Identification of mumps-specific IgM antibodies in serum or plasma by enzyme immunoassay analysis (EIA) in samples taken during the acute phase is the most-widely used serologic test for the diagnosis of mumps, due to its simplicity, high sensitivity and specificity. When the infection cannot be confirmed by serologic tests, the diagnosis can be made by direct methods such as isolation of the virus in cell cultures or detection of the viral genome by molecular analysis This work describes these three methods: serologic testing, cell culture and gene amplification.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"3 1","pages":"86-88"},"PeriodicalIF":0.0,"publicationDate":"2010-02-16","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104265","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, various transmission chains occurred in the indigenous population after the introduction of an imported case, and this caused a total change in the existing situation of elimination. The objective of this study was to analyze the epidemiological and clinical characteristics of the measles outbreak in Catalonia in 2006. Cases reported between August 28 2006 and July 8 2007 were collected. Cases were microbiologically confirmed by determination of measles-specific IgM and/or detection of the viral genome in urine samples. A total of 381 cases of measles were confirmed, representing an incidence of 6.6 per 100,000 inhabitants. Laboratory confirmation was made in 87.1% of the cases and all specimens sequenced belonged to the D4 genotype. A total of 89.5% of the cases occurred in non-vaccinated people, above all in infants aged <16 months.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"3 1","pages":"42-47"},"PeriodicalIF":0.0,"publicationDate":"2010-02-16","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104461","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Both dominance of a single genotype and co-circulation of different genotypes in the same geographical area, as well as temporal replacement of genotypes have been described in different countries. The different histories of genotype importation, variations in vaccine coverage and the use of different vaccine strains in each country results in a complex picture that could be the cause of the different geographical patterns of mumps virus genotype circulation observed in different countries. Lack of full cross-protection between different genotypes has been reported and has been suggested as a cause of vaccine failure, especially for vaccine strains belonging to genotype A, which is genetically distant to the remaining genotypes that include most of the currently circulating wild strains. Finally, a differential ability to invade the neural system has been suggested for some particular strains belonging to genotype D.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"3 1","pages":"37-41"},"PeriodicalIF":0.0,"publicationDate":"2010-02-16","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104130","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Vaccines against infectious diseases with a human reservoir can have the following beneficial effects : control, elimination, eradica- tion and extinction of the disease. Vaccines have facilitated the eradication of smallpox in 1980 and a massive reduction in cases of poliomyelitis, of which only 1,655 were recorded worldwide in 2008. The elimination of poliomyelitis in the European region was certified by the WHO in 2002. The essential conditions for the elimination of communicable diseases are political commitment, the implementation of vaccination programmes and exhaustive disease surveillance.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"3 1","pages":"19-24"},"PeriodicalIF":0.0,"publicationDate":"2010-02-16","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104572","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The measles virus was detected by culture in 31% of cases. The genotype was determined in 90.7% of positive samples, all of which were the B3 genotype (subgenotype B3.1), with sequencing in various genes that indicated the same viral lineage. The outbreak lasted 198 days. The age range was 2 months -37 years and 84.6% of cases were Spanish-born. A total of 9.8% of cases presented complications, 37.4% required hospital admission, and 9.2% had a documented history of immunization. Twenty-four clusters were identified in families, health workers and infant schools. Conclusions: Control measures included advancing the infant MMR vaccination schedule and reinforcement vaccination in health care workers. While the measles virus still circulates, surveillance of suspected cases and maintenance of high vaccination coverages, especially in health care workers, are essential.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"3 1","pages":"48-54"},"PeriodicalIF":0.0,"publicationDate":"2010-02-16","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104474","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Eight genotypes have been identified (47% of European origin). Hospitalization was required in 18% of cases and the most common complication was otitis. One death has been recorded. Methods: Analysis of the evolution of rubella incidence and vaccination coverages (l980 and 2007). Morbidity data were obtained from the National Epidemiological Surveillance Network and mortality and population data from the National Institute of Statistics. Results: In 1979, a policy of selective rubella vaccination was introduced in Spain, targeting 11-year-old girls. In 1981, immunisation with the measles-mumps-rubella vaccine was introduced, at 15 months of age and a second dose was introduced in 1995. Vaccination coverages since 1999 exceed 95% for the first dose and 90% for the second. The incidence of rubella has fallen drastically to < 1/100,000 since 2000 in all regions. Only small outbreaks or clusters have been detected, except for one large outbreak in Madrid that affected young adult males and fertile women from countries with low vaccination coverages. Since 1997, 13 cases of congenital rubella syndrome have been detected. Conclusions: The situation in Spain favours the elimination of rubella. However, pockets of susceptible subjects remain, mainly young adult males and immigrants, who have increased substantially in numbers in the last decade. Fertile women with high birth rates from countries with a high rubella incidence have a high risk of congenital rubella syndrome.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"95 1","pages":"55-59"},"PeriodicalIF":0.0,"publicationDate":"2010-02-16","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104163","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The tegumental surface of adult worms presents Sm60, a mannose-binding protein, which induces neutrophil migration and mast cell degranulation. Here we demonstrated that Sm60 is able to block the in vitro antigen-specific or polyclonal spleen cell proliferation induced by keyhole limpet haemocyanin (KLH) or con- canavalin A (Con A), respectively. To address the mechanism of inhibition, we evaluated some cytokines in culture and observed that Sm60 decreased significantly the synthesis of interleukin (IL)-2 induced in response to KLH, but not other cytokines. To block the suppression triggered by Sm60, exogenous IL-2 or α-methyl-mannoside were added to KLH- stimulated cultures pulsed with Sm60. Only IL-2 abolished the Sm60 effect, suggesting that Sm60 blocked the lymphop- roliferation through the inhibition of IL-2 production, in a carbohydrate recognition domain (CRD)-independent mecha- nism. Our results suggest that Sm60 could participate in the immune evasion mechanisms of S. mansoni.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"3 1","pages":"1-6"},"PeriodicalIF":0.0,"publicationDate":"2010-01-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104282","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Most of these vaccines were developed empirically and there has not been a clear understanding of the immunological principles that contribute to this success. Recent advances in systems biology are being applied to the study of vaccines in order to determine which immunological parameters are the best predictors of success. New approaches to vaccine development include the identification of peptide epitopes and the manipulation of the immune response to generate the most appropriate response. Vaccines are being developed to prevent and/or treat such conditions as cancer and autoimmunity in addition to infectious diseases. Vaccines targeting this diverse group of diseases may need to elicit very different types of immune responses. Recent advances in our understanding of the functions of dendritic cells (DC) and cytokines in orchestrating qualitatively different immune responses has allowed the design of vaccines that can elicit immune responses appropriate for cancer, autoimmunity or infectious organisms. We previously observed that polyphosphazenes enhance immune responses in mice when injected subcutaneously against X:31 influenza antigens. Here, we investigated the potential of polyphosphazenes as mucosal adjuvants for enhancing influenza-specific immune responses. Vaccine combinations with soluble polyphos- phazenes (PCPP or PCEP) and influenza X:31 antigen were administered to BALB/c mice intranasally. Antigen-specific antibody responses were assayed in serum and mucosal washes by ELISA, while antigen-specific cytokine production was assayed in spleen cells by ELISPOT assay. We observed that the formulation of either of the two polyphosphazenes with X:31 induced significant and prolonged serum IgG1 antibody responses as early as 2 weeks after primary immuniza- tion. Interestingly, only PCEP + X:31 induced a significant and prolonged serum IgG2a response. These results implied that the PCEP+X:31 formulation induced better Th1 immune responses, suggesting increased cell-mediated immunity. To confirm this, we determined that IFN- , a Th1 cytokine, was produced at significantly higher levels in spleens from mice that were vaccinated with the PCEP+X:31 formulation, while IL-4, a Th2 cytokine, was produced at higher levels from both vaccinated groups. Finally, we determined that these polyphosphazenes were indeed effective as adjuvants in induc- ing mucosal immune responses, as IgA and IgG antibodies were detected in lung and vaginal washes. We conclude that polyphosphazenes increase mucosal immune responses effectively, and can be used to modulate the quality of Th1 and Th2 immune responses.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"2 1","pages":"134-143"},"PeriodicalIF":0.0,"publicationDate":"2009-11-18","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104240","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
