Orafidiya","doi":"10.15406/JAPLR.2021.10.00362","DOIUrl":"https://doi.org/10.15406/JAPLR.2021.10.00362","url":null,"abstract":"The aims of this study were to determine the emulsifying properties of Theobroma cacao pod husk ash (CPHA) methanolic extract combined with shea butter and explore the antibacterial activities and physicochemical characteristics of resulting emulsions toward the development of a topical antibacterial lotion formulation for shaving bumps treatment. The ash resulting from combustion of pod husks of freshly harvested ripe cocoa fruits was extracted with methanol and the extract evaporated to dryness. Shea butter was also extracted by traditional method from kernels from the shea tree. These natural-source materials were combined with pharmaceutical ingredients (buffer, viscosity enhancer, preservative) to develop fluid emulsion formulations. Stability characteristics (droplet size, viscosity, creaming, and pH) of the formulations were evaluated as well as their antibacterial activities against microorganisms isolated from after-shave bump swabs of adult male volunteers and against reference organisms; in order to select product(s) of best qualities suitable as shaving bumps medication. The prototype formulations exhibited suitable physicochemical properties and demonstrated inhibitory activities against several isolated shaving bump microbes and the reference organisms namely, Staphylococcus aureus, Bacillus subtilis and Pseudomonas aeruginosa. Two formulations were finally selected as having physicochemical and antibacterial qualities most suitable for shaving bumps therapy, which contained shea butter (20%), citrate buffer (5%), and parabens (0.3%), prepared using 5% CPHA extract solution with and without methyl cellulose (2%), respectively. It has antidepressant and anti-anxiety activities. This method's main aim and the objective was to develop and validate a bio-analytical method of Vilazodone in human plasma by LC-MS/MS (API-4000) and its application to estimate pharmacokinetics. Method: For reporting this investigation, Analyst software, 1.6.3 used. The mobile phase was acetonitrile with 0.1% formic acid as an organic solvent and Milli Q water with 10Mm ammonium acetate and 0.1% formic acid using the gradation method 7.0min run time. The calibration standard concentrations were 1.0 to 64 ng/ml. Plasma precipitation was by protein precipitation technique. Result: The accuracy of calibration concentrations of Vilazodone was 93.5-104.39% and stability study showed 96.41-106.71%, 94.77-96.36%, 92.22-101.38%, 94.15-98.47%, 93.95-95.75% remaining for freeze-thaw, short term, long term, benchtop and autosampler stability respectively. Recovery was to be 98.10-98.99%; the matrix factor was 0.94-0.96. The maximum plasma concentration of reference preparation was 13.445±2.842ng/ml (Cmax) at a time 6.792±0.846hr. (Tmax). The maximum plasma concentration of test preparation was 13.218±3.231ng/ml (Cmax) at a time 6.958±0.793hr (Tmax) The relative bioavailability of the test preparation was to be 94.66 % of that of the reference preparation. Conclusion: The present investigation was highly selective, sensitive, reproducible, low matrix effect, high recovery and low time-consuming method. This article provides information on reporting and interpreting the numerical results of a vaccine trial. It aims to be a practical guide for students and health care professionals. It gives a simple definition of the common terminology, the vaccine efficacy, how to calculate it, and how the confidence interval can be found (though formulae for the latter are not given here). USH1B is characterized by hearing loss due to abnormalities in the inner ear and vision loss due to retinitis pigmentosa. Here, we present the model of human MYO7A homodimer, built using homology modeling, and refined using 5 ns molecular dynamics in water. Global computational mutagenesis was applied to evaluate the effect of missense mutations that are critical for maintaining protein structure and stability of MYO7A in inherited eye disease. We found that 43.26% (77 out of 178 in HGMD) and 41.9% (221 out of 528 in ClinVar) of the disease-related missense mutations were associated with higher protein structure destabilizing effects. Overall, most mutations destabilizing the MYO7A protein were found to associate with USH1 and USH1B. Particularly, motor domain and MyTH4 domains were found to be most susceptible to mutations causing the USH1B phenotype. These two drugs are currently used as drugs in first line treatment of patients with Acquired Immunodeficiency Syndrome (AIDS). In order to predetermine parameters for extrusion and the maximum concentration of API to be used without any recrystallization, binary blends were first processed in the mixing chamber at 130°C using a roller type rotor at 10, 20 and 30 rpm for 7 min, giving rise to ASD of both API, at least up to 20 wt% of the drug. Both 3TC and TDF were then extruded individually with Kollidon in a single screw extruder. HME produced ASD with high concentration of both Kollidon/API combinations, being part of the API soluble in the polymer matrix. It is highly prevalent global disease; affecting approximately 300 million individuals worldwide. Different medicinal plants have been discovered and used in traditional medicine practices for the treatment of asthma in different part of the world since prehistoric times. The aim of this review article is to explore and compile the ethno medicinal uses, phytochemistry and method of preparation of commonly used medicinal plants for the management of asthma in Ethiopia. A comprehensive and systematic web-based search of full length experimental, ethno-botanical and ethno medicinal survey articles from scientific database like Pub Med, Science Direct, Web of Science and Google Scholar, were undertaken using key words: asthma, medicinal plants, ethno-botanical, herbal, phytochemistry so as to document this review article. About 23 medicinal plants like Papilionaceae, Caricaceae, Moringaceae, Liliaceae, Asteraceae etc. were found among the most common used medicinal plants in the society for the treatment of asthma. A good number of contemporary medicines have evolved from traditional medicines thus in the present circumstances, our traditional system must be given an objective and critical examination. Further research must be carried out to determine the fraction(s)/compounds that are mostly responsible for the observed anti-asthmatic activity as this will lead to drug development. As well, the potency of plants used in combination as beside being used individually should be further researched. Seven varieties of lipase (PPL, Candida rugosa, OS cepacea, OS cepacea immobilized on diatomaceous earth, Novozyme 435, Lipozyme and Lipozyme RM) and seven enzymatic sources from a vegetable origin (latex of the species Ficus benjamina, Manihot glaziovii Muell. Arg., Calotropis procera, Jatropha curcas L., Ficus pumila, Euphorbia tirucalli L.) were tested. Enzymatic sources of plant origin were submitted, initially, the hydrolytic activity tests to evaluate their potential as biocatalysts and, from the results obtained, two samples were selected for the experiments further synthesis of biodiesel. Validation of the method of production of biodiesel evaluated linearity, reproducibility (precision), recovery (accuracy) and the limits of detection and quantification. Among the sources of enzymes used in the synthesis of biodiesel presented the best result was the commercial enzyme Novozym 435 with 55.33% conversion yield of biodiesel in initial tests. OCA1 exists in two forms: OCA1A and OCA1B. OCA1A is caused by a full loss of the human tyrosinase protein (Tyr), leading to an absence of pigment in skin, hair, and eyes, while OCA1B has reduced Tyr catalytic activity and pigment. The current understanding of the disease is hampered by the absence of information regarding the alterations of protein structure and the effects leading to either form of OCA1. Here, we used computational methods to find a general mechanism for establishing this link. Tyr and mutant variants were built through homology modeling, glycosylated in silico, minimized, and simulated using 100 ns molecular dynamics in water. For OCA1B mutants, cavity size is linked to ΔΔG values for mutants, suggesting that partial loss of Tyr is associated with the destabilizing effect of the EGF-like domain movement. In OCA1A, active site mutation simulations indicate that the absence of O2 leads to protein instability. OCA1B mutants are described in severity by the size of the cavity within the EGF–Tyr interface, while active site OCA1A mutants are unable to fully coordinate copper, leading to an absence of O2 and Tyr instability.
