The percentage of O. lienalis completing the molt was similar in the three culture media and two gas phases tested ranging from 44.8% (1:1 IMDM:NCTC + 5% CO2: 95% N2) to 56.7% (L-15 + 5% CO2: 95% air). Percent molting in O. volvulus ranged from 0% (F12(K) + 5% CO2: 95% N2) to 33.3% (L-15 + 5% CO2: 95% N2). All media were supplemented with either 20% FCS or 20% horse serum. Molting by O. lienalis occurred on days 2-5 in culture. Molting by O. volvulus was observed as early as day 5 and as late as day 10. Incomplete casting of the third-stage cuticle was frequently observed in O. volvulus. Larvae of both species entered a lethargus 24-48 hours prior to the onset of molting. Maximum survival in culture was 42 days for O. lienalis and 25 days for O. volvulus. To examine whether modifications in immune effector parameters correlate with the pattern of LCM growth, in vivo experiments were carried out in C57BL/6J mice infected intraperitoneally with LCM. Immigrant peritoneal and intra-LCM leukocytes were tested for the loss of cell surface receptors, membrane-bound and internalized immune complexes (ICs) and lysosomal enzyme contents during the restrictive and progressive growth phases of the LCM. The proportion of Fc and C3b receptor-bearing intra-LCM leukocytes decreased 2-fold and 31 to 72% of the leukocytes contained ICs during the progressive phase of the LCM. Acid phosphatase and beta-glucuronidase activities of macrophages declined sharply during the restrictive phase but they gradually increased as the infection progressed. DEC at 5 micrograms/ml promoted antibody-mediated neutrophil adherence and cytotoxicity with sheathed L. carinii microfilariae. With exsheathed parasites, the drug promoted significantly complement-and antibody-mediated cellular adherence and cytotoxicity. In the case of B. pahangi, antibody-dependent cell-mediated cytotoxicity (ADCC) of neutrophils to sheathed and exsheathed microfilariae were similarly stimulated by DEC. Pre-incubation of microfilariae with DEC promoted the cellular adherence and cytotoxicity whereas pre-incubation of cells had no such effect. DEC stimulated the adherence of eosinophils to both sheathed and exsheathed microfilariae of the two species but the adherence resulted in cytotoxic effect only on the L. carinii microfilariae. DEC at higher concentrations (100 or 500 micrograms/ml) inhibits these processes. From 2231 extirpated nodules 3327 male and 5713 female macrofilariae were isolated. About 98% of the male worms and 88% of the female worms were found alive. The sex ratio of the live male and female worms was 1:1.5 in Liberia and 1:1.6 in Upper Volta. Less than 1% of all live macrofilariae were immature in both countries. 22% of the live male worms in Liberia were regarded as old. The percentage of old male and female worms in Upper Volta and that of the old female worms in Liberia were similar (5-7%). The portion of old worms was independent of the age of the patients in Liberia. Dead worms were found in 66% of the Liberians and in 85% of the Voltaics. The percentage of patients with dead worms increased with the age. About 0.5% of all male and 8% of all female worms were calcified. In Liberia the percentage of calcified worms increased with the age of the patients. In both countries the highest number of live and dead worms were found on the pelvic girdle. On the thorax many more macrofilariae were found in Upper Volta than in Liberia. This may contribute to the severe eye lesions in this savanna area. The presented findings provide some basic information for the planning and performance of trials with macrofilaricidal drugs in hyperendemic areas of West Africa. Trypanosoma brucei procyclic forms were also agglutinated by midgut and hindgut extracts, but not by haemolymph. The 3 fractions tested failed to react with erythrocytes of four other vertebrates and other trypanosomatids (Leishmania hertigi and Crithidia fasciculata) examined. The reciprocal titre of agglutinating activity of the 3 fractions against calf, guinea pig and chicken erythrocytes were approximately the same at a dilution of 64. Both midgut and hindgut agglutinins showed higher titres against T. brucei at reciprocal dilutions of 128 and 256 respectively. Sialidase treatment, reduced the agglutinating activity to half its normal range while trypsinisation had no effect. The results of attempts to inhibit the agglutinating activity using a variety of sugars and glycoproteins revealed that midgut and hindgut were specific for D+glucosamine suggesting that midgut and hindgut agglutinin were possibly of the same origin. Phosvitin turned out to be the preferred exogenous acceptor protein, when homogenates of adults of Brugia malayi, Dipetalonema viteae, Setaria cervi and Litomosoides carinii were used as source of enzyme. In contrast to adults the L3-larval and the microfilarial stages demonstrated much lower phosvitin phosphorylating activity and the preference for acidic and basic proteins as phosphate acceptors was about equal. The results showed that the percentage of positive reactions (greater than 1:10) was higher in the sera of mothers than in their new borns (24.8 versus 12.4). In young children (6 months to 3 years old) the percentages of positive sera varied between 4.4 and 9.6. The values were much higher for persons between 15 to 70 years (28.3% to 43.7%). These flagellates were only agglutinated by the lectins from Canavalia ensiformis and Pisum sativum. Furthermore four day-old culture forms of the T. cruzi strains Morcego 1354, P-60, Téhuantépéc and WA 301/130 as well as the T. rangli strains San Augustin, Venezuela and DA-3412 were treated with neuraminidase and the supernatant examined by the Aminoff TBA assay. While the supernatant of the T. cruzi strains showed a red colour complex the supernatant of the T. rangeli strains was weak yellow-green. In contrast to the T. cruzi strains no N-acetylneuraminic acid was detectable on the cell surface of T. rangeli. During the first 6 to 8 weeks postinfection (p.i.) the larval cyst masses (LCM) grew slowly, weighted 1 g and contained 3 to 4 X 10(6) leukocytes/100 mg of LCM. Subsequently, (8 to 16 weeks p.i.) the growth of the LCM was rapid and was accompanied by 3 to 4-fold reduction in the absolute leukocyte count; monocytoid cells, neutrophils, eosinophils and T-lymphocyte significantly decreased during the progressive growth phase of the LCM. The cumulative data show a strong inverse relationship between the mean LCM weights and the absolute or relative counts of intra-LCM leukocytes.
