{"title":"Comparative biochemical and genetic studies of the GOX-1 (HAO-1) and the XDH-1 polymorphism in the rat (Rattus norvegicus).","authors":"B Brysch, I C Reetz, H J Hedrich","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Biochemical and genetic studies have been carried out to test whether the GOX-1 (HAO-1) and the XDH-1 polymorphism in the rat are determined by one locus or by two different closely linked loci. Besides similar migration behaviour upon electrophoresis the phenotypes of both polymorphisms share major biochemical properties such as responses to heat inactivation at 50 and 70 degrees C and isoelectric points. This suggests that they represent only one enzyme. The one-enzyme hypothesis is supported by linkage analyses performed in 50 offspring of a backcross population [(SPRD-Cu3 x BN)F1 x SPRD Cu3] and 14 LXB recombinant inbred strains (progenitor strains LEW and BN) in which not a single recombinational event can be observed. The latter finding agrees well with the GOX-1/XDH-1 distribution pattern investigated in 93 inbred strains and substrains (BENDER et al. 1984). {"title":"Herniation of the liver through the diaphragm in Mol: WIST rats.","authors":"E Kristiansen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The occurrence of herniation of liver through the diaphragm in Mol: WIST rats from toxicological studies conducted over a period of approximately 15 years at the Institute of Toxicology, National Food Agency, Denmark, is described. The incidence of diaphragmatic hernia in male rats indicates a tendency toward an increase through the years. The long term carcinogenicity studies initiated in 1971, 1976 and 1982 (age of the rats at necropsy approximately 2 1/2 year) revealed no diaphragmatic hernias in the 1971 study and 4% in both the 1976 and the 1982 study. In females, 0%, 1% and 1% respectively with diaphragmatic hernias were found. This method was used successfully in 190 animals, the complication rate being 3.2%. The hazards consisted of laryngospasm (1.6%) and fatal bleeding (0.5%). {"title":"[Leukocyte agglutination test for the determination of microbial burden].","authors":"N C Juhr, R Stuckenberg","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Transfer of laboratory animals from the breeder to the experimental unit includes in most cases a change of the microbial environment even under SPF-conditions. Many experimental treatments may also disturb the balance between host and microbial load and provoke infections. With the aim to detect such undesired effects in the course of the experiment the suitability of the leukergy test was investigated. SPF-rats, endotoxin-treated rats, and conventionalized germ-free rats were checked for leukocyte agglomeration, and the findings were compared with total leukocyte number, blood picture, endotoxin content in the blood and body temperature. {"title":"Seromonitoring in small laboratory animal colonies. A five year survey: 1984-1988.","authors":"V Kraft, B Meyer","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>From 1984 to 1988 one thousand serologic investigations of laboratory animal colonies originating from 10 different European countries were performed. The most prevalent infections in mouse stocks were caused by Mouse hepatitis virus (MHV), Minute virus of mice (MVM), Theiler's encephalomyelitis virus (TEMV), Reovirus type 3 (Reo3), Sendaivirus, and Pneumonia virus of mice (PVM). In mice no infections with Lymphocytic choriomeningitis virus (LCM), Polyomavirus, Mouse adenovirus, and K-virus were recorded. Only two colonies were infected by Ectromelia virus. The first six virus infections of mice were also found in rat colonies as well as the rat-specific Coronaviruses (Sialodacryoadenitisvirus--SDA, Rat corona virus--RCV) and Parvovirus (Kilham rat virus--KRV, Toolan H-1 virus) being endemic. Antibodies to Bacillus piliformis were detectable in about 50% of rat stocks screened. This is in contrast to the mouse, where only about 10% of the colonies were found to be positive. A similar picture was seen for M. pulmonis which is primarily an infection of the rat. In mice no case was detected during the last two years. The number of investigations performed from guineapig, hamster and rabbit colonies was relatively low. Nevertheless, antibodies against the following antigens were detectable: In guineapig stocks: Reo3, PVM, Sendai, Simian virus 5 (SV5) and B. piliformis; in rabbits: Reo3, Sendavirus, SV5, and B. piliformis; in hamsters: PVM, LCM and B. piliformis. The overall contamination rate showed a continuous decrease until 1988. Thereafter nearly no changes in lipid peroxidation occur. The relatively high rate of lipid peroxidation in fetal liver microsomes is of interest. The well known inducers of the monooxygenase system phenobarbital and beta-naphthoflavone change lipid peroxidation only in liver microsomes of newborns. Lipid peroxidation is connected with the formation of reactive oxygen species and organic radicals characterized by the chemiluminescence with the amplifiers luminol and lucigenin. Chemiluminescence increases with increasing age depending on the amplifier. {"title":"The relation of amyloidosis to social stress induced by crowding in the Syrian hamster (Mesocricetus auratus).","authors":"P G Germann, M Kohler, H Ernst, H Baumgart, U Mohr","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The aim of the presented study was the investigation of a probable influence of social stress on spontaneous amyloidosis. As stress-inducing parameter crowding of the animals was used. 220 Syrian hamsters were kept individually (controls) or with 3, 5, 7 animals per cage. The crowded animals showed a significant decrease in mean survival time. This was linked to a histopathological examined significant increase in the extent and incidence of amyloidosis in several organs of both male and female hamsters. The kidneys and adrenals were most affected. Chronic inflammation as one probable amyloidosis-inducing factor, was not related to the observed morphological alterations. Furthermore the increase of amyloidosis was statistically not connected with an age-dependent development of amyloidosis. After the natural end of the animals all pathological findings were recorded.
