Two prophylactic vaccines containing combinations of the major capsid protein (L1) of HPV16 and HPV18 have been shown to efficiently prevent infection by inducing capsid-specific neutralizing antibodies. Since the year 2006 these vaccines have been implemented in many countries in the hope that the incidence of cervical cancer will be drastically reduced in coming decades. Nevertheless, the real efficacy of the present HPV vaccines in preventing cervical cancer is not known. This review summarizes the clinical studies that led to assess the prophylactic power of these vaccines and discusses some open questions and controversy on cervical cancer prevention in relation with the HPV vaccine.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"2 1","pages":"123-133"},"PeriodicalIF":0.0,"publicationDate":"2009-08-27","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104199","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This descriptive qualitative study of HPV vaccine policy development used an iterative, inductive, theme-based approach to data analysis. The study was conducted in four developing countries—India, Peru, Uganda, and Vietnam. Study participants were comprised of a total of 237 national policymakers, legislators, officials, and senior managers from ministries of health, finance, and planning; leaders of medical and health professional associations; cancer institutes; heads of nongovernmental organizations; and women's health advocates. While differences existed among low-income countries in specific cervical cancer, women's health, adolescent health, or immunization policy environments, we found the policymaking process itself, specific concerns related to HPV vaccines, and the information needs of policymakers for HPV vaccine introduction to be strikingly similar. Data on burden of cervi- cal cancer, HPV vaccine safety and efficacy, and cost-effectiveness and vaccine affordability were top issues reported by policymakers. Advocacy strategies need to address these issues in order for HPV vaccine policy formulation and approval to be successful.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"2 1","pages":"113-122"},"PeriodicalIF":0.0,"publicationDate":"2009-08-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104157","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Certain diseases have haunted humanity for centuries but are now extinct due to vaccines. On the other hand, some diseases such as salmonellosis, that were uncontrollable in the past, still cause pandemics today. There are more than 2500 serovars of Salmonella and vaccines made from any one serovar do not confer cross-protection against another, no matter how much antigenic similarity there is between the two. Salmonella strains are able to cause disease and to adapt to different types of animals whilst still maintaining their zoonotic and interspecies transfer potential. Three major types of vaccines are being used to control salmonellosis: killed bacteria, subunit vaccines and live attenuated vaccines. Effective vaccines against some host adapted and common serovars have been developed but their use has led to the emergence of other serovars. The problem has become more complex because increased international trade and travel has helped Salmonella strains to cross continental boundaries. It seems unlikely that we will be able to develop an effective Salmonella vaccine in the near future that is able to control all forms of salmonellosis, even in a single animal species. Recent advances in Salmonella vaccines will be reviewed including the use of Salmonella as vector for delivery of multivalent DNA and recombinant vaccines for controlling salmonellosis and other infectious diseases as well as for the control of cancer.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"192 1","pages":"100-112"},"PeriodicalIF":0.0,"publicationDate":"2009-07-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104150","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Immunization of mice with the Streptomyces mycelium induced high levels of specific antibodies against proteins released in the culture supernatant of the analyzed strain. Splenocytes from the Streptomyces-immunized animals were able to secrete high levels of IFN- (2103.9 pg/mL) and to proliferate in vitro on stimulation with proteins released by Streptomyces. The analysis of cross-reactivity against M. tuberculosis secreted proteins showed that the immunization of mice with Streptomyces led to a comparable level of cross-reacting antibodies as in the BCG immunized mice. Similarly sera antibodies from Streptomyces immunized group recognized whole BCG cells to the same degree as antibodies raised against BCG reacted with Streptomyces mycelium, indicating that these two actinobacteria cross-react immunologically. The Streptomyces lividans strain was highly immunogenic in mice showing an enduring Th1-dependent immune response. This is the first report demonstrating the potential of Streptomyces as an attractive vehicle for developing a live TB vaccine.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"2 1","pages":"85-91"},"PeriodicalIF":0.0,"publicationDate":"2009-06-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104139","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The objective of this work was to present an up-to-date review of the efficacy and the trend of mandatory vaccination regimens among children in Egypt, and its efficacy in decreasing the burden of vaccine-preventable diseases among children under 5 years in Egypt during the period 1995-2005. The sources of data depend on the bibliographic review on MEDLINE, published studies and reports, and WHO and EMRO databases that covered the period from 1995 to 2005. Statistic analysis of the available data revealed that the coverage rate of the existing vaccination program in Egypt has greatly expanded over years and is significantly associated with a swift reduction in the burden of the vaccine-preventable childhood diseases (measles, mumps, rubella, neonatal tetanus, pertussis, diphtheria, poliomyelitis, and hepatitis B). Varicella vaccine is strongly recommended to be added to the mandatory vaccination of children in Egypt.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"2 1","pages":"77-84"},"PeriodicalIF":0.0,"publicationDate":"2009-06-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104426","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In the past 5 years, our newly developed CLIPS TM technology has proven an extremely valuable tool for 1) binding site mapping of therapeutically relevant mAbs, 2) generating hyperimmune sera via immunization with CLIPS peptides, and 3) the generation of monoclonal antibodies (mAbs) via the use of hybridoma technology. We currently have data available for more than 50 therapeutic targets. For this reason, genetic modification of DCs functions is an attractive approach to treat disease, both using mature DCs (mDCs) to immunize patients, or immature DCs (iDCs) to induce tolerance. Viral vectors are efficient at transducing DCs, and we have investigated the effect of transduction with viral vectors on the phenotype and function of DCs. Adenovirus (Ad5), and Lentivirus (LV) are able to up-regulate costimulatory molecules and major histocompatibility complex (MHC) class II expression on DCs, as well as inducing production of Th1 and proinflammatory cytokines. Moreover, the function of virally infected DCs is altered. In fact, iDCs have an increased, and mDCs a decreased, ability to stimulate a mixed lymphocyte reaction (MLR). Similar results were observed when the capability of transduced-DCs to present recall- antigen was investigated. Of interest, in the context of vaccine design, these vectors were able to induce an antigen- independent T lymphocytes proliferative response. These data are relevant not only in the context of genetic manipulation of DCs for vaccine development and active immunotherapy, but also in our understanding of the response of DCs to viral infection.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"2 1","pages":"68-76"},"PeriodicalIF":0.0,"publicationDate":"2009-06-12","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104409","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In order to induce maximal protective mucosal immunity, mucosal adjuvant is essential. Current studies are searching vaccinal antigens that are derived from the outer membrane protein (OMP) of Neisseria meningitidis. This study investigated the immunogenicity of intranasally administered class 5C protein of Neisseria meningitidis B employing purified LPS of the immunotypes (L3, 7, 9 or L8), cholera toxin and whole cells of Bordetella pertussis as mucosal adjuvants for the development of new meningococcal vaccine. In conclusion, the nasal delivery of class 5C with mucosal adjuvants has considerable potential in the development of a mucosal vaccine against serogroup B meningococci in a prime boost immunization in the presence of the adjuvants used. A monoclonal antibody against class 5 C protein using popliteal lymph nodes was obtained.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"5 10 1","pages":"45-55"},"PeriodicalIF":0.0,"publicationDate":"2009-05-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104561","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

An attenuated VZV vaccine has been approved for general immunization to prevent varicella and zoster in the United States. Although this vaccine provides a high degree of protection against virus infection, the virus becomes latent in dorsal root ganglia and will reactivate to produce zoster. Therefore, there is a need for developing additional VZV vaccines that are capable of eliciting immune response to the virus but will not establish viral latency. Our studies have been focused on the development of a truncated secretory VZV glycoprotein (VZVgE) subunit vaccine. The results from our studies have shown that the VZV subunit vaccine: (1) elicits the induction of neutralizing antibodies in animals as well as in humans; and (2) stimulates the induction of VZVgE-specic antibodies in VZV-seropositive human mononuclear cells. Such a VZV glycoprotein antigen, therefore, may have the potential to be used as a candidate VZV glycoprotein subunit vaccine for prevention of primary VZV infection (Varicella) or boosting immune response against VZV reactivation (Zoster) in adults, the elderly and immunosuppressed individuals.","PeriodicalId":88763,"journal":{"name":"The open vaccine journal","volume":"2 1","pages":"17-27"},"PeriodicalIF":0.0,"publicationDate":"2009-04-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68104550","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
