Vaccine strains in contrast to virulent ones lacked glycolipid 8, possessed new glycolipid 8-a and a higher level of glycolipid 7. Low immunogenic strains had a decreased content of glycolipids, while avirulent non-immunogenic strain (15-a) nearly completely lost them. {"title":"Alternation of exo- and endotrophy during the mitotic cycle of the yeast cells.","authors":"V N Ivanov,&nbsp;A A Pindrus","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The utilization of the intracellular and extracellular sources of carbon and energy during the mitotic cycle of yeasts Saccharomyces cerevisiae, Kluyveromyces marxianus, Candida boidinii, Candida tropicalis has been studied. Increase in the consumption rate of carbon and energy sources and in the exogenous respiration rate at G1- and G2-phases of the mitotic cycle is shown. The rate of the endogenous respiration of the cells at these phases decreased. The hypothesis has been proposed that during the mitotic cycle of the yeast cell the regular alternation of exotrophy (the utilization of the extracellular carbon and energy sources by a cell) and endotrophy (the process of the utilization of the intracellular carbon and energy sources by a cell) occurs. It is possible to reveal the exotrophic cells by the cytological method which is based on the calculation of dead cells after incubation of the yeast suspension in amyl alcohol solution. This method has revealed that exotrophic and endotrophic processes do not predominate one over another but alternate at the mitotic cycle. Exotrophy and endotrophy are phase-specific processes. The scheme of isolation consists of purification stages on columns with phosphocellulose, DNA-cepharose CL-8B and phenylcepharose. DNAase was not bound to phosphocellulose, its volume was equal to zero. Then this DNAase was passed through column with DEA-cepharose CL-6B (elution by gradient KCl from 0.1 to 1.8M): enzyme was eluted at KCl concentration in the eluting buffer from 0.1 to 1.2 M. The enzyme was purified to the homogeneous state on column with phenylcepharose (elution by linear gradient of ethylene glycol from 30 to 80%): enzyme was eluted at the concentration of ethylene glycol in the eluting buffer from 43 to 80%. Among the cultures isolated from human and animal organism 60% of strains were characterized by the presence of extracellular chinases, among the collection strains--37%, among those isolated from soil and from insects--40 and 43%, respectively. The cultures of Bacillus subtilis as well as B. coagulans, B. megaterium and some other possessed the highest activity on the liquid medium. Whittenbury and Y. Heyer were screened for restriction endonucleases. Type II restriction endonucleases were detected in IMV B-3112 (= 12 b), IMV B-3027 (= 26), IMV B-3019 (= 9 c), IMV B-3017 (= 17 c), IMV B-3226 (= 26 v), IMV B-3033 (= Y), IMV B-3100 (= 100) and IMV B-3494 (= 1E494). The results obtained were indicative of relatively high frequency of restriction enzymes occurrence in methane-oxidizing bacteria. There were Kpn I (Asp 7181) restriction endonuclease isoschizomers in crude extracts of IMV B-3112, B-3017, B-3019, B-3027 isolated from fresh-water silt as well as in IMV B-3226 strain isolated from waste-water silt. Although these isolates had bee previously considered as untypical strains of M. ucrainicus, more detailed study of their properties allowed placing them with Methylovarius luteus (= Methylococcus luteus). IMV B-3494 strain was identified as Methylococcus capsulatus. Strain IMV B-3033 had earlier been allocated to Methylovarius whittenburyi (= Methylococcus whittenburyi). Specificity of restriction endonucleases of this strain was not tested. Therefore, for the first time restriction endonucleases were detected in methane-oxidizing bacteria. 8 strains (3 species) among 51 strains (13 species) were found to produce restriction endonucleases displaying three different types of specificity in the least.
