Lipid was observed in zona reticularis parenchymal cells but droplets were especially numerous in macrophages. Enlargement of lipid droplets was facilitated by an envelopment with lysosomal membranes. Focal areas or entire parenchymal cells may then undergo degeneration and necrosis and be incorporated into large macrophages recognizable by distinct areas of lipid-containing material as well as by small mitochondria with short cristae. Oestrogen may well act directly on zona reticularis cells inasmuch as nafoxidine, an antioestrogen, prevents these changes. Zona fasciculata cells were identical to those in controls and an intermediate region between the zona fasciculata and zona reticularis persists, although concentric whorls of rough endoplasmic reticulum seen in controls were replaced by disorganized, randomly dispersed cisternae of rough endoplasmic reticulum. The epithelium, the connective tissue cells, the endothelial cells and the pericytes of the capillaries are filled with lysosomal inclusions with pleomorphic structure. The nerve fibres show destructive process of the myelin sheaths and accumulation of lipid material in the Schwann cells. The extension of the lesions is similar, as shown by morphologic and morphometric analysis in early and terminal stages of the diseases. Restoration of relative resistance to metastasis formation in thymectomized sub-lethally irradiated mice by spleen cells.","authors":"M Suurküla,&nbsp;B Boeryd","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Adult thymectomy and/or sub-lethal irradiation increased spontaneous metastasis formation in several syngeneic murine systems (MCA-induced sarcomas MCG101 in C57BL/6J mice, MCG12 in CBA mice and MCA-induced epidermoid carcinoma EpCa1 in CBA mice). In order to differentiate between immunological and non-immunological effects of these treatments on tumour spread the effects of presumed selective restoration of immune competence by transfer of syngeneic spleen cells from normal donors shortly after irradiation (1.5 months before tumour transplantation) was perfomed. This procedure brought the resistance of thymectomized irradiated mice to tumour spread back to the level seen in untreated mice, providing further evidence for the conclusion that the indrease of tumour spread in this experimental model to a major part was immune-mediated. In thymectomized irradiated mice adoptive transfer of concomitant immunity by spleen cells from tumour-bearing mice soon after tumour transplantation completely inhibited the spread of the highly antigenic MCG101 and reduced that of the weakly antigenic MCG12 to control level. {"title":"Demecolcin and vinblastine sulphate as stathmokinetic agents for different tissues of the hairless mouse.","authors":"O Nome","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The dose/response relationship of the kinetostatic effect of demecolcin (ColcemidR) and vinblastine sulphate (VelbeR) was estimated in the epidermis, the forestomach, the jejunum and the colon of the hairless mouse (hr/hr) at 3 hours after intraperitoneal (i.p.) injection. A dose of 0.15 mg Colcemid per animal was found to be the lowest effective dose in all the organs tested. The value did not differ much from the corresponding effective dose of Velbe. This dose of Colcemid was then used to study the accumulation of arrested mitoses during the first 4 hours after i.p. injection. An almost linear increase in the mitotic count was found up to 4 hours in the epidermis, 3.5 hours in the forestomach and 3 hours in the jejunum and the colon. A dose of 0.15 mg Colcemid per animal i.p. with an observation time of 3 hours is thus a reliable method for estimation of the mitotic rate in many organs. Examination with the naked eye, dissecting microscope, light microscope and electron microscope of tissues from these animals sacrificed minutes, hours, weeks and up to 23 months after the injection revealed phogocytosis of most of the infected carbon by the professional phagocytes of the reticuloendothelial system (RES). However, in the non-RES sites carbon was trapped in significant amounts in the endothelial cells, the interendothelial spaces, pericytes and perivascular macrophages of the subepithelial capillaries and venules in the anterior nasal mucosa. The catchment area often showed partially fenestrated capillaries and redundant porous basement membrane around the venules. The leakage of carbon through these vessels was through these vessels was thought to be caused partly by the unusual porosity of the vascular walls and partly due to vascular stasis induced by constant loss of heat to the inspired air stream in post-natal life. Ultrastructural lesions, characterized by the hypertrophy of the smooth endoplasmic reticulum and the presence of lamellate cytosomes appeared in both tissues. They were less marked in hepatoma than in non-neoplastic cells. As in control livers, they were related to an invrease in pohspholipid concentration due to the decrease of phospholipid breakdown. In addition, chemical analysis demonstrated differences between hepatoma, non-neoplastic aflatoxin-treated liver and control liver: total and free cholesterol were decreased, relative concentration of sphingomyelin increased and that of phosphatidylethanolamine decreased by about 50%, in both hepatoma and non-neoplastic liver. Phospholipid concentration was decreased by 50% in tumour cells. A similar irradiation 1 or 10 days prior to i.v. DMBA showed a significant increase of tumours in the irradiated region, as compared with the contralateral abdominal region. The two thoracic regions, however, still had a very high tumour incidence. This was equal to, or even higher than that of the irradiated area. The same pattern also prevailed in animals receiving extra-mammary irradiation.
