This provides powerful approach in designing novel treatment strategies. RNA interference (RNAi) is a natural phenomenon that can aid in the study of post-transcriptional regulation of genes. This mechanism can be stimulated via introduction of double stranded RNA (dsRNA) in a cell. Synthetic short/small interfering RNAs (siRNA) can be utilized to trigger down-regulation of desired genes via transfecting into mammalian cells. Recently, utilization of bio-based polymers has been an attractive approach in drug delivery for medical applications. This review article describes the advantages of exploiting bio-based polymeric carriers for siRNA targeting, as bio-therapeutics. Here, we report the current developments, safety and delivery of bio-based siRNAs via polymeric carriers. Sporulasyon evresinde, sporlarla birlikte bircok bocek cesidine karsi insektisit etki gosteren kristal proteinleri de uretir. Insektisidal kristal proteinler Cry ve Cyt proteinler olup; aktivitelerini duyarli boceklerin orta barsak membranlarinda bulunan reseptore baglanarak gosterirler. Bu reseptor baglantili ozgul aktivitelerinden dolayi zirai mucadelede onemlidirler. Daha sonralari, insektisit olanlara gore dogada daha genis dagilim gosteren insektisit olmayan kristal proteinler uzerindeki calismalar yeni bir protein sinifi olan parasporinleri ortaya cikarmistir. Parasporinler ayricalikli olarak kanser hucrelerine karsi sitotoksik etki gosterirken normal hucrelere etki etmez ve hemolitik etki gostermez. Şimdiye kadar alti parasporin ailesi tanimlanmis olup toplamda 19 alt grup bilinmektedir. Parasporinler de insektisidal kristal proteinlere benzer sekilde reseptor baglantili etki gosterdiginden kanser tedavisiyle ilgili calismalarda yeni bir arastirma alaninin ortaya cikmasini saglamistir. Ozellikleri belirlenen orneklerden fenollu besiyerine inokulasyon yapilmis ve 19 izolat elde edilmistir. Bu izole edilen 19 izolattan morfolojik olarak farkli oldugu dusunulen 9 izolatin tanimlamasi yapilmistir. Izolatlar arasinda yogun olarak Pseudomonas turleri belirlenmistir. Tanimlanan bu 9 izolatin atik sulardaki aritim potansiyellerini degerlendirebilmek icin antibiyotik duyarlilik testi gerceklestirilmistir. Ayrica izolatlarin amilaz ve proteaz enzim uretim aktiviteleri degerlendirilmistir. Izolatlarin ofloksasin, azitromisin ve streptomisine duyarli olduklari belirlenmistir. Intein kullanimi ile protein ayrismasi uzerinde yapilan arastirmalar, intein ve protein saflastirma sureclerinin daha verimli hale getirilmesini amaclamaktadir. In vivo ve in vitro olarak gerceklesen sureclerde, intein ve hedef protein iceren poliklonal PCR urununun cogaltilmasi ve ekspres plazmidlerin klonlanan suslara tasinmasi islemleri yapilmaktadir. Inteinin proteinden ayrilma aktivitesi ve ayrilma orani sabiti kinetikleri, degisik pH, sicaklik ve suzme surelerinde belirlenmekte ve hedef proteinlere iliskili olarak karsilastirilmalari yapilmaktadir. Arastirmalarda degisik mikrobiyal dogal ve yapay inteinlerin protein ayristirma aktiviteleri incelenmis ve yapay Nostoc punctiforme DnaE C-inteinlerinin ayrisma aktivitelerinin en yuksek oldugu belirlenmistir. Bununla birlikte, sucul sistemlerin Tl’a maruz kalmasinin sonuclari ile ilgili cok az bilgi mevcuttur. Bu calismanin amaci, sucul oligoket Tubifex tubifex ’in antioksidan biyolojik belirteclerinde ( metallotionein (MT), toplam glutatyon (GSH), glutatyon peroksidaz (GPx), glutatyon S-transferaz (GST) ve katalaz (CAT)), TI’un subletal dozlarina yanit olarak meydana gelen degisikliklerin arastirilmasidir. Deney gruplari 0.25, 0.5 ve 1 µg l -1’ lik konsantrasyonlarda, 7 ve 15 gunluk sureler boyunca talyum asetat’a maruz birakilmistir. MT seviyelerinde ilgi cekici bir azalma gozlenmistir. 0.25 µg l -1’ lik konsantrasyon toplam GSH, GPx ve GST aktiviteleri uzerinde etki gostermezken, 0.5 ve 1 µg l -1’ lik konsantrasyonlar toplam GSH ve GPx aktivitelerini 7 gunun sonunda induklemistir. Daha uzun maruz kalma suresi GPx ve GST aktiviteleri ile birlikte toplam GSH’un da tum konsantrasyonlarda azalmasina neden olmustur. CAT aktivitesinde tum deney gruplarinda gozlenen anlamli artisa ragmen, lipid peroksidasyonunun (LP) artan konsantrasyon ve sureye bagli olarak arttigi gozlenmistir. Metal bilesik sisplatin cesitli kanser turlerine karsi tibbi olarak kullanilmis ve yan etkiler gostermistir. Biz bu calismada sisplatine alternatif ajan olarak dusundugumuz titanyum dioksitin A549 hucreleri uzerindeki etkilerini ve apoptotik mekanizmasini arastirdik. Titanyum dioksitin 24, 48 ve 72 saatlik surelerdeki zamana ve konsantrasyon araligina bagli olarak antiproliferatif etkilerini MTT canlilik analizini kullanarak belirledik. Titanyum dioksitin apoptozisi tetikledigini saptadik. Titanyum dioksitin IC 30 konsantrasyonu, 72 saat suresince A549 hucreleri uzerinde uygulandiginda erken ve gec apoptozisi tetikledigi belirlendi. Titanyum dioksitin IC 30 konsantrasyonu, 72 saat suresince A549 hucreleri uzerine uygulandiginda mitokondriyal membran potansiyelini azaltarak apoptozisi tetikledi. Buna ragmen kaspaz-3 aktivitesi gozlenmedi. Titanyum dioksit kaspazdan bagimsiz bir yol izleyerek apoptozisi tetiklemistir diyebiliriz. Titanyum dioksitin IC 30 konsantrasyonun 72 saatlik sure sonundaki hematoksilen ve eozin, TUNEL, BrdU, Bcl-2 ve Bax immunositokimyasal analizleri sonucunda apoptotik indekslerinde artis belirlendi. Titanyum dioksitin IC 30 konsantrasyonun uygulandigi A549 hucrelerinde 72 saatlik sure sonunda konfokal ve TEM mikrokobisi kullanilarak cesitli apoptotik yapilar gozlendi. On dort istasyonun 10’unda Cryptosporidium ookistlerine LAMP yontemiyle rastlanilmistir. Cryptosporidium ookistleriyle kontamineli istasyonlar: Tersakan–Havza; Tersakan-Suluova, Celtek; Tersakan-Kanlidere; Tersakan-Bogazkoy; Tersakan-Karasu; Tersakan-son; Yesilirmak-Amasya Koprusu; Yesilirmak-Yassical; Yesilirmak-Durucasu; Yesilirmak-Tasova’dir. Arastirilan tum istasyonlarin icinde Tersakan–Suluova, Celtek ve Tersakan-Bogazkoy istasyonlari Cryptosporidium ookistleri acisindan en kirli istasyonlar olarak belirlenmistir. Bununla beraber Amasya ve Samsun’dan gecen Yesilirmak Nehri’nin Tersakan Cayi’na oranla daha temiz oldugu saptanmistir .
