{"title":"[A clinical and histological study of the efficacy of betadine on gingival inflammation].","authors":"F Cigana, B Kerebel, J David, F Doumenjou, R Da Costa Noble","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>12 patients (7 men and 5 women) with advanced periodontitis and probing depths of 4mm or more were selected to assess the effectiveness of an antiseptic: betadine used in situ, as a subgingival irrigation in periodontal pockets. Although widely used in other fields of medicine such as gynecology and dermatology, betadine has not been used in periodontology. However, betadine is an excellent antiseptic with a wide range of action. The slow release of iodine explains why this antiseptic is well tolerated by mucosae. The oral cavity was divided in 2 parts: the left side was irrigated with betadine while the right side was irrigated with saline which was used as a control. Irrigations were performed once a day during a period of 15 days. Patients were able to carry out the irrigations themselves after careful, instruction. Thc practitioner carried out the irrigation on day 0 and day 8. The comparison of clinical and histological data showed a decrease in gingival inflammation after 15 days of treatment. By means of the Student \"t\" test significant decreases in the plaque index, the gingival, index and the bleeding index were observed between day 0 and day 15 as well as a significant increase in recession. These clinical results were correlated with histological findings. {"title":"Toothbrushing with the root of Euclea natalensis.","authors":"I Stander, C W Van Wyk","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The custom of cleaning teeth and the gums with a chewed root of the tree Euclea natalensis, in the belief that it benefits oral health, is practised in South Africa by married women of an African Zanzibari community. The root of E. natalensis contains naphthoquinones which are bactericidal and, as they are also pigments, the mouths of the root users are stained bright orange. Samples of fresh root were tested against Streptococcus mutans and organisms in saliva and from deep periodontal pockets. Aerobic as well as anaerobic bacterial growth was suppressed in all instances. In addition we compared the plaque index (Silness et Löe), the periodontal index (CPITN), the DMFT and the state of the oral mucosa of root users and non-users. No significant differences were recorded in any of the variables. The two populations of fibroblasts were very similar in morphology, the only difference found in the parameters measured being the increased number of microtubules in the enamel-related connective tissue. (1989) and Regad et al. (1989). Two liner materials were selected for this experiment which had already been subjected to both in vivo and in vitro tests. After 2, 7 and 14 days of contact with the test materials, the cell responses were evaluated under the light microscope by their growth pattern and morphological aspects. Cell synthesis of fibronectin and type I and III collagen was studied by immunofluorescence. Surface appearance of the cells in contact with the materials as well as their structures were observed by scanning electron microscopy. This study confirmed the viability of the in vitro cytotoxicity assessment methods with cell cultures. {"title":"Mineral content of different areas of human dentin in hypophosphataemic vitamin D-resistant rickets.","authors":"E L Hietala, M A Larmas","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Calcium, phosphorus, fluoride, sodium, magnesium and zinc estimations were carried out on teeth from a patient with hypophosphataemic vitamin D-resistant rickets (HVDRR) and from a patient with acquired rickets with the aim of determining differences in the composition of dentine in these two types of rickets. Normal deciduous teeth served as controls. Mineral analyses were carried out using an electron probe micro-analyser after carefully polishing the hemisected specimens. After the analyses the specimens were coated with gold-palladium for more detailed SEM studies. The Ca, P, F and Zn contents of the calcospherites were normal, while there was more Na and less Mg in the dentine of HVDRR teeth than of controls. The significance of this remains unexplained. The mineral content of the interglobular spaces was very limited, but there was more Zn in these than in other parts of the HVDRR teeth, in the acquired rickets teeth or in the control teeth. The excess of Zn in the interglobular spaces is thought to have an effect on the mineralisation process in HVDRR teeth. The globular nature of HVDRR teeth is thought to be genetically controlled and the result of a reduction in the number of calcification nuclei. The globular nature of the HVDRR teeth was not due to lack of Ca and P, as the serum levels of these minerals were maintained within normal limits during tooth development by controlled phosphate supplementation. {"title":"Possible functions of alkaline phosphatase in dental mineralization: cadmium effects.","authors":"J H Wöltgens, D M Lyaruu, T J Bervoets","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In mineralizing dental tissues the non-specific alkaline phosphatase, using paranitrophenylphosphate (p-NPP) as substrate, is also capable of splitting inorganic pyrophosphate (PPi). In contrast to the p-NPP-ase part of the enzyme, the PPi-ase part requires Zn2+ as a cofactor for its hydrolytic activity. The PPi-ase activity of the enzyme can be inhibited by cadmium ions (Cd2+), perhaps by replacing Zn2+ from the active site of the enzyme molecule. In addition to splitting PPi, the PPi-ase part of the enzyme may also be involved in the phosphorylation process of yet undetermined organic macromolecules. Cd2+ inhibits this phosphorylation process. Inhibition of the PPi-ase activity can also be accomplished by ascorbic acid known for its capacity to complex bivalent cations. Ascorbic acid may accordingly also remove Zn2+ from the active site of the PPi-ase. The target antigen was the hemagglutinating adhesin HA-Ag2 of Bacteroides (Porphyromonas) gingivalis precipitated from a crude antigen extract with a rabbit antiserum in crossed immunoelectrophoresis. The immunization protocol used included: subcutaneous injection of 1 IP in Freund's complete adjuvant to Balb/c mice on day 0 and of 0.5 IP on day 8, followed by an intravenous booster injection of outer membranes on day 15, 4 days before the fusion. Of the 9 MAbs obtained, 8 were specific for HA-Ag2 since they reacted with its characteristic electrophoretic bands at 43 and 49 kDa. The ninth MAb was specific for the B. gingivalis lipopolysaccharide. After the treatment both parts were demineralized, and the artificial caries lesions were analyzed by means of quantitative microradiography. Treatment during only 1 minute with an aqueous solution of 1.5 mmol/l of the surface active compound 2-O-stearoyl-glycerol 1,3-bisphosphate (Glydip) resulted in a decrease in the rate of enamel demineralization by about 60%. When Glydip was dissolved in a toothpaste/water slurry it inhibited the demineralization by about 30%. When the enamel was covered by a salivary pellicle prior to the application of the slurry, the inhibition of the demineralization disappeared. However, when Glydip was applied in an aqueous solution, the presence of a salivary pellicle did not interfere with the decreasing effect of Glydip on the demineralization. {"title":"Immunolocalization of a 110 kD molecule and a 150 kD molecule in rat incisor and mandibular bone.","authors":"H Chardin, D Septier, M Goldberg","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In the present study, antibodies against rat dental proteoglycans were used to characterize and localize the proteoglycans in rat incisor and mandibular tissues. Polyclonal rabbit antibodies were raised against a CPC-precipitated fraction of a sulfated dental extract. In unpurified dental extract these antibodies recognized two molecules of 110 kD and 150 kD. The 150 kD molecule was susceptible to chondroitinase ABC digestion but the 110 kD molecule resisted this enzymatic degradation. Immunocytochemically these two molecules were seen to be located in the pulp, the enamel organ and the mandibular bone. In each tissue only the periphery of the cells was stained and not the intracellular compartment. In the mineralized area of bone, dentin and forming enamel no staining was seen. These results indicate common epitopes in the proteoglycans from pulp, predentin, enamel organ and bone.
