Methods CIA model was established in Sprague-Dawley rats. Successfully modeled rats were randomly divided into the model group, the positive drug ,oup, high and low dose TMD groups, 6 in each group. Besides, a normal group was set up (n =6). Deionized water (10 mL . kg⁻¹ . d⁻¹) was administrated to rats in the normal group and the model group by gastro- gavage. Leflunomide (1. 87 mg . kg ⁻¹ . d ⁻¹) was administrated to rats in the positive drug group by gastro- gavage. TMD (31. 8 g crude drugs . kg ⁻¹ . d ⁻¹ and 15. 9 g crude drugs . kg ⁻¹ . d ⁻¹) was administrated to rats in high and low dose TMD groups respectively by gastrogavage. The intervention lasted for 12 suc- cessive weeks. Protein and mRNA levels of PPARy, P65, and IL-17 were detected at the end of intervention. Results Compared with the normal group, mRNA and protein expression levels of PPARγ, P65, and IL-17 were up-regulated in the model group (P <0. 01). Compared with the model group, PPARγ pro- tein expression level was up-regulated, mRNA and protein expression levels of P65 and IL-17 were down-regulated in high dose TMD group (P <0. 01). mRNA and protein expression levels of PPARγ were up-regulated, mRNA and protein expression levels of P65 and IL-17 were down-regulated in the positive drug group and low dose TMD group (P <0. 01). {"title":"[Effect of Xinfeng Capsule on Beclin1/PI3K-AKT-mTOR of Adjuvant Arthritis Rats].","authors":"Ya-Li Wang, Jian Liu, Lei Wan, Li-Ping Ruan, Wen-Fang Ye","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Objective To observe expression levels of autophagy related 5,7,12 mRNA (Atg5, 7,12), microtubule-associated protein 1 light chain 3 (LC3-II), Beclin1, phosphatidylinositol 3-kinase (PI3K) , protein kinase B (AKT) , mammalian target of rapamycin (mTOR) , IL-1 β, TNF-α, IL-4, and IL- 10 in adjuvant arthritis (AA) rats, and effects of Xinfeng Capsule (XFC) on them. Methods Totally 48 male SD rats were divided into 4 groups, i.e., the control group, the model group, the Western medicine (WM) group (leflunomide, 5. 0 mg/kg) , the Chinese medicine (CM) group (Xinfeng Capsule, 3.0 g/kg) , 12 in each group. Thirty days after medication body weight (BW) , toe swelling degree (E%) , arthritis in- dex (AI) , and pathological changes of ankle joint and ultrastructural changes were observed. mRNA ex- pressions of Atg5, 7, 12, protein expressions of LC3-L , Beclin1 , PI3K, AKT, mTOR, serum contents of IL-1 β, TNF-α, IL-4, and IL-10 were detected. Results Compared with the normal group, E%, Al, IL-1 β and TNF-α increased; BW, levels of IL-4 and IL-10, mRNA expressions of Atg5 and Atg12, protein ex- pressions of LC3-ll and Beclin1 decreased (P <0.01, P <0.05), protein expressions of PI3K, AKT, mTOR increased (P<0.01) in the model group. Compared with the model group, E%, Al, mRNA expres- sions of IL-1β , TNF-α-, and Atg12, protein expressions of PI3K, AKT, and mTOR decreased (P <0.01, P<0.05), IL-4, IL-10, protein expressions of LC3-II and Beclinl increased (P <0.01, P <0.05) in the two medicated groups. Atg5 mRNA expression decreased (P <0.01) , Atg7 mRNA expression increased (P < 0.05) in the WM group. Compared with the WM group,BW, IL-4, mRNA expressions of Atg5 and Atg12, protein expressions of PI3K and mTOR increased in the CM group (P <0.01 , P <0. 05). Conclusions The level of autophagy in AA rats was decreased, leading to excessive proliferation of synovial cells, swollen joints, elevated proinflammatory factors, decreased inflammatory factors, resulting in inflamma- tory reactions of joints. Methods Different concentration of LPS (0. 01,0. 1 ,1. ,10 mg/L) was applied to the HPMECs to induce acute HPMECs injury. The HPMECs mod- els (n =3) were intervened by Alpinetin(40,80,160,320 mg/L) . Normal HPMECs were selected as control group. The viability of HPMECs was observed,mRNA and protein expression of intercellular cell adhesion molecule-1 (ICAM-1) ,TNF-α,APQ-1 were detected. Results Compared with control group, the protein expression of ICAM-1 , TNF-a were increased, the protein and mRNA expression of APQ-1 and the vi- ability of HPMECs were decreased in model group (P <0. 05). Compared with model group,ICAM-1 and TNF-α protein expressions were significantly inhibited in Alpinetin (80,160 mg/L) group, the mRNA and protein expression of APQ-1 and the viability of HPMECs were significantly increased (P <0. 05, P < 0. 01). Methods Totally 1 200 patients with mixed hemorrhoid were assigned to the control group(600 cases) and the treatment group(600 cases) according to randomized, parallel controlled,multi-center trial design. Patients in the control group received PPH with the simplified M-M surgery, and patients in the treatment group received RPH with the simplified M-M surgery. Postop- erative complications, operation time,the postoperative hospitalization days and the efficacy were ob- served. Results Compared with the control group, the numbers of postoperation hemorrhage, postop- erative uroschesis, anal fissure and anorectal stenosis in treatment group were decreased(P <0. 01 , P < 0. 05), operation time and the postoperative hospitalization days were decreased (P <0. 01 , P <0. 05 ), the cure rate for 3 and 12 months after operation were increased (P <0. 01, P <0. 05). Methods Totally 70 patients with chronic erosive gastritis were selected and assigned to turbidity toxin intrinsic syndrome group and Gan-wei disharmony syndrome group, HGF expression level of ser- um,and HGF,c-Met expression level of gastric mucosa tissues were measured;the correlation of HGF and c-Met in gastric mucosa tissues, and the correlation of HGF in serum and gastric mucosa tissues were analyzed. Results The expression level of HGF and c-Met in turbidity toxin intrinsic syndrome group was higher than that in Gan-wei disharmony syndrome group (P <0. 05) ; the expression level of HGF in gastric mucosa tissues was positively correlated with c-Met(r =0. 831 , P <0. 05) ; the expression level of HGF in serum was positively correlated with that of gastric mucosa tissues(r =0. 656, P <0. 05). {"title":"[Three Waves of Arsenic Therapy for Leukemia and Related Innovations].","authors":"Yong-Ming Li","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>There were three waves of arsenic therapy for leukemia in the history and every time, it started with a particular medical innovation. German doctor first found that Fowler arsenic solution might improve symptoms of leukemia patients in 1865 but the therapy was never popular due to uncertainty. In 1931, American hematologists reported a series of chronic myeloid leukemia cases successfully managed by arsenic solutions , and published the pathological data supporting the specificity of arsenic action on leu- kemic cells. Not until late 1970s, after the publication of new FAB classification of leukemia, Chinese doctors from Harbin Medical University who had been exploring novel applications of traditional Chinese medi- cines, discovered that the new leukemic entity, acute promyelocytic leukemia (APL) was the best indica- tion for arsenic containing remedy. The theory was supported by many APL survivors and later trials with As₂Ο₃, as a monotherapy. The Harbin Protocol was soon repeated successful in China and in the West, and As₂Ο₃, became a FDA approved new drug for APL in 2001. The discovery of arsenic therapy for APL was ap- parently not an incidental finding, but a timely advance following medical innovation. Methods Myocardial ischemia model was established in healthy male SD rats via subcutaneously injec- ting ISO (85 mg/kg) multipointedly (medial root of limbs and the back). Then they were randomly divided into 4 groups, i.e., the model group, Neiguan (PC6) group, Lieque (LU7) group, non-acupoint group, 10 in each group. Besides, another 10 healthy rats were recruited as the control group. Corresponding EA was performed at respective acupoints to rats in Neiguan (PC6) group, Lieque (LU7) group, non-acu- point group, with dense-sparse wave, 2 -3 mA, 2 -20 Hz, needle retaining time of 20 min, once per day for 7 successive days. mRNA expression levels of Kir6. 1 and Kir6. 2, SUR2A, SUR2B, PKA, PKG, and PKCβ₂ in left ventricular myocardium were analyzed by Real-time PCR. Results Compared with the con- trol group, mRNA expressions of each index increased in the model group (P <0. 01). Compared with the model group, mRNA expressions of each index significantly decreased in Neiguan (PC6) group and Lieque (LU7) group (P<0. 01). Compared with Neiguan (PC6) group, mRNA expressions of each index significantly increased in Lieque (LU7) group and non-acupoint group (P <0. 01). Compared with Lieque (LU7) group, mRNA expressions of each index significantly increased in non-acupoint group (P <0. 05). {"title":"[Serum Proteomic Analysis and Construction of Protein-protein Interaction Networks about Ejaculation Praecox with Shen-yang Deficiency].","authors":"Cong-E Tan, Xiang-Yun Huang, Mi-Qu Wang","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Objective To identify serum proteome of ejaculation praecox(EP) with Shen-yang de- ficiency, and to explore its pathogenesis of EP in the protein-protein interaction ( PPI) network. Methods The serum samples were respectively collected from 4 EP with Shen-yang deficiency patients and 4 healthy controls. After the serum proteome of EP with Shen-yang deficiency was obtained, the technology of isobaric tags for relative and absolute quantitation (iTRAQ) was adopted for identification. The STRING data- base was applied to construct the PPI network whose function was analyzed through bioinformatics meth- ods. Results A group of 238 serum proteins were identified in total, of which, 162 proteins reached the strict quantitative standard. Nine proteins were differently expressed, including 1 up-regulated and 8 down-regulated. The constructed PPI network was constituted by 72 protein nodes and 283 protein couples, and could be clustered to 16 clusters, in which 10 clusters were composed of 3 or more proteins. Each cluster could be found with a core protein correspondingly. The core protein of C3,C5,C1S and MASP2 were all main constituents of complement system, whose function involves in biological process of complement ac- tivation.
