The animals were divided into 3 groups namely; A, B and C and groups A and B were subdivided into 2; AI, A2, B1 and B2. All the groups and subgroups contained 6 albino rats each and were given water and food ad libitum. Groups A1 and B1 with A2 and B2 were treated with 1.85 mg kgG and 3.70 mg kgG of 90% ethylene glycol intraperitoneally once, respectively. Groups B1 and B2 were also given 1 mL of the plant extract, respectively by oral incubation for 21 consecutive days while group C received only the water and food. The average body weights of the treated groups decreased significantly at p<0.05 compared to control. The result showed no significant change at p<0.05 in total protein levels and significant decrease in albumin, total and direct bilirubin levels compare to control. Juice yields were increased greatly with enzymatic treatment (p<0.05), The amounts of flavonoid and polyphenols as well as clarity of the juices were greatly varied because of different enzyme treatment. Juice treated with pectinase and diastase showed better clarity and sugar content than the untreated juice (p<0.05). The relative viscosity of the juice were also decreased in clarified and fermented juice of both fruits. The mass spectra of fermented juice of M. citrifolia exhibits 14 important peaks out of which 4 peaks are having larger % area. Food products can contain non-halal components like porcine gelatine. One of food suspected to use gelatine is soft candy. Gelatin can be made from pork or beef or other animal. The presence of porcine gelatine in any food products is not allowed for Moslem community, therefore an analytical method offering reliable results must be developed. This study is intended to use Real-Time Polymerase Chain Reaction (RT-PCR) for analysis of porcine gelatine in soft candy. Isolation of DNA was performed with mitochondrial DNA Isolation Kit K280-50 (Bio-Vision). The DNA was analyzed by RT-PCR using primer D-Loop 318. Analysis for the primer specificity was performed on fresh tissue (pig, cows, chickens, goats and rats) and gelatin sources (beef, pigs and catfish). Primer D-loop318 can amplify porcine DNA at the optimum temperature 61.4°C. Repeatability test demonstrated amplification of all positive response samples containing porcine DNA in serial dilution of 10000-1 pg). The Coefficient of Variation (CV) is 6.32%. The repeatability test was also performed on soft candy 100% having CV of 1.06%. Parker, Uroko Robert Ikechukwu","doi":"10.3923/AJB.2016.24.33","DOIUrl":"https://doi.org/10.3923/AJB.2016.24.33","url":null,"abstract":"The study investigated phytochemical, antihyperglycaemic and lipid profile effects of methanol extract fraction of Ricinus comminus seeds in alloxan induced diabetic male Wistar albino rats alloxan. Eighteen mice were used for the lethal dose (LD50) study while 54 Wistar albino male rats were used for the antihyperglycaemic and lipid profile, respectively. The result of the phytochemical analysis showed the richness of the extract in important phytochemicals that could confer some health benefits. The methanol extract fraction had LD50>5000 mg kgG 1 b.wt., which indicate that the extract was safe to larger extent. The baseline blood glucose concentration showed non-diabetic glucose level 105.10±4.61 mg dLG. Diabetes was induced with alloxan monohydrate. Significant decrease (p<0.05) in blood glucose levels in the test animals were observed when compared with positive control. The extract had significant effect on the blood glucose levels and lipid profile of the treated rats closer to that of the standard drug (metformin) with the serum total cholesterol and triacylglycerol levels decreasing significantly (p<0.05) in all test groups relative to positive control. However, serum HDL showed significant increase (p<0.05) in group 4 and 5 when compared with positive control. The results of the study showed that the methanol extract fraction of Ricinus communis seeds exhibited significant antihyperglycaemic activities against alloxan induced diabetes in Wistar albino rats. Firstly, the antidiabetic activity of petroleum ether, chloroform, methanol and water extract of C. iners leaves was evaluated. Next, the potential extract which was the methanol extract was fractionated to obtain ethyl acetate, n-butanol, chloroform and aqueous fraction. The active chloroform fraction was subjected to subfractionation process to obtain subfraction 1 (SF 1) and subfraction 2 (SF 2). Cinnamomum iners leaves active portion (methanol extract, chloroform fraction and subfraction 1) was standardized using cinnamic aldehyde. The antidiabetic activity of standardized C. iners leaves was assessed by using Intra-Peritoneal Glucose Tolerance Test (IPGTT). The antidiabetic mechanism of active portion of C. iners leaves were also determined by measuring glucose absorption from the intestine. This study showed significant glucose tolerance effect of active portion of C. iners leaf after repeated oral administration in STZ-induced diabetic rats. The flavonoidal constiteuents in EEHh and GFHh was confirmed by HPLC. The behavioral analysis of forced swimming test, tail suspension test, locomotor activity in mice has been conducted. Brain superoxide dismutase, catalase, glutathione and lipid peroxidation were estimated biochemically. Brain monoamine oxidase (A and B) activity has been carried out by double beam spectrophotometer.
