This study investigated the predictive value and clinical associations of inflammatory markers and depression in cancer patients using machine learning (ML) and statistical modeling. Pancreatic cancer patients (n = 328; mean age, 65 years; majority with stage IV disease) were assessed using the Patient Health Questionnaire-9 (PHQ-9; depression defined as PHQ-9 ≥ 10). Clinically significant depression was present in 35% of subjects at baseline, and the rate declined at follow-up. Four ML models (logistic regression, random forest, support vector machine, and extreme gradient boosting; XGBoost) were trained using routinely collected clinical data and showed comparable performances with moderate but consistent discriminative capacity (AUC: 0.70-0.72). Permutation importance analysis revealed C-reactive protein (CRP), neutrophil-to-lymphocyte ratio (NLR), and albumin as key predictors of depression. Generalized estimating equations further confirmed that elevated CRP (OR = 1.32, p = 0.001) and NLR (OR = 1.55, p = 0.001) were independently associated with depression. These findings suggest that inflammatory markers can not only help to identify patients at risk for depression but also underscore the linkage between inflammation and depression. Beyond merely impeding regular respiration, adverse events (AEs) such as hemothorax or pneumothorax can hinder the patient's recovery. Herein, we aim to validate potential predictive factors for AEs among adults with BCT who were admitted concurrently through the dataset focusing on the limited information available upon their arrival at the emergency department (ED). Seventeen variables-including patients' demographics, comorbidities, and vital signs/hemogram data upon arrival at the ED-were investigated. A penalized logistic regression model was applied to the derivation cohort and validated in a subgroup using the same dataset (80%:20%). In addition, we employed the least absolute shrinkage and selection operator (LASSO) logistic regression to develop a nomogram, which enhances the accuracy of estimating individual probabilities for AEs after admission for BCT. Our retrospective review encompassed 3,668 adult patients between 2017 and 2021, and the incidence of AEs was 15.6% (572 out of 3,668). Penalized logistic regression was conducted both without and with the hemogram data (Model 1 and Model 2), yielding relatively satisfactory results (R<sup>2</sup>: 0.271 vs. 0.291; area under the curve: 0.784 vs. 0.797, respectively). Despite the model's relatively high predictive value in the derivation cohort, the validation data still maintained an acceptable accuracy of 0.7456 and 0.7049, respectively. Employing our penalized logistic regression analysis, the recently formulated nomogram exhibited proficiency in predicting AEs following BCT. However, the impact of environmental and seasonal factors on SPM remains unclear. This study investigated their association with SPM onset and clinical outcomes. We conducted a 12-year retrospective review of SPM cases, comparing clinical characteristics and outcomes between patients with and without pneumorrhachis. A case-crossover design was used to assess short-term associations between environmental exposures and SPM incidence, analyzed via conditional logistic regression. A total of 70 consecutive patients were identified, with 9 classified as SPM with pneumorrhachis and 61 as SPM without pneumorrhachis. While both groups were predominantly managed with hospitalization, those with pneumorrhachis had longer hospital stays (median: 7 vs. 3 days, p = 0.002) and were more often associated with severe-grade SPM and identifiable triggers (p < 0.001 and p = 0.009, respectively). No significant environmental exposure differences were observed between groups. Seasonally, SPM incidence was significantly higher in autumn and winter (p < 0.001), consistent with elevated air pollutant levels. Linear regression showed that standardized β coefficients for PM<sub>2.5</sub> were higher in autumn and winter (β = 1.15 and β = 1.18), indicating a seasonal association between PM<sub>2.5</sub> and SPM onset. Despite experiencing more triggers and longer hospitalization, patients with pneumorrhachis had similarly favorable clinical courses. This study aimed to elucidate the mechanism of Kruppel-like factor (KLF1) in NSCLC progression. Clinical samples were collected, after which the expression levels of KLF1, LINC02159, and dynein cytoplasmic 1 heavy chain 1 (DYNC1H1) in tissues and cells were initially detected, and NSCLC cell proliferation and invasion were measured when KLF1 was up- or downregulated. The binding relationships among KLF1, the LINC02159 promoter, DYNC1H1, and serine and arginine-rich splicing factor 1 (SRSF1) were analyzed. The colocalization of LINC02159 and SRSF1 was verified. DYNC1H1 stability upon actinomycin D treatment was assessed. Combined experiments were designed to confirm the interaction of the LINC02159/DYNC1H1 pathway in NSCLC development. Finally, xenograft tumors were generated in nude mice to validate the mechanism involved. KLF1, LINC02159, and DYNC1H1 were upregulated in NSCLC tissues and cells. KLF1 overexpression promoted NSCLC cell proliferation and invasion, whereas KLF1 knockdown inhibited NSCLC cell proliferation and invasion. Mechanistically, KLF1 transcriptionally activated LINC02159, which could recruit the SRSF1 protein and increase DYNC1H1 mRNA stability in the cytoplasm. Combined experiments revealed that LINC02159 and DYNC1H1 overexpression could counteract the inhibitory effect of KLF1 silencing on NSCLC cell proliferation and invasion. Since initial nodule volume may impact the efficacy of radiofrequency ablation, this study evaluated its long-term outcomes across varying nodule sizes, focusing on regrowth, new growth, and clinical management implications. This retrospective study included 160 patients who underwent thyroid radiofrequency ablation for benign thyroid nodules at a Taiwanese tertiary center between July 2016 and April 2018. Patients were classified into three groups based on nodule size: small (< 10 mL), medium (10-30 mL), and large (> 30 mL). Treatment efficacy was assessed over a period of up to 5 years, focusing on volume reduction rate, regrowth, residual volume, and new growth. The initial ablation rate of all benign thyroid nodules was 99.46%. After the 5-year follow-up, the volume reduction rate was 92.96%. The small nodule group demonstrated the highest volume reduction rate. The incidence of increased residual vital volume was 3.57%. The overall regrowth rate was 9.82%, with a mean time to regrowth of 2.8 years. No nodules required retreatment due to regrowth. New growth was observed in 22.32% of patients, with the highest incidence in the large nodule group (34.29%). Radiofrequency ablation is effective in the long-term management of benign thyroid nodules across various sizes, achieving substantial volume reduction rate with minimal complications. Although ozone therapy has shown protective potential against I/R injury in animal models of various organs including skeletal muscle, its precise mechanistic underpinnings require further elucidation. This investigation evaluates the therapeutic potential of major ozonated autohemotherapy (MOAH) for skeletal muscle I/R injury management. Utilizing a rat femoral artery ligation/release model, we demonstrated that MOAH pretreatment substantially alleviated histopathological damage through hematoxylin-eosin/Masson staining analyzes, diminished skeletal muscle apoptosis via terminal deoxynucleotidyl transferase dUTP nick-end labeling and Western blot, and reduced tissue edema as quantified by wet weight ratios. Serum biomarker assessments confirmed decreased creatine kinase and lactate dehydrogenase levels with MOAH administration. In oxygen-glucose deprivation/reoxygenation (OGD/R)-treated L6 myoblast models, ozone pretreatment enhanced cellular proliferation capacity while attenuating apoptosis and mitochondrial dysfunction. Subsequent analyzes revealed ozone's regulatory effects on oxidative stress markers (MDA content, SOD and CAT activity) and inflammatory factors (TNF-α and IL-1β) across both in vivo and in vitro models. Mechanistic evaluations through Western blot and reverse transcription quantitative real-time polymerase chain reaction techniques identified MOAH-induced activation of the Nrf2/HO-1 signaling pathway, with observed abolition of protective efficacy under Nrf2 knockdown conditions.
