Gupta","doi":"10.9734/bbj/2016/20401","DOIUrl":"https://doi.org/10.9734/bbj/2016/20401","url":null,"abstract":"Aim: To produce bovine herpesvirus-1 (BoHV-1) vaccine strains in MDBK cells using an oscillating bioreactor, BelloCell. Place and Duration of Study: Division of Veterinary Biotechnology, Indian Veterinary Research Institute, Izatnagar-243122, India, between November 2013 and June 2015. Methodology: MDBK cells were cultured in BelloCell bioreactor seeded with MDBK cells. After 6 days incubation at 37°C under 5% CO 2 tension, the MDBK cells were infected with 0.001 multiplicity of infection (MOI) of either wild-type (wt)-BoHV-1 or glycoprotein E deleted (∆gE)-BoHV-1vaccine strains and incubated further for 96 h. To analyse the vaccine potential of the harvests, inactivated wt-BoHV-1 and ∆gE-BoHV-1 were inoculated in guinea pigs. Sera from immunized guinea pigs were collected at 90 days after primary immunization and analysed for virus neutralizing (VN) antibody response specific to wt-BoHV-1. Results: Using BelloCell bioreactor, the MDBK cell density reached to a maximum number of Original Research Article Rajan et al.; BBJ, 12(1): 1-7, 2016; Article no.BBJ.20401 2 1.5 x 10 to 2.0 x 10 cells after 6 days cultivation. After infection with wt-BoHV-1 and ∆gE-BoHV-1 strains, the total virus yield at 96 h post-infection was 0.5 x 10 and 0.5 x 10, respectively. There was no significant difference in VN antibody response in sera collected from immunized guinea pigs. Aims: To screen for resistance 10 improved and 4 local cultivars of taro against taro leaf blight disease. Study Design: A randomized complete block design study. Place of Study: Studies were conducted at the International Institute of Tropical Agriculture (IITA) Yaounde Nkolbisson from July 2013 to January 2014. Methodology: Taro cultivars from tissue culture were planted in the screen house conditions and Original Research Article Charles et al.; BBJ, 15(1): 1-15, 2016; Article no.BBJ.14317 2 tested for virulence and pathogenicity with 4 isolates of Phythophthora colocasiae at spore density of 3×10 4 spores /ml of distilled water. Plants were planted in the field to assess disease incidence and severity. Results: The results obtained on the different taro cultivars, revealed that all the 4 isolates showed variable pathogenicity. They caused lesions on inoculated leaves. There was variability in pathogenicity based on the small lesion lengths produced on cultivars, these included BL/SM132 and Red petiole. Isolate 3 showed a stronger sensitivity to leaf collapse and defoliation irrespective of the cultivar tested. There was a significant difference (p = 0.05) in tissue collapse and leaf defoliation on exposure to the different fungal isolates. The result of field infection rates of P. colocasiae at 126 DAP-154 DAP on 10 improved and 4 local cultivars indicated that there was significant variability (p = 0.05) in incidence and disease severity, with high incidence and severity occurring at 154 DAP in all cultivars. Improved cultivar BL/SM132 showed no classic symptoms of P. colocasiae and therefore it was resistant to Phytophthora colocasiae. Conclusion: The results obtained on virulence and pathogenicity of Phythophthora colocasiae on the different taro cultivars revealed that all the 4 isolates showed variable pathogenicity. They caused lesions, on inoculated leaves. Isolate 3 showed a stronger sensitivity to leaf collapse and defoliation irrespective of the cultivar tested.
