It has been difficult in the past to move a tooth only over this small distance in order to activate the periodontal receptors without at the same time activating other receptors in related structures. This paper describes a simple solution to this problem based on the use of von Frey hairs (in the form of nylon filaments) rather than a rigid probe, to move the tooth. Filaments of different stiffness are mounted on the moving coil of a loudspeaker to apply different forces to the tooth. The timing of the contact of the filament with the tooth is measured precisely with a simple impact detector. The sensation elicited by this stimulator is abolished by local alveolar anaesthesia, indicating that the stimulation was localised to the periodontal area. The flexible probe is an important safety feature, as it limits the force applied to the tooth, and therefore the distance that the tooth can be displaced. In addition, these data demonstrated as that 0.006% F- given either as the quaternary ammonium compound or NaF completely prevented demineralization by acting primarily on the dentine mineral. The purpose of the present study was to re-examine the questions of the existence, nature and significance of Von Korff's fibers using light microscopy and immunohistochemistry. Specimens were obtained from 3 days-old CD-I mice and mandibles were carefully dissected under constant irrigation and immediately fixed in 10% neutral buffered formalin for light microscopy. Sections were treated or not with collagenase prior to silver staining. For immunohistochemistry, specimens were fixed in 95% ethanol and embedded in paraffin. Sections were reacted with goat anti-human-bovine type I or type III collagen and a rhodamine (RITC) labelled rabbit anti-goat IgG was then reacted as a secondary antibody. Slides were then examined under a Zeiss II photomicroscope equipped with epifluorescence. Our results have confirmed the presence of argyrophilic material concentrated at the periphery of the dental papilla and stretching from the subodontoblastic layer to the future dentino enamel junction. The distribution of type III collagen was very similar to the distribution of the silver staining at the cervical loop area. Type I collagen distribution was different and concentrated in areas where odontoblasts were fully differentiated. Our study showed that Von Korff fibers are not artefactual. Monoclonal antibodies to CD1a (specific for Langerhans cells and thymocytes) and HLA-DR (class II major histocompatibility antigens - (MHC)) were used to identify Langerhans cells within gingival biopsies taken on days 0, 7, 14 and 21. HLA-DR antibody stained dendritic cells within the oral epithelium which were morphologically identical to the CD1a+ Langerhans cells. Class II MHC LC numbers rose and plateaued between day 7 and 14 then decreased to baseline by day 21. As plaque accumulated and initial inflammation developed there was an increase in the number of CD1a+ Langerhans cells which peaked at day 7 and stayed high (day 14). As inflammation developed there was a statistically significant decrease in the number of CD1a+ Langerhans cells by day 21 (p = 0.028). These events were observed clinically during three consecutive intervals. Only correlations, significant during at least two random intervals were discussed. The Snyder test determined at 24 hours was significantly positively correlated with most events confined to the enamel surfaces and only the Snyder test determined at 48 hours in stimulated saliva appeared to be significantly positively related with deeper processes such as progression. In addition, the mineral composition of Ca2+, P and Mg2+ in saliva was significantly related to cariogenic events in the enamel surfaces of girls, and its viscosity for boys. Finally, and most interesting from a clinical point of view, was the fact that caries progression was negatively related with P concentrations in both unstimulated and stimulated saliva for girls. The difference in acid resistance between a pair of enamel pieces obtained from the same tooth was examined with atomic absorption spectrometry. After etching with a lactate buffer solution at pH 4.5, the amount of Ca dissolved from the enamel in each pair by brushing without dentifrice was significantly lower than that when brushing with an by abrasive paste. Under SEM, the brushing with paste caused prism structures to appear clearly on the enamel surface, while brushing without dentifrice caused a nonbacterial organic film, i.e., salivary pellicle, to cover the surface. These results indicate that the pellicle is protected by mineral deposits when brushing without dentifrice but brushing with an abrasive paste tears off the unmineralized pellicle. The aim of this study was to measure the adherence potential of Actinobacillus actinomycetemcomitans (A.a.) for buccal epithelial cells (BEC). In the second part of the study, the inhibitory potential of saliva on adherence between A.a. and BEC was tested. Saliva specimens were obtained from three groups of subjects: a group of normal subjects (NS), a group of subject with localized juvenile periodontitis (LJPS) and a group of subjects with adult periodontitis (APS). Clinical strains of A.a. showed a major adherence potential for BEC (25.5 A.a./BEC). In contrast, reference strains of bacteria appeared to possess a much lower potential for adherence (12.6 A.a./BEC). The culture conditions were standardized in order to evaluate simultaneously the expression of the three antigens in four different culture passages of the two fibroblast types. The optical density values of immunostaining intensities were quantified, the integrated optical density per cell was calculated, and the results were analyzed by a variance test. It was found that all three collagen types were present in the tissues, and in both gingival and pulp fibroblasts after three to nine culture passages. A non-parametric statistical analysis of the staining intensity variances revealed significant differences between antigenic levels depending on the tissue origin of the fibroblasts and an effect of culture passages. This IR method allows a dish to be placed directly on the sample beam window of a normal double beam IR spectrometer. Placing a blank dish on the reference beam window of the spectrometer compensates for the high background absorption due to the dish on the sample beam window and enables an identifiable spectrum to be obtained. Using this technique with a Petri dish, a high correlation factor of 0.998 was found between the PO4 peak absorbance and the weight of synthetic hydroxyapatite in the range of 0.5-2.5 mg/cm2. The IR pattern, however, showed a shift in the positions of the absorption band and the appearance of ghost peaks. Using a thin based Petriperm dish, these difficulties were overcome producing a correlation factor of 0.992 at the lower range of 0.05-1.45 mg/cm2. Vitamin D3 a true endogenous or alimentary prohormone is converted into its main metabolite, calcitriol, by successive hydroxylations in the liver in position 25 and in the kidney in position 1, the production of which is controlled by several factors including parathyroid hormone, blood calcium and phosphorus or insulin as well as by the metabolites of the hormone itself. It controls the synthesis of numerous peptides by acting on gene expression. Indeed, several structural proteins are involved including procollagen alpha 1l, core protein of proteoglycans, diverse regulatory peptides such as protooncogene c-myc and growth factors, \"Tumor Necrosis Factor or TNF\" and \"Nerve Growth Factor or NGF\" or hormones such as parathyroid hormone, and finally constitutive proteins of the mineralized tissues such as osteonectin, osteocalcin, osteopontin and calbindins. Therefore, it modulates very different cellular processes. It acts via a nuclear receptor the structure and function of which have been investigated by genetic engineering (cloning of genes encoding for the receptor and hormono-dependent peptides, transfection assays, directed mutagenesis). Actual studies investigate its role in the formation of the complex for transcription initiation near ADN sites, the \"Vitamin D Responsive Element or VDRE\", located upstream vitamin D-responsive genes and approximately RNA polymerase II.
