tumor mass, or GSK3 mR- NA expression among all groups (P >0. 05). Conclusions The mechanism for SHSBG treating myelo-suppression was related to regulating Wnt signaling pathway. Methods Totally 56 patients with diabetic foot were assigned to the control group (29 cases) and the treatment group (27 cases) according to Wagner grading method (the range and the degree of foot lesion). Patients in the control group received basic treatment (anti-inflammation, blood glucose control, anti-coagulation, debridement, drainage, and so on) for 8 weeks. Patients in the treatment group additionally received XYP for 8 weeks. The wound healing was observed. Contents and protein expressions of VEGF/SDF-1 a/CXCR4 were detected using SP method and Western blot. Results The wound healing rates after 2, 4, 8 weeks of treatment were signifi- cantly higher in the treatment group than in the control group (all P <0. 05). Contents and protein expres- sions of VEGF/SDF-1 a/CXCR4 protein expression at week 8 after treatment were all significantly higher in the treatment group than in the control group (P <0. 05). Methods MCAO model was pre- pared in male SPF grade SD rats by suture method. Totally 90 rats were divided into the sham-operated group, the model group, and the EA group according to random number table, 30 in each group. Rats in the sham-operated group only received surgical trauma. Rats in the model group only received MCAO I/R injury. Rats in the EA group received EA at Baihui (DU20) and Dazhui (DU14) , once per day, 30 min each time. Nerve defects of rats were tested by neural function defect scale at day 1 , 7, 14 of treatment, respectively. Meanwhile, 6 rats were executed randomly from each group. Their hippocampus tissues were isolated. Then the proliferation and differentiation expression of eNSCs in the hippocampus area were detected by immunofluorescence method. Results (1) The scores of nerve function defect scale: The scores of the model group increased at day 1, 7, 14 of treatment, being higher as compared with those of the sham-operated group (P <0. 05). The scores of the EA group were lower than those of the model group at day 1, 7, 14 of treatment (P <0. 05). (2) The expression of BrdU positive cells: Com- pared with the sham-operated group, the expression of BrdU positive cells in the model group were in- creased at day 1, 7, 14 of treatment (P <0. 05). Compared with the model group at each time points, the expression of BrdU positive cells in the EA group were increased more at day 1, 7, 14 of treatment (P < 0. 05). (3) The expression of Nestin positive cells: The expression of Nestin positive cells were in- creased more in the model group than in the sham-operated group at day 1 , 7, 14 of treatment (P < 0. 05). Compared with the model group, the expression of Nestin positive cells increased more in the EA group, but only with statistical difference at day 7 of treatment (P <0. 05). (4) the expression of DCX positive cells: the expression of DCX positive cells were increased more in the model group than in the sham-operated group at day 1 and 7 of treatment (P <0. 05). Methods Registry method was used in this study. Risk factors and information on symptoms of CM collected from 1 823 OP high risk females (40 -65 years old) in Dongcheng District of Beijing and Xuhui District of Shanghai (collected in our previous research). Meanwhile, Cox proportional hazard model was also a- dopted to conduct single-factor and multiple-factor analyses. Risk factors and symptoms of CM were screened and analyzed. Results A total of 1 498 subjects attended follow-up surveys, with the response rate of 82.17%. Of them, 52 had brittle fracture. In Cox single-factor analysis, when taking the time for occurrence of postmenopausal OP as the outcome variable, average age (RR = 1. 083, P = 0. 003 ) , whether menopause (RR = 5. 462, P = 0. 004), average menopausal year (RR = 1. 060, P = 0. 010) , menopausal year ≤10 years (RR =4. 890,P =0. 008), menopausal year >10 year (RR =7. 720,P = 0. 001) , delivery times ≤3 times (RR =5. 870, P =0. 003), dizziness (RR =3. 037, P =0. 001) , spasm of lower limbs (RR =2. 315, P =0. 003) , reduced bone mass in bone mass density (BMD) diagnosis (RR = 2. 376, P =0. 008) , β >0 and RR >1 in BMD diagnosis (RR =2. 801 ,P =0. 013) indicated they were risk factors for postmenopausal OP fracture. When taking BMD diagnosis, age, menopausal year, delivery times, dizziness, and spasm of lower limbs in multiple-factor analysis, menopausal year <10 years (RR =3. 257,P =0. 032) and dizziness (RR =2. 655,P =0. 005) were associated variables.
