Earthworms were exposed to soils contaminated with different concentrations for various duration. Residues were monitored in soil and earthworms after 7, 15, 30, 45, and 60 days of exposure to different isoproturon concentrations. Acute toxicity of isoproturon was determined together with growth rate and total soluble protein content of worms. These parameters were related to isoproturon concentration in soil and earthworms. No lethal effect of isoproturon was observed even at the highest concentration tested (1.4 g/kg soil) after 60 days after treatment. Residues of isoproturon have caused a significant reduction of the growth rate. Additionally a reduction of total soluble protein was observed in all treated worms. Decrease of isoproturon concentration in soil was slow and depended on herbicide initial concentration. In the worms, it increased during the first 15 days and decreased thereafter. This study is suggesting the use of the growth rate of earthworms as biomarker of exposure to isoproturon.</p>","PeriodicalId":85134,"journal":{"name":"Mededelingen (Rijksuniversiteit te Gent. They were homogenized, mixed and stored at constant humidity in a non-heated room. Sub-samples of soils were subsequently collected at regular intervals, dried and sieved through a 1.8-mm mesh before DNA was extracted. Specific primers were then used for the detection of plant DNA by hot start PCR. Results reveal that, under laboratory conditions, transgenic and non-transgenic sugar beet DNA was still detected after 25 days incubation in the soil taken from a sugar beet experimental plot while detection of chicory DNA was still possible after 50 days incubation in soil taken from the chicory experimental plot. This might be in correlation with the stronger resistance of chicory radicles to decomposition as compared to radicles from sugar beets.</p>","PeriodicalId":85134,"journal":{"name":"Mededelingen (Rijksuniversiteit te Gent. All samples were washed and the nematodes were extracted by Centrifugal Flotation Technique. They were fixed and transferred to glycerin, then permanent slides were mounted. Morphological and morphometrical characters of each species were studied. In this study 31 species belonging to 17 genesa of the order Tylenchida were identified as follows. Amplimerlinius macrurus, Anguina tritici, Aphelenchoides centralis, A. limberi, A. parietinus, Aphelenchus avenae, Boleodorus thylactus, Ditylenchus dipsaci, D. destructor, D. kheirii, D. medicaginis, D. myceliophagus, Filenchus cerealis, Irantylenchus clavidorus, Helicotylenchus cavenessi, H. dehystera, H. digonicus, H. pseudodigonicus, H. pseudorobustus, Heterodera avenae, Meloidogyne incognita, M. javanica, Paratylenchus projectus, P. tateae, Pratylenchus neglectus, P. thornei, Psilenchus hilarulus, P. iranicus, Quinisulcius capitatus, Seinura oostenbrink and Zygotylenchus quevarai. Special investigations have been carried out to clear up the mechanisms of spore-adhesion at the insects with fluorescence-microscopy. After direct soil applications we have found a lot of spores/larva while applying low spore-concentrations. The loading with spores after indirect applications was very high as well. Glucosinolates (GLS) and their degradation products from Brassica species are attractants and feeding stimulants for Brassicaceae specialist insects but are generally repellent and toxic for generalist herbivores. The impact of these compounds on crucifer specialist insects are well known but their effect on generalist predators is still not well documented. Prey host plant influence on reproduction of an aphidophagous beneficial, the two spot ladybird, was determined using the cabbage aphid, Brevicoryne brassicae reared on a crucifer plant, namely Brassica napus containing low GLS levels. As ladybird developmental and reproductive parameters were already shown to be strongly affected by the allelochemical presence in its preys, the unsuitable aphid and host plant combination was only momentary used to feed the A. bipunctata adults. A strong impact of the diet was observed on the beetle fecundity and the emerging offspring. Changing B. brassicae aphid to a suitable prey slowly improved the temporary negative effect of the former diet. These results enhance the food environmental effect and the importance of tritrophic relations in pest management strategies by predators. Indeed, more than the choice of the beneficial species, the prey host plant has a major influence on the potential efficacy of biological agent to control herbivore species such as aphids.</p>","PeriodicalId":85134,"journal":{"name":"Mededelingen (Rijksuniversiteit te Gent. The main breeding programmes are focused on scab resistance conferred by monogenic sources of resistance, specifically the Vf gene originated from Malus floribunda 821. Since 1993, evidence of the breakdown of this resistance mechanism appeared in different countries like United Kingdom, Germany, The Netherlands and France. In 1997, a first evidence of the Vf gene breakdown in Belgium was observed on the cultivar 'Judeline' planted in an untreated orchard at Aubel where a cider making company, with our collaboration, search for cultivars adapted for juice and cider processing. Therefore, a glasshouse experiment was planned in order to characterize which scab race was present. In 1999, the artificial inoculation of some cultivars in controlled conditions showed that the scab race collected the year before at Aubel presented the same characteristics than race 7 described by other works. At the same time, an orchard was established by our Department as planned in an European FAIR project named 'Durable Apple Resistance in Europe' (D.A.R.E) with the aim to study the resistance durability of some partial scab resistant apple cultivars inside an European orchard network. Sixteen repetitions of 20 different cultivars are planted and are originating from 8 participating countries. Five apple clones with specific host resistance towards 4 specific scab races are also present. Incidence and severity assessments of the disease are made 3 times a year on the whole tree according to two international standardized scales. For the first time in 2000, a specific scab race was observed on Malus floribunda 821 which caused very serious injuries. Those results confirm that new breeding strategies must be developed for reducing the risk of scab resistance breakdown in the future.</p>","PeriodicalId":85134,"journal":{"name":"Mededelingen (Rijksuniversiteit te Gent. Worms were then exposed to these concentrations (LC10, LC25, and LC50) of endosulfan and (LC10 and LC25) of aldicarb. Growth rate, total protein content, and insecticide residues were determined. Aldicarb was more toxic than endosulfan and both insecticides caused a significant reduction of growth rate and total protein content of earthworms. Residues remaining in the soil after 2 to 15 days ranged between 37.75% and 68.54% of applied concentration for endosulfan and between 10.13% and 67.71% of applied concentration for aldicarb. Less than 1% of soil concentration was detected in worms and accumulation was more important in the case of endosulfan.
