Desmosomes in the endometrium of rats treated at 40 mg/kg/day were similar to those of controls. Abnormalities included an increase in desmosome number with prominent filamentous interconnections, asymmetry, disorganization of tonofilaments, missing tonofilaments, missing desmosomal plaques, and desmosomal plaques that did not appear to line up with those of the adjacent cell. It is thought that the antifertility actions of gossypol may be due to a disturbance of desmosome formation. This would cause a disruption of endometrial cell adhesion as well as an alteration in the microenvironment within cytoplasmic domains. The S-layer has tetragonal symmetry, with a unit cell dimension of 12.0 nm and a thickness of 5.6 nm. Infected RS 537 rabbit fibroblasts were treated with tunicamycin, which inhibits the glycosylation of many glycoproteins, or monensin, which inhibits the transport of proteins to the cell surface, and were compared with untreated infected cells. Tunicamycin treatment almost entirely suppresses the perinuclear envelopment of viral capsids, induces the nuclear export of unusually numerous naked viral capsids, and prevents the proliferation of the Golgi apparatus. On the other hand, perinuclear envelopment of viral capsids still occurs following a monensin treatment; however, enveloped viral capsids are not released into the extracellular space; in addition this treatment induces the proliferation of the rough endoplasmic reticulum (RER). Our surveys have shown that, in addition to the expected storage function, these hypogeal cotyledons also develop a definite photosynthetic function supported by well-organized chloroplasts. The initial presence of chloroplasts with rather large thylakoid stacks in the quiescent cotyledons of two species does not appear to represent an advantage fro them over the two other species, where the white quiescent cotyledons have plastids without thylakoid stacks. The following criteria were used to determine that intact whole disks were isolated: (1) the disk diameter, 1.4 μm; (2) the presence of one incision extending toward the center of the disk; (3) the edge structure; and (4) the presence of particulate material with the dimensions of the particles corresponding to those present in the particulate fracture face of freeze-fractured outer segment disks. The disks were analyzed after negative staining or after freeze-drying and surface replication. The surface of the disks appeared smooth. In damaged disks negative staining revealed the presence of particulate material located below the disk surface. The lipid intracuticle, proposed as the main desiccation barrier in tuns, is thickest in species most tolerant of rapid drying, supporting a multilayer lipid model for the variable cuticle permeability (J. C. Wright, <em>Tissue Cell</em>, in press). The cuticle shows the greatest variation in thickness (chiefly attributable to inter-/intrasegmental differences) in species effecting the greatest surface area reduction during tun formation, suggesting that thinner intersegmental cuticle allows for greater infolding. This is substantiated by the demonstration that the greatest variation in thickness—for all species—occurs in the presumably stiff, chitinous procuticle, with intermediate variation in the partly tanned epicuticle, and minimum variation in the untanned, probably highly flexible, intracuticle. The cyst wall develops from randomly dispersed particulate material which, during midspermiogenesis, tends to collect along the irregularly outlined cell membrane of the spermatid. Further accumulation of such a material results in the sponginess of the cyst wall. In late spermatid, the intracellularly produced wall material becomes thick, compact, and electron dense. Later, the nongerminal testicular cells produce extracellular material which is deposited on the outside of the sperm cell membrame. Application of Thiéry's method confirms the presence of carbohydrates in the cyst wall. Finally the cyst wall assumes such a consistency that it becomes impermeable to the fixing fluids. In addition to the cyst wall, the sperm tail in <em>Diplotemnus</em> sp. presents some unusual morphological features; the axoneme is enclosed in a “flagellar tunnel” and the axoneme plus the flagellar tunnel, in turn, is enclosed in a unique structure—the “flagellar sheath”. The flagellar tunnel is formed by the close apposition of the outer leaflets of the invaginated cell membranes. This tunnel along with the axoneme retracts into the spermatid. By such a disposition the tunnel separates the motile apparatus from the mitochondria. The species differ with respect to the number of spiral coils. This unique axonemal structure is a characteristic of Asphondyliids and has not been found in some related groups. The axonemal microtubular doublets have outer arms only as is the rule in the spermatozoa of other gall-midge flies. This peculiar kind of axoneme is able to move even in the absence of inner dynein arms.
