Three weeks after implantation [<sup>3</sup>H] cholesterol linolenate and arachidonate were resorbed to a significantly greater extent than free [<sup>3</sup>H]cholesterol, [<sup>3</sup>H] cholesterol linolenate and the more saturated labelled sterol esters. [<sup>3</sup>H]Cholesterol was more rapidly mobilized than the saturated and monounsaturated [<sup>3</sup>H] cholesterol esters. The resorption of esterified cholesterol did not appear to depend on rapid hydrolysis of the fatty acid moiety and slow removal of the sterol nucleus, for <sup>3</sup>H and <sup>14</sup>C labels disappeared at nearly equal rates from implants of [<sup>3</sup>H]cholesterol-[l-<sup>14</sup>C]linolenate. Addition of antioxidants to the polyunsaturated ester implants did not significantly accelerate resorption, even though it did reduce chromolipid formation. One group was supplied with distilled (ion-exchanged) water, the other with normal drinking water. The arteriosclerotic lesions were studied using histologic and planimetric methods. Serum lipids were measured. The protein content was 8 % casein. Carbohydrate (sucrose) levels of 45.5 %, 57.5 % and 79.5 % were used to balance the range in amount of fat. Within the 40 % fat group, 65 % of the mice developed atrial thrombosis as compared to a 36 % and 30 % incidence in the 28 % and 6 % fat level groups. The highest incidence of atrial lesions occurred during 10–12 weeks in the 6 % fat group, and during 18–22 weeks in mice fed the 28 % and 40 % fat levels. After feeding the diets for as long as 20 weeks the incidence of atrial thrombosis decreased rapidly in the 6 % and 28 % fat level groups. Deux d’entre elles proviennent de l’orifice aortique,une autre était insérée en position mitrale et la quatrième dans l’orifice tricuspide. Ces deux dernières proviennent du měme patient. Les lipides des valves ont ététudiés par Chromatographie en couche mince de silica-gel après extraction par le mélange de <span>Folch</span><em>et al.</em> Les 4 valves avaient un aspect jaunâatre, très différent de celui des valves neuves. L’une d’elle était rétractée. Dans toutes les prothèses, on a trouvé tous les lipides du plasma normal: esters du cholestérol, triglycérides, acides gras, cholestérol, phospholipides et de plus, on y a découvert la présence de monoglycérides et de diglycérides. En outre, les valves tricuspide et mitrale renferment une substance que l’on retrouve également dans le plasma laiteux. La prothèse tricuspide est de loin la plus riche en lipides, notamment en esters du cholestérol et en cholestérol, mais ces divers résultas devront ětre confirmés par une étude statistique.</p><p>Ces résultats permettent de dire que les lipides présents dans toute l’épaisseur des valves proviennent de la pénétration profonde des lipoprotéines du sang dans le matériel apparemment inerte de la prothèse. En outre, le rôle probable de l’état du sang circulant et de la pression sanguine dans l’apparition des altérations des valves de Starr rappelle à bien des égards, ce qu’on sait des conditions circulatoires qui provoquent l’athérosclérose. A ce titre, l’étude du devenir des billes en matière plastique placées dans les orifices cardiaques fournit un modèle de recherche particulièrement intéressant pour l’étude de cette maladie.</p></div><div><p>In an effort to contribute to the study of serum factors and haemodynamics in atherosclerosis, we analysed lipids present in the plastic balls of four starr valves which had been in position for 34, 28, 4 and 2.5 months, respectively. Two balls originated from an aortic valve prosthesis; the third came from a mitral and the fourth from a tricuspid valve prosthesis. The last two came from the same patient. The lipids were studied by thin-layer (silica-gel) chromatography after extraction with the aid of the mixture of <span>Folch</span><em>et al.</em> The four valves showed a yellowish appearance quite different from that of new valves. One was retracted. In all prostheses we found all the normal plasma lipids—cholesterol esters, triglycerides, fatty acids, cholesterol and phospholipids—and in addition we discovered monoglycerides and diglycerides. The tricuspid and mitral prostheses contained a substance which is also found in the milky plasma. The tricuspid prosthesis had by far the largest content of lipids, particularly cholesterol ester and cholesterol. The level of lipoprotein lipase in the plasma fell over the first 3-7 days, and then rose to a level higher than that present on the first day of the heparin infusion. This secondary peak was followed by a further fall in plasma activity.</p><p>The fluctuations in plasma lipoprotein lipase activity may have been due to changes in the sensitivity of the patients to heparin. Whether this was the result of acute myocardial infarction is not clear, as no control studies have been performed. Evidence is presented for the presence of chondroitin sulphate A in approximately equal amount to chondroitin sulphate B. Saxe","doi":"10.1016/S0368-1319(68)80004-3","DOIUrl":"10.1016/S0368-1319(68)80004-3","url":null,"abstract":"<div><p>By using modifications of the Dole free fatty acid (FFA) extraction procedure, the present study demonstrated that ethyl-<em>p</em>-chlorophenoxyisobutyrate (Clofibrate or Atromid-S) given orally and intraperitoneally to rats at doses of 50, 100 and 800 mg/kg/day for 9–30 days produced 40–80 % decreases in the levels of plasma and red blood cell FFA. Near maximum FFA-lowering effects were obtained with the lower doses. These observations support the concept that some of the clofibrate-induced reductions in the circulating levels of cholesterol, phospholipids, and triglycerides are due to a reduction in FFA availability. Besides decreasing the circulating levels of FFA, Clofibrate treatment resulted in certain alterations in the relative composition of the plasma FFA.
