However, NG has been criticized as a non-standard (several grading schemes used) and subjective (significant interobserver variability) method for predicting the biologic behavior of this tumor. Our results on 106 invasive ductal breast carcinomas demonstrate that NG correlates with morphometric prognostic index (MPI) (p < 0.007) (lower value of MPI is connected with lower NG of 1-2 and better prognosis), with estrogen receptor (p < 0.0002) and progesteron receptor status (p < 0.04) (hormonal receptor positive tumors having lower NG). NG correlates with s-phase fraction (SPF), p < 0.04, values lower than 9.6% corresponding to lower NG. The lesion consisted of interlacing bundles of spindle cells arranged around small vessels with hemangiopericytomatous appearance. Neoplastic cells were positive for vimentin and actin. Desmin, cytokeratins and Factor XIIIa were negative. Two different modes to detect DNA-aneuploidy were compared: the conventional application of a threshold at 2.2c for the modal DNA value and the use of the Kolmogoroff-Smirnow (KS) test to compare GO/1 phase fractions of the cells under analysis and the internal reference cells. Whereas the specificity for both types of analysis was 100%, the sentitivity of the latter method was 73.3% compared to 48.3% of the conventional procedure. Therefore, the application of the KS-test for the identification of DNA- stemlines as aneuploid results in an increased detection rate was compared with the conventional threshold procedure. DNA histograms were obtained in 186 cases, whereas in 44 samples the cell number was too low for detailed estimation of cell kinetics. Aneuploidy was observed in 71 of 90 samples where cancer cells were detected by cytology, in four of 20 samples where breast cancer could only be suspected and in 12 of those 76 where no cancer cells occurred in cytologic specimens. The average S-phase and S+G2M values were higher in aneuploid than in diploid cell samples. In the group of 40 cases (34 breast cancers, 6 benign lesions) comparative analysis of DNA histograms obtained on aspirates and tissue fragments revealed a concordance in 88.9%. Modified radical mastectomy was performed in 41 breast cancers and in 33 cases ductal infiltrating carcinomas were recognized by histology and scored according to Bloom-Richardson score. Cell features assessed by cytology in these cases did not correlate with histology. In most cases, however, nuclear polymorphism and numerous naked nuclei were present in cytologic specimens. Frequently, malignancies of the jawbones appear clinically as inflammatory diseases. Recognition of these cases usually occurs after the inflammatory-like symptoms have failed to respond to conventional therapy. This case report presents an extranodal non-Hodgkin lymphoma of the maxilla, which clinically imposed as odontogenic infection in a 45-year-old woman. The definite diagnosis was made by histologic means and confirmed by polymerase chain reaction (PCR). In this study, immunohistochemistry on 15 different human tissue probes of gastrointestinal and colonic mucosa was carried out to visualize possible deposits for TGF-alpha in the mucosa and to compare different compartments of the upper and lower gastrointestinal epithelium. Most interestingly, in the colon there is a strong TGF-alpha-like immunoreactivity in neuroendocrine cells as evidenced by double labeling experiments with common neuroendocrine markers and by immunoelectron microscopy. In the upper gastrointestinal epithelium, however, all neuroendocrine cell types are negative for TGF-alpha, but parietal cells of the gastric corpus epithelium show strong cytoplasmic immunostaining for TGF-alpha. The neuroendocrine cells of the colon-harboring TGF-alpha-like immunoreactivity in their neurosecretory granules belong morphologically to the so-called L-cell type. Along with the well-known presence of EGF-receptor in the mucosa of the gastrointestinum, TGF-alpha, which is very likely to be released by this neuroendocrine cell type of the colon, acts in a paracrine mode. Histomorphology may poorly correlate with clinical outcome and the tumors within the same type in classification schedule exhibit different clinical courses. Prognostic relevance of various cell proliferation markers has been investigated in many types of human cancer, recently including salivary gland tumors. Evaluation of DNA content by flow cytometry and by cytophotometry, AgNOR technique, and immunohistochemical detection of antigens in cycling cells such as the Ki67 antigen and proliferating cell nuclear antigen (PCNA) have been applied to a variety of benign and malignant salivary gland tumors in only few studies so far. Cell proliferation, assessed with the MIB1 antibody, that recognizes the Ki67 antigen in proliferating cells, represents a significant prognostic factor for acinic cell carcinomas and mucoepidermoid carcinomas of salivary gland origin. Moreover, much lower proliferative activity as assessed with the MIB1 antibody helps to distinguish difficult cases of polymorphous low grade adenocarcinomas from adenoid cystic carcinomas and may contribute to differentiation of solid myoepithelial cell-rich pleomorphic adenomas from various malignant tumors. Using predominantly formalin-fixed and paraffin-embedded material, we performed polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) analysis of exons 4-8 and direct sequencing. Molecular genetic findings were correlated with immunohistochemical detection of p53 protein. A total of eight alterations (19%) were identified. Two splice mutations were detected in one case of a highly malignant osteosarcoma and its metastasis, and in one osteoblastoma with focal malignant transformation. Four of the mutations were missense mutations, one was of the silent type. These data correspond to the results found in the literature on bone and soft tissue tumors. The point-counting method was utilized to study the volume densities of following tissue compartments: red pulp, perifollicular zone, white pulp (this compartment was divided into two subcompartments: follicles and periarteriolar lymphatic sheath), marginal zone and connective tissue. The following stereologic parameters of lymphoid follicles were determined: areal numerical density (the number of follicles per mm2 of tissue section), the numerical density (number of follicles per mm3 of tissue), and the mean follicle diameter. The identity of tissue compartments was verified using immunohistochemical staining for B- and T- lymphocytes. Significantly increased volume densities of perifollicular zone and periarteriolar lymphatic sheath were registered in patients treated for traumatic spleen injury. On the other hand, the significantly increased number of lymphoid follicles per mm2 and per mm3 of splenic tissue were registered in the group of cancer patients.
