TNF alpha treatment of human umbilical vein endothelial (HUVE) cells induced urokinase-type plasminogen (uPA) activity, increased HUVE uPA-dependent extracellular matrix (ECM) degradation, and accelerated matrix remodeling and endothelial differentiation into tubes or cord-like structures. All of the aforementioned effects of TNF alpha on HUVE uPA-dependent activities were abrogated by co- or pretreatment with IFN gamma. In contrast, endothelium derived from human lung (HLE) exhibited high constitutive uPA and uPA-dependent matrix degradation and rapid tube formation in Matrigel, activities all unaffected by TNF alpha or IFN gamma. Endothelium derived from human rheumatoid synovium (HSE) exhibited uPA-dependent activities intermediate between the HLE and HUVE. TNF alpha or IL-1 treatment of HUVE potently induced surface ICAM-1 expression, whereas these cytokines were relatively ineffective on HLE and HSE ICAM-1 expression. Co-incubation with IFN gamma synergistically elevated TNF alpha or IL-1 induced ICAM-1 expression in HUVE, HLE, and HSE. The major prostaglandin synthesized by HUVE was PGI2, in contrast to HLE and HSE which produced PGE2 as the major product. Although cytokine treatment increased prostanoid production in all three cell types, HLE were not responsive to IL-1, and HSE demonstrated the greatest increase in prostaglandin synthetic capacity. These studies underline important differences not only in the \"constitutive\" activities expressed by EC from different vascular beds, but also in the responsiveness to proinflammatory cytokines alone or in combination. These observations further emphasize the need to study the endothelial cell derived from the vascular bed of interest rather than extrapolate from results obtained with HUVE or other macrovessel-derived endothelium.
","PeriodicalId":75604,"journal":{"name":"Blood cells","volume":"19 2","pages":"325-39; discussion 340-2"},"PeriodicalIF":0.0,"publicationDate":"1993-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"18903128","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The assumption has been that they are therefore of no significance. They are commonly seen in blood films from patients with lymphoproliferative syndromes and particularly in chronic lymphocytic leukemia (CLL). In 96 patients with CLL a statistically significant correlation existed between the basket cells observed in films and the lymphocytes with dense chromatin (DC) determined by flow-cytometry. There was no statistically significant correlation between the number of BC and DC, and the anatomic-clinical stage of the disease.
","PeriodicalId":75604,"journal":{"name":"Blood cells","volume":"19 3","pages":"573-81; discussion 582-9"},"PeriodicalIF":0.0,"publicationDate":"1993-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"19014749","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

{"title":"Vasoactive hormones and autocrine activation of capillary exchange barrier function.","authors":"V H Huxley, M K McKay, D J Meyer, D A Williams, R S Zhang","doi":"","DOIUrl":"","url":null,"abstract":"

Capillary barrier function is subject to changes in Starling forces via hemodynamic status (hydrostatic pressure) or protein milieu of fluids bathing the wall (oncotic pressure). Venular function is sensitive to inflammatory mediators leading to white cell sticking, fluid and formed element extravasation, and flow disruption. Thus, we hypothesized that vasoactive hormones and autocrines alter preferentially the venular-capillary (VC) barrier. Hydraulic conductivity (Lp) of frog mesenteric venular- and true-capillaries (TC) was measured by the modified-Landis technique under control (LpC), then during atrial natriuretic peptide (ANP, 10(-7) to 10(-8) M), bradykinin (BKN, 10(-7) M), acetylcholine (ACh, 10(-6) to 10(-5) M), angiotensin II (AII, 10(-7) M), or norepinephrine (NE, 10(-6) M) perfusion. All agents, except AII or NE, elevated Lp: LpANP/LpC = 2.9 +/- 0.3 (mean +/- SEM; (n = 55), LpBKN/LpC = 3.3 +/- 0.8 (n = 16), LpACh/LpC = 1.6 +/- 0.1 (n = 26), LpAII/LpC = 1.1 +/- 0.2 (n = 8), and LpNE/LpC = 1.1 +/- 0.2 (n = 9). Contrary to our hypothesis, VC and TC responded similarly: 3.0 versus 2.9 for ANP, 3.4 versus 3.2 for BKN, and 1.6 versus 1.6 for ACh, respectively. These data are consistent with putative vasodilators lowering capillary barrier resistance independent from changes in Starling forces.
","PeriodicalId":75604,"journal":{"name":"Blood cells","volume":"19 2","pages":"309-20; discussion 320-4"},"PeriodicalIF":0.0,"publicationDate":"1993-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"19300498","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

First, cells were stimulated with N-formyl methionyl leucyl phenylalanine (FMLP) with or without depletion of free intracellular calcium (by incubation with BAPTA/AM in calcium-deprived medium). Control cells displayed a rapid and transient rise of free intracellular calcium (peaked after a few seconds, returned to the basal level after 2-3 minutes), extensive morphological polarization (more than 90% polarized cells after 15 minutes), and reorganization of actin filaments (as assessed by image analysis on cells labeled with a fluorescent phallacidin derivative). Calcium-depleted cells displayed no calcium rise, but both morphological polarization and actin reorganization were indistinguishable from those of controls. In a second set of experiments, individual granulocytes were labeled with fluorescent calcium probes and aspirated in a micropipette on the stage of a confocal laser microscope. About half of the cells displayed a transient calcium rise, which was concomitant with the formation of a protrusion in most cases. However, extensive protrusions were shown by the cells that did not exhibit calcium flashes. Furthermore, if cells were treated with ionomycin, in calcium-containing or calcium-deprived medium, then aspirated, they showed protrusions comparable to those of controls. Finally, when cells were treated with ionomycin during micropipette aspiration, a calcium rise was followed by a moderate increase of the rate of protrusion growth. It is concluded that free calcium alone cannot play a major role in the control of cell shape.
","PeriodicalId":75604,"journal":{"name":"Blood cells","volume":"19 1","pages":"115-29; discussion 129-31"},"PeriodicalIF":0.0,"publicationDate":"1993-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"19386039","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This deoxyadenosine analog was selected from 25 candidate compounds because it was resistant to deamination by ADA and because it had a favorable therapeutic index. The lymphoid specific action of this drug is a function of the high deoxycytidine kinase and low 5' nucleotidase activity of lymphocytes. The drug appears to trigger apoptosis, very likely by preventing the repair of nicks in double-stranded DNA. Accordingly, 2-CdA has been found effective against a variety of lymphoid neoplasms. Hairy cell leukemia has proved to be most sensitive to its action, with approximately 80% of patients achieving a complete, long-lasting remission after a single five- or seven-day infusion. 2-CdA also shows promise in the treatment of autoimmune disorders.
","PeriodicalId":75604,"journal":{"name":"Blood cells","volume":"19 3","pages":"559-68; discussion 569-72"},"PeriodicalIF":0.0,"publicationDate":"1993-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"19014748","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The experimental data are those of Nakahata et al. [J. Cell. Physiol., 113, 455-458, 1982], who studied the development of primary and secondary murine hemopoietic colonies in vitro. The cumulative probability distributions of the secondary colonies are simulated by a simple proliferation tree model. The model consists of four subsequent generations of distinct cells undergoing self-renewal and differentiation governed by an unspecified control process. It is assumed only that the control mechanism is so complex that the resulting commitment can be well described by probabilistic formalism. The calculated and experimental distributions are found to be in reasonable agreement. Furthermore, 25 additional experimental distributions have been examined, corresponding to various sets of in vitro and spleen-grown colonies reported in the literature. It is shown that most of the cumulative distributions analyzed are well approximated by the exponential function f(n) = exp(alpha n), where f(n) is the relative number of originating elements (e.g., cells or clones) producing groups having at least n members. A formula is suggested relating the exponent alpha and the probability of self-renewal for a simple single-generation case.
","PeriodicalId":75604,"journal":{"name":"Blood cells","volume":"19 2","pages":"435-60; discussion 461-73"},"PeriodicalIF":0.0,"publicationDate":"1993-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"19299105","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Immunoglobulin genes in chronic lymphocytic leukemia. 慢性淋巴细胞白血病的免疫球蛋白基因。

{"title":"Immunoglobulin genes in chronic lymphocytic leukemia.","authors":"T J Kipps","doi":"","DOIUrl":"","url":null,"abstract":"

Common B cell chronic lymphocytic leukemia (CLL) cells frequently produce polyreactive autoantibodies. Molecular studies have revealed that the leukemia cells in this disease use a restricted repertoire of conserved immunoglobulin variable region genes (Ig V genes) that generally have not undergone somatic mutation. Studies indicate that this polyreactive autoantibody activity expressed in CLL is dependent upon the third complementarity determining region (CDR) of the heavy chain variable region and on proper pairing of the Ig light and heavy chains. In this light, the high frequency at which CLL patients have leukemia cells that make IgM autoantibodies does not appear secondary to the genetic constraints on the potential diversity of the antibodies that may be expressed in this disease. Rather, even though they express nonmutated Ig V genes, CLL B cells appear to have been selected for their ability to bind self-antigens. Conceivably, normal B cells that express such autoantibodies may be perpetually stimulated, thereby increasing their risk for malignant transformation into CLL. Alternatively, anti-self-reactivity may enhance the survival of a B cell clone subsequent to its malignant transformation. In either case, the autoantibody activity of the immunoglobulins expressed by leukemia B cells may be an important factor in the etiopathogenesis of this disease.
","PeriodicalId":75604,"journal":{"name":"Blood cells","volume":"19 3","pages":"615-25; discussion 631-2"},"PeriodicalIF":0.0,"publicationDate":"1993-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"19014751","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
