METHOD Cell proliferation was assessed by MTT assay. Cell cycle distribution, apoptosis and mitochondrial membrane potential were measured by high content screening (HCS). The protein expression of caspase-3, caspase-8, caspase-9, Bcl-2, Bax, tBid and cytochrome c in the HepG2 cells were evaluated by Western Blots. RESULT beta-Sitosterul exerted significant antiproliferative effects in HepG2 cells. Furthermore, beta-sitosterul also induced HepG2 cells apoptosis, lost mitochondrial membrane potential, activated caspase-3, caspase-8 and caspase-9, up-regulate Bax, tBid protein, down-regulation Bcl-2 protein. However, beta-sitosterul had hardly any effects on QSG7701 cells. METHOD A GC method was established and the contents of volatile oil and beta-elemene were determined in Sarcandrae from different length of time to grow, different medicament portions, different collection periods and different length of time to dry. RESULT The length of time to grow had no significant effect and the collection periods had effect on the volatile oil yield and content of beta-elemene. The volatile oil yield and content of beta-elemene were the highest in Sarcandrae harvested in December. The yield of volatile oil in different medicament portions descended in an order of roots, leaves and stems. The content of beta-elemene was the highest in leaves and the lowest in stems. With the increasing of the length of time to dry, the volatile oil yield and content of beta-elemene decreased calculated on an anhydrous basis. CONCLUSION The established method is simple, accurate and repeatable. It can be used for the quality control of beta-elemene in Sarcandrae. METHOD: Wistar rats were randomly assigned to two groups, nomal group and BXD group After evaluated by macro-behavior observation and open-field test, MTT assay and the whole-cell patch clamp recording were performed respectively to evaluate the effects of serum from BXD capsule-treated rats on the viability and GABA(A) R-induced currents of cortical neurons in vitro. RESULT: In the open-field test, the crossing score, rearing score and total score of BXD rats decreased significantly (P < 0.05), compared by the normal rats. Compared with cells exposed to serum of normal rats, the viability values of those incubated with serum of BXD group for 24 h and 48 h significantly increased (P < 0.05), measured by MTT assay. The results of whole-cell patch clamp recording showed that concentration-response relationship curves revealed an EC50 value of (29.0 +/- 4.4) micromol x L(-1) and a Hill coefficient of 1.07 for normal-exposed cultures, (63.5 +/- 8.2) micromol x L(-1), 1.04 for BXD-exposed cultures after incubation for 24 h. Furthermore, the difference in EC50 values was statistically significant (P < 0.01), that in the Hill coefficient was not obvious. CONCLUSION: BXD capsule could significantly decrease crossing score and total score of open-field test and effectively enhance the neuron viability and GABA(A) receptor activity in rat cortex. METHOD The compounds were isolated and purified by silica gel, macroporous adsorption resin and Sephadex LH-20 column chromatographic methods. Their chemical structures were elucidated on the basis of physicochemical properties and spectral data. RESULT Fourteen compounds were isolated and identified as syringic acid (1), 3-methoxy-4-hydroxycinnamic acid (2), vanillic acid (3), 4-hydroxybenzoic acid (4), mauritianin (5), (+)-xylopinidine (6), (+)-oblongine(7), (+)-tembetarine (8), eythritol (9), D-mannitol (10), ethyl-beta-D-glucopyranoside (11), (+)-magnoflorine (12), stepharanine (13), (2S, 4R)-4-hydroxy-2-piperidine-carboxylic acid (14), respectively. METHOD: The trace elements, the structure, and the thermal stability of Nanhanshuishi were assayed and calculated by X-ray fluorescence spectrometry (XRF), inductively coupled plasma optical emission spectrometer (ICP-OES), atomic fluorescence spectrometry (AFS), inductively coupled plasma mass spectrometry (ICP-MS), X-ray power diffraction (XRD), TG-DTA. RESULT: The results indicated that the phase is mainly made up of CaCO3 (Rhombohedral, R-3c) in Nanhanshuishi. The analysis of elements show that Nanhanshuishi is rich in Ca and O, and contains other more than 20 minor elements, such as Si, Mg, Fe, Al, Na, K, Zn, Mn, Pb, As, Hg etc. The result of TG-DTA show that the weight of Nanhanshuishi starts to decline from near 700 degrees C and get steady above 850 degrees C. METHOD: A water stress experiment with pot culture was applied to study the effects of different soil water content on the seedlings growth, biomass and the accumulation of tanshinone, salvianolic acid B and mineral nutrition of S. miltiorrhiza. RESULT: Soil water content had serious influence on the growth, yield, outer appearance and inner quality of S. miltiorrhiza when the soil was under severe drought or with too much water. But the shoot and root biomass of S. miltiorrhiza was increased significantly under mild drought. As well as the content and cumulant of dihydrotanshinone I , cryptotanshinone, tanshinone I , tanshinone IIA and salvianolic acid B in root of mild drought were increased. It also enhanced the P, K, Ca, Mg, Mn, Zn, Cu and Fe contents of S. miltiorrhiza under mild drought. CONCLUSION: The overall results from the experiment suggest that the appropriate soil water content is 55% to 60% in seedling stage of S. miltiorrhiza.
