Priya","doi":"10.3923/AJB.2016.59.67","DOIUrl":"https://doi.org/10.3923/AJB.2016.59.67","url":null,"abstract":"The present study was undertaken to evaluate the Ethanolic Extract of Tecoma stans (EETS) flowers for its protective effect on gentamicin-induced nephrotoxocity in rats. Nephrotoxocity was induced in rats by intraperitoneal administration of gentamicin 80 mg kgG dayG for seven days. Effect of concurrent administration of EETS at a dose of 500 mg kgG dayG given by intraperitoneal route was determined using serum and urinary creatinine and blood urea nitrogen as indicators of kidney damage. As nephrotoxocity of gentamicin is known to involve the induction of oxidative stress, so in vitro antioxidant activity, malondialdehyde (MDA) and glutathione (GSH) levels were also evaluated. Gentamicin-induced glomerular congestion and necrosis of kidney cells were found to be reduced in the group receiving EETS with gentamicin. Rats treated with gentamicin developed significant kidney dysfunction was observed from increased levels of urea, creatinine, sodium and decreased levels of protein, uric acid and potassium. In vitro studies revealed that the EETS posses significant antioxidant activity. Yousif","doi":"10.3923/AJB.2015.252.266","DOIUrl":"https://doi.org/10.3923/AJB.2015.252.266","url":null,"abstract":"Multipara metric analysis of compound toxicity at the level of individual cells using flow cytometry and cellular imaging-based approaches such as High Content Screening (HCS) have played key roles in the detection of toxicity and classification of compounds based on observed patterns of reversible and irreversible cellular injury. Gold III complex (AuL2) of bidentate ligand derived from the cyclization reaction of Schiff base of 4-amino-5-phenyl-4h-1,2,4-triazole-3-thiol with thioglycolic acid was synthesized and characterized by using melting point, FTIR spectroscopy, HNMR, UV-Visible spectroscopy and elemental analysis. The presence of chloride counter ion in complex was supported by conductivity measurement and the presence of hydrated water was supported by thermal gravimetric studies. We examine the cytotoxic effects of gold complex and its ligand in one cultured cellular models (MCF7 cell line) by High Content Screening (HCS) and analysis and cell viability assay (MMT assay). The inhibitory effect of AuL2 on breast cancer cell growth was due to induction of apoptosis as evidenced by annexin V staining and cell shrinkage. The study found that AuL2-mediated lead to disruption of Mitochondrial Membrane Potential (MMP), cell membrane permeability, nuclear condensation, fragmentation and release of cytochrome c from the mitochondria into the cytosol and also suggesting AuL2 as a potential MCF7 inhibitor compared to doxorubicin as positive control. This study investigated anticonvulsant and analgesic potentials of leaf and root extracts. The extracts were prepared with deionized water and ethylacetate. Extracts doses of 200, 400, 600 and 800 mg kgG b.wt. were used for the investigations. Anticonvulsant potential of the extracts against pentylenetetrazole-induced convulsion was tested in albino rats, by measuring time for convulsion onset, duration of convulsion and plasma glucose and Ca levels before, during and after convulsion, using diazepam as standard. The hot plate method was used to investigate the analgesic property of the extracts using morphine sulphate as standard. Pretreatment of the animals with different doses of the extracts delayed the onset of convulsion in a dose-dependent manner. Convulsion was not observed in 800 mg kgG b.wt. of Deionized Water Leaf (DWL) extract and in diazepam pretreated groups. The extracts decreased significantly (p<0.05) the severity of convulsion and prolonged the duration of convulsion induced by pentylenetetrazole. The plasma concentration of glucose and Ca decreased significantly (p<0.05) in the test groups through the course of convulsion, while they did not change significantly (p>0.05) in the non-convulsed groups. All the extracts at all doses and 4 mg kgG of morphine sulphate (an analgesic) showed a significant (p<0.05) percentage inhibition against hot plate induced pain. The difference between the analgesic potentials of 800 mg kgG of DWL extract and 4 mg kgG morphine sulphate was not significant (p>0.05). Group I served as control while group II, III and IV received 50, 100 and 150 mg kgG b.wt. dayG ammonium molybdate orally respectively for 60 days. Group V rats received 150 mg kgG b.wt. dayG for 60 days and thereafter left for recovery study for 60 days. The present study examined the effects of ammonium molybdate on serum testosterone, Follicle Stimulating Hormone (FSH) and Luteinizing Hormone (LH) along with sperm parameters. Estimations were also made for marker biochemical and antioxidant parameters in testis. Ammonium molybdate treatment resulted in a dose dependent reduction in serum testosterone, FSH, LH, sperm count, motility and viability. The concentration of total protein and sialic acid as well as activities of acid phosphatase (ACP) and alkaline phosphatase (ALP) were decreased in a dose dependent manner while there was a significant increase in the glycogen and cholesterol level in testis. The level of Thiobarbituric Acid Reactive substance (TBARs) was increased while the activities of superoxide dismutase and concentration of glutathione and ascorboic acid was decreased significantly in testis of rats treated with ammonium molybdate. All the parameters showed significant improvement after 60 days of treatment withdrawal in highest dose group. Environmental stresses are caused by biotic factors such as fungi, bacteria and herbivore and abiotic factors such as temperature, water, light and salinity. Salinity stresses cause a decrease in the production of banana, because they affect the growth of the plant in the changing balance of Na and ClG ions and changes in the plant’s primary metabolites including fatty acid compounds. The purpose of the research was to identify the fatty acid compounds. The research method was conducted in 2 stages: phase (1) Developing in vitro culture of banana shoots with NaCl treatment of 0, 50, 100, 150 and 200 mM concentrations, (2) Gas Chromatography Mass Spectrometry (GCMS) analysis on the roots of in vitro banana planlets. The results showed that the fatty acid compounds involved in the protection against salt stress. The fatty acid the highest area compound in 0 mM treatment 9 Octadecanoic acid methyl ester were (14:36%), 50 mM treatment 9-octadecanoic acid methyl ester 13.13% and 100 mM treatment Tridecanoic acid 11.8%. The effect of ethanol extract on malaria infected mice was studied by assaying percentage parasitaemia, some liver marker enzymes and kidney function markers. The acute toxicity study showed that the ethanol extract had LD50 above 2900 mg kgG 1 b.wt. Malaria infected mice treated with the ethanol extract had significant (p<0.05) decrease in the mean percentage parasitaemia when compared with group 2 mice (malaria untreated). The AST activity showed significant (p<0.05) increase in all the test groups when compared to normal control on day 14 but significantly (p<0.05) reduced in all the treated groups when compared to group 2 mice on day 28. The ALT and ALP activities decreased significantly (p<0.05) in all the test groups when compared to group 2 mice. The urea concentration significantly (p<0.05) decreased in the normal control, group 4 and 5 when compared with group 2 mice. Creatinine concentration decreased significantly (p<0.05) in the normal control, group 3 and group 4 when compared to group 2 mice on day 14, however, on day 28, no significant (p>0.05) differences were observed in the creatinine concentrations in all the groups when compared to normal control. Worldwide, there were about 2.5 million new cases of HIV reported in 2011. The HIV continues to be a major global public health issue with approximately 34 million people living with HIV in 2011. However, the development of anti-viral has blunted the AIDS epidemic in the western world but globally the epidemic has not been curtailed. Negative Regulatory Factor (Nef) is one of the accessory genes found only within HIV and SIV genomes and thought to play a key role in the progression to AIDS. Nef is considered as a potential anti viral target for preventing or at least delaying HIV pathogenesis. It suggesting that, the Nef protein that was detected not identically synthesis through post translation modification though it was expressed in the cytoplasm of HEK 293 cells. Five groups (n = 6) of rats, A-E were respectively exposed to 6.0, 12.0 and 24.0 mg kgG b.wt. of esculetin, 0.2 mL of the vehicle control (10% dimethylsulfoxide (DMSO)) and 0.2 mL of the normal control (Distilled Water (DW)) per orally and daily for 28 days. Compared with the controls, the haematological parameters change in the esculetin-exposed rats was not significant (p>0.05) but decreased numerically with increasing dose viz: hemoglobin (99.83±5.04 g LG-91±7.04 g LG), packed cell volume (30±1.41%-27.33±2.16%), white blood cell count (9.63±0.91×10 LG7.95±0.58×10 LG), neutrophil (1.93±0.51×10 LG-1.72±0.34×10 LG) and lymphocyte count (7.64±1.04×10 LG-6.18±0.63×10 LG). The reduction in the serum cholesterol (3.39±0.23 mmol LG3.01±0.78 mmol LG) and triacylglyceride (0.49±0.45 mmol LG-0.15±0.05 mmol LG) concentrations in the esculetin-dosed rats was significant (p<0.05) and followed the numerical decreasing trend. The serum MDA concentration (1.90±0.53 mmol LG-2.82±1.50 mmol LG) decreased (p<0.05) but increased numerically with increasing dose. Many basic and specialized cellular functions depend on the activity of this enzyme, hence, its adaptability to changing cellular and physiological stimuli. This review presents an overview of the many adaptations or mechanisms that regulate sodium pump activity in a tissue-specific manner. These mechanisms include regulations by substrates, membrane associated components such as cytoskeletal elements and the γ-subunits and circulating endogenous inhibitors as well as a variety of hormones, including corticosteroids, peptide-hormones and catecholamine.
