This study compares both the drop in peroxidase activity following filtration (49.3% +/- 16.4%; N = 12) or centrifugation (33.6% +/- 15.9%; N = 6) and the drop in total protein concentration after filtration (33.9% +/- 13.4%) and centrifugation (23.6% +/- 8.7%). After filtration, the loss was not only due to retention of the large particle-bound fraction but also to adsorption on the filter membranes. Limits related to weight agreed well with the usually recommended ranges for supplemented fluorides. However, for children up to 6 years of age limits related to energy intake were twice as high as those related to body weight. These differences diminished with increasing age and were small between 16 and 19 years of age. New conservative limits were based on an interval for adults ranging from 1.45 to 2.9 mg fluoride per day. These antimicrobial factors were also analysed in relation to the salivary levels of mutans streptococci (MS). The amount of MS correlated significantly (+0.31, p less than 0.05) with the number of initial caries lesions (Di) but not with other caries indices (DMFT, DMFS, DS). The group with no Di (N = 17) had significantly (p less than 0.05) more hypothiocyanite (HOSCN/OSCN-) and anti-S. mutans IgA antibodies in whole saliva than those with initial caries lesions (N = 42). None of the antimicrobial factors alone showed any significant association with salivary MS counts. The soluble dentine proteins were isolated according to their extractability in successively: 4M guanidine-HCl (GuHCl), 0.5M acetic acid pH 3 (after extraction with GuHCl), and 0.01M EDTA pH 7.4 and 0.1M EDTA pH 7.4 (after complete demineralization with acetic acid). The amounts isolated were 1.2, 3.3, 6.1 and 3.7 mg protein per gram dentine, respectively. The GuHCl extractable proteins exhibited the lowest affinity and the acetic acid extractable proteins the highest affinity for HAP. Acetic acid extractable proteins, were significantly less adsorbed than the other protein fractions at pH 5.0. In the present study the effects of the diet were assessed after 90 days. Serum P was significantly reduced (p less than 0.05) in experimental animals; serum Ca was unaltered. Disturbances in body growth had occurred as shown by a reduction in body weight gain (p less than 0.05) in the first 45 days of the experiment and by a decrease in femoral size (p less than 0.02). Osteopenia had also developed: the epiphyseal TBV decreased (p less than 0.05) as well as the thickness of both epiphyseal and metaphyseal trabeculae (p less than 0.001). However signs of partial recovery from this condition were observed as shown by an increased body weight gain in the last 45 days of the experiment (p less than 0.02) and lack of difference in metaphyseal TBV, assumed to be the more labile, between the 2 groups. For such investigations, routine methods are needed. In the present study, a modification of the original method of Tunell and Hart (1977) was used, in which three features of the original alkaline preincubation method (composition, incubation time and pH) were modified. These allowed a single step differentiation of the various fibre types found in rat masticatory muscles, for which the classical technics gave only a weak contrast, not suitable for image analysis. Acid preincubation was also tested but failed to give new information. Among 288 wearers of complete dentures 50 patients severely affected by denture stomatitis and heavily colonized by Candida, were selected. The patients were assigned randomly to two groups: a miconazole varnish group and a gel group. All patients were examined 7 times: before starting the treatment (day 0); during treatment (day 3, 7 and 14); after treatment (day 21, 28 and 35). At each examination a photograph of the palatal mucosa was obtained and quantitative cultures of Candida from the lesions and the fitting denture surface were performed. At day 14 the inflammation was reduced to the same degree in the two groups of patients. A comparison of the antifungal effect, i.e. reduction of the yeast score, showed no significant difference between the two groups. Maximum reduction of the number of yeasts was obtained at day 14. From that moment, recolonization started; thus, about 60% of the treated patients whether it was with the gel or the varnish was positive of Candida by day 35. In conclusion, the present study showed no difference in the clinical and antifungal effect of a single-dose application of miconazole varnish compared with miconazole gel. The varnish seemed to be better with respect to the posology as the total dose of miconazole is minimal and only one application is necessary. Cases of gingival hyperplasia associated with their use are repeatedly reported, therefore the question deserves to be restated. Under these conditions and in the presence of physiological level of calcium, colony formation was enhanced and stratification was slowed down. The strong attachment of the cells to glass slides permitted immunocytochemical examination of cytokeratin (CK) expression and their organization within individual cells during the different stages of epithelial maturation. The present results showed that during the stage of cell migration and colony formation, the cells express the same set of cytokeratins (basal cell marker 14, simple epithelial markers 8, 18 and 19, and marker of hyperproliferation 16) which forms a well-defined network of organized filaments. At the stratification stage, the filament network became dense by the additional expression of the markers of differentiation in non-keratinized stratified epithelia (CK 4 and 13). These appeared once individual cells started to overlap the basal cells, a period during which the cell-temporarily changed morphology. Whilst the suprabasal cells exhibited dense filament network labelled for CK 4 and 13, the density of labelled filaments for CK 14, 8 and 18 was much lower, indicating that these cells contained newly-formed filaments lacking the basal and simple epithelial keratins. The simple epithelial cytokeratins became weakly labelled in older cultures. The uncoupling of paired expression of cytokeratins 4 and 13 was observed in non-colony forming aged cells.
