{"title":"Alterations in the sensitivity to endogenous proteolysis due to storage of fructose-1,6-bisphosphatase extracts of liver and muscle of young and old rats.","authors":"S K Trigun, S N Singh","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The sensitivity of fructose-1,6-bisphosphatase (FBPase: EC 3.1.3.11) to endogenous proteolysis was studied in liver and muscle extracts of young (28 weeks) and old (97 weeks) rats (stored at 4 degrees +/- 2 degrees C) by monitoring the alterations in its properties. Apart from the level of total enzyme activity, decrease in activity ratio (at pH 7.0/9.2) and sensitivity to AMP inhibition no age-dependent significant change was found during storage of the liver extract. However, a more significant increase in activity, comparatively less decrease in activity ratio, and retention of sensitivity to AMP inhibition were observed during the storage of muscle enzyme extract from old rats than from young rats. Using gel electrophoresis, an extra degradative active product was observed in all samples except those of old rat muscle. These results may indicate tissue-specific alterations in the susceptibility of FBPase to endogenous proteolysis during old age in the rat.</p>","PeriodicalId":77698,"journal":{"name":"Comprehensive gerontology. It allows in vivo retrieval of 3 x 3 mm2 mouse epidermis biopsies. Separation takes place exactly at the epidermal-dermal junction. Young skin and iso-grafted skin require suction with a lower bar value than skin from young and old non-grafted animals. Langerhans cells in biopsies were identified both with ATPase staining and with monoclonal antibody. {"title":"Sulfate incorporation into glycosaminoglycans of human lung fibroblasts as influenced by the number of subcultures and glycosaminoglycan polysulfate.","authors":"K Sames, B Lücht","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The influence of a glycosaminoglycan polysulfate (GAGPS) on the glycosaminoglycan (GAG) metabolism was investigated in relation to in vitro ageing of cultured human lung fibroblasts (Flow 2002). The incorporation of 35S-sulfate was determined following proteolytic digestion and specific degradation methods for the individual GAGs. In untreated cultures the pericellular matrix and medium showed a decrease of chondroitin sulfate (CS) 35S-radioactivity of the higher passage numbers, while the pericellular heparan sulfate (HS) showed increasing values. In GAGPS-treated cultures the decrease of 35S-sulfate incorporation into CS and dermatan sulfate of the cells and medium as well as into the pericellular CS was negatively correlated to the passage numbers, since GAGPS preferentially increased the radioactivity in cultures of low passage numbers. The HS values, however, were changed in the same direction as observed in untreated controls. Thus, age-related changes in the GAG metabolism become visible when exogenous GAGPS is used as a stimulus.</p>","PeriodicalId":77698,"journal":{"name":"Comprehensive gerontology. This effect of grafting was independent of whether young or old skin was transferred to young or old recipients. {"title":"Plasma selenium and glutathione peroxidase in relation to cancer, angina pectoris and short-term mortality in 68-year-old men.","authors":"B Akesson, B Steen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Plasma selenium and plasma glutathione peroxidase were measured in 483 68-year-old men participating in a longitudinal population study, in which the probands were representatively selected. The prevalence of cancer and coronary heart disease was assessed by a structured interview. In plasma the concentration of selenium was 1.08 +/- 0.18 mumol X l-1 and the activity of glutathione peroxidase 540 +/- 90 mumol X l-1 X min-1. Plasma selenium was correlated to glutathione peroxidase activity (r = 0.49, p less than 0.001), but not significantly to plasma albumin levels (r = 0.18). Plasma selenium and glutathione peroxidase values in subjects with cancer or angina pectoris were not significantly different from those in subjects without these diseases. Plasma selenium concentration and glutathione peroxidase activity in the subjects who died within 1 year of screening were 89% and 88%, respectively, of the values among survivors (p less than 0.01). For the subjects who died during the second year after screening, these variables did not differ significantly from the values among survivors, 97% and 99%, respectively. Since the relation of low selenium and glutathione peroxidase to mortality was observed only for the first year, it was probably due to a disturbed nutritional state prior to death, rather than to a long-term negative influence of low plasma selenium on the mortality risk. Information regarding hearing problems was collected by questionnaire, and hearing was tested with pure-tone audiometry after wax extraction. Only 55 patients were able to participate in the hearing test due to the high prevalence of dementia. For this category of patients it is probably necessary to use objective neurophysiological tests, such as brain stem audiometry and electrocochleography. All patients tested had considerably impaired hearing. Mean hearing loss in the speech area was 51 dB, indicating that a majority of the patients needed hearing aids. However, only one patient in 10 had such. Technical devices for amplification in the wards were scarce. It was evident that impaired hearing among geriatric long-stay patients was an underestimated and rather neglected problem, and a need for education among the nursing staff regarding communication problems was noted.</p>","PeriodicalId":77698,"journal":{"name":"Comprehensive gerontology. {"title":"Screening of patients admitted to a geriatric hospital with supposed organic dementia.","authors":"K Hedner, L Gustafson, G Steen, B Steen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>75 patients, aged 69 to 97 (mean 84) years, admitted to a geriatric clinic with symptoms or signs of organic brain failure, were examined with a wide test battery including chemical analyses, electroencephalogram (EEG), regional cerebral blood flow (rCBF) measurement, and psychometric tests. There was a prevalence of 89% organic dementia, 3% treatable dementia, and 8% non-dementia conditions. Thus the prevalence of treatable conditions was rather low (11%). Multi-infarct dementia was more prevalent (52%) than dementia of Alzheimer type (31%). All but one of the non-dementia conditions were due to confusional reaction. In no case was depression, drug intoxication, or deafness the only cause of symptoms. After a follow-up period of 6 months, 33% of the patients had died. {"title":"Craniomandibular disorders in elderly with psoriasis.","authors":"M Könönen, P Ekholm, E Mäkilä","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>A questionnaire survey was carried out to determine the prevalence of craniomandibular (CM) disorders in relation to age in psoriasis. Of the 1776 psoriatics, 85.4% responded; 1050 of them had psoriasis with skin symptoms only (P), and 400 had both skin and joint symptoms (PA). No statistically significant differences could be found between older (greater than or equal to 65 yrs) and younger (less than 65 yrs) psoriatics in either the P-group or the PA-group as regards the severity of subjective symptoms of CM disorders. In the P-group the younger psoriatics reported more frequent (30%) subjective symptoms than the older ones did (19%) (p less than 0.05), but no difference was found in the PA-group (51% and 41%, respectively) (NS). The most frequently reported symptoms in all age groups were temporomandibular joint (TMJ) sounds and TMJ pain during function. TMJ sounds were significantly less frequently reported by the older than the younger psoriatics both in the P-group and the PA-group (p less than 0.01 and p less than 0.05, respectively). It can be concluded that, apart from TMJ sounds, the frequency and severity of symptoms of CM disorders are not related to age in psoriasis.</p>","PeriodicalId":77698,"journal":{"name":"Comprehensive gerontology.
