OTA insan ve hayvanlarda kanserojen, mutajenik ve teratojenik etkiler gostermektedir. Gerceklestirilen bu calismada yerel marketlerden temin edilen 30 adet kahve ornegi icerisindeki OTA miktari Yuksek Basincli Sivi Kromatografisi (YPSK) cihazi ile tayin edilmistir. Calismalar floresans dedektor kullanilarak Ex 330 Em 450 nm dalga boylarinda gerceklestirilmis, kolon sicakligi 25°C, sistem basinci 83 bar olarak olculmustur. Akis hizi 0.9 ml/dk olarak secilmistir. Kahveleri cozmek icin etanol kullanilmistir. Analiz boyunca C18 (200 mm 4.6 mm I.D., 5 µm partikul boyutuna sahip kolon kullanilmistir. OTA standardi ve numunelerin yaklasik 11.287 dk da sinyal verdigi gorulmektedir. Mobil faz olarak (su:asetonitril:asetik asit) (49.5:49.5:1) (h:h:h) kullanilmistir. {"title":"Determination of peptidase production potential of Bacillus strains isolated from ropey bread and optimisation of some culture conditions for peptidase production","authors":"M. Then, optimum enzyme production medium composition and conditions for two of the strains which were found to have the highest peptidase activity were optimised with one factor at a time approach. For this purpose, carbon source, nitrogen source, carbon/nitrogen ratio, agitation rate and pre-culture medium which ensure the best peptidase activity were determined by considering the cost-effectiveness. The best carbon and nitrogen source was determined as glucose and yeast extract, respectively. Cyclophosphamide is a chemotherapeutic agent but has also toxic effect on testes. Testicular cells can be protected from toxic effects by antioxidant systems, including vitamin E. The aim of this study is to reveal what kind of an effect the vit-E exerts upon the experimentally induced CP toxicity on testicular tissue of rats. {"title":"ISOLATION, SCREENING, PARTIAL PURIFICATION AND CHARACTERIZATION OF PROTEASE FROM HALOPHILIC BACTERIA ISOLATED FROM INDONESIAN FERMENTED FOOD","authors":"S. Fitriani, K. During the study, 4 halophilic protease producers were isolated from tauco and terasi . Among these isolates, halophilic bacterial isolate TANN 4 was recorded as the best protease producer. Extracellular protease from isolate TANN 4 was partially purified using ammonium sulfate precipitation. The protease was partially purified with final yield of 72.87 % and 25.41 fold purity. This moderate thermoactive and alkaliphilic protease showed a pH optimum of 8.0 and temperature optimum was 50 °C. The enzyme was also active at salt concentrations ranging from 1 to 15 % (w/v), with optimum activity at 1 % NaCl (w/v). Ethylenediaminetetraacetic acid (EDTA) completely inhibited the enzyme activity suggesting that it was a metalloprotease. Among metal ions, the Ca 2+ , K + and Mg 2+ ions enhanced the activity of enzyme. The K M and Vmax values exhibited by partially purified protease were 0.0649 mM and 216.45 U mg −1 using casein as substrate. The molecular weight was estimated to be 19.8 kDa on SDS PAGE. The enzyme also fairly stable in Triton X-100, SDS, 1 % commercial detergents (OMO and Ariel) and 25 % methanol. This enzyme was capable of hydrolyzing casein, hemoglobin and bovine serum albumin (BSA). Automated ribotyping analysis revealed that 3 isolate (TANN 4, TR 2 and TR 4) resembled Halobacillus trueperi that exhibited 71, 68 and 69 % similarity respectively, and isolate (TR 1) resembled Virgibacillus pantothenticus with 64 % similarity. Although hepatotoxic effects related to quetiapine treatment were reported in a few studies, potential hepatotoxicity of this drug was not identified clearly. Therefore, it was aimed to evaluate possible hepatotoxic effects of quetiapine by oral administration of this drug at 10 and 20 mg/kg doses to rats for 30 days in our study. For this purpose, plasma aspartate aminotransferase, alanine aminotransferase, total bilirubin and direct bilirubin levels as markers of hepatotoxicity were determined and histopathological examination was performed in liver tissues. According to our results, serum aspartate aminotransferase and alanine aminotransferase levels were significantly increased in quetiapine-administered groups, whereas total and direct bilirubin levels were significantly increased in high dose group. Histopathological examination of liver tissue indicated that necrotic areas were observed in 10 mg/kg quetiapine-administered group whereas necrotic areas were present and sinusoidal dilatation was observed in 20 mg/kg quetiapine-administered group.
