In order to define the mechanism of subunit specific action of ginkgolic acid we have performed comparative analysis of the amino acid sequences of at and α2 subunits of glycine receptor. Amino acids that contribute to the different action of ginkgolic acid on glycine receptors formed by these subunits were determined. Using whole-cell configuration of patch-clamp recording, we have demonstrated that mutation of three residues in α2 subunit for corresponding ones from αl subunit makes α2 receptors sensitive to the potentiation by ginkgolic acid. Currents, mediated by α2 mutant receptors, increased by 89±14% after application of ginkgolic acid. Similarly to ai receptors α2 mutant receptors have shown a decrease in EC₅₀. for glycine under the action of ginkgolic acid. {"title":"CALIX[4]ARENES AS MODULATORS OF ENERGY-DEPENDENT CA2+_ACCUMULATION AND FUNCTIONING OF THE ELECTRON TRANSPORT CHAIN IN SMOOTH MUSCLE MITOCHONDRIA.","authors":"H V Danylovych, Yu V Danylovych, R V Rodik, V I Kalchenko, A Ju Chunikhin","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The influence of supramolecular macrocyclic compounds calix[4]arenes (C-97, C-99, C-107) at a concentration of 100 nM in the process of energy-dependent Ca²⁺-transport in isolated mitochondria of smooth muscle, as well as autofluorescence mitochondrial coenzyme NADH, FAD and hydrodynamic diameter of these organelles was investigated. Using Ca²⁺-sensitive fluorescent dye Fluo-4 AM it was shown that the selected calix[4]arenes can suppress energy-dependent accumulation of Ca²⁺ by mitochondria. Accumulation of Ca²⁺ (80 jiM in the medium) accompanied by the growth of the fluorescent probe response from a conventional unit to a value of 1,57±0,04 (n=5). Calix[4]arenes C-97, C-99, C-107 falls fluorescent signal below the 0,88±0,08, 0,92±0,08 and 0,78±0,04 respectively. Thus, the selected calix[4]arenas lead to release of previously accumulated Ca2+ from mitochondria. Under the influence of C-97 and C-99 fluorescent signal from NADH reduced to -0,11±0,02 and -0,12±0,02, respectively, in relation to the reference value - -0,05±0,01 (n=5). Analysis of fluorescence response NADH and FAD in a suspension of isolated mitochondria suggests that the effects of test compounds on the functional activity of the electron transport chain is associated with the initial stimulation of its 1-th complex and subsequent inhibition of Ca²⁺-dependent NAD- containing Krebs cycle dehydrogenases. Along with this, the use of photon correlation spectroscopy to assess changes in the volume of mitochondria (their hydrodynamic diameter) under the action of selected calix[4]arenes has shown that interference with the electron transport chain leads to changes in the osmotic balance between the matrix of the mitochondria and the external environment. The result is the growth of isolated organelles volume. In particular, the hydrodynamic diameter of mitochondria increased by 22±6 % and 34±8 % (n=5) in presence of C-97 or C-99. It is shown that young men who systematically performed muscular work have a high content of markers of different ways of generation of superoxide radical, a reactive oxygen species for products of lipid peroxidation and markers of nitrosative stress. The increase in the degree of adverse effects on the body intensive training and competitive loads is accompanied by pronounced adaptive changes in the hierarchy of oxidizing constitutive de novo synthesis of nitric oxide, as well as its nonoxide reutilization synthesis (in 3 times higher). Disadaptation of the organism of boys at the end of the competition period is reflected in growing levels of generation of ROS (superoxide radical: 3,5 times higher, hydrogen peroxide: 2,5 times higher). The method of microfluorescent microscopy and Ca2+ sensitive dye fura- 2AM. Registration of changes in the concentration of intracellular Ca2+ was performed by using the ratio of fluorescence signals measured at two wavelengths (R = F1/ F2). Researches were conducted on cultured neurons of rat dorsal ganglia (DRG neurons). Application of AITC and Caps on soma of DRG neurons resulted in an increase in intracellular Ca2+. Consistent repeated Caps applications resulted in a significant reduction in the amplitude of Ca2+ transients ( desensitization of TRPV1 channels), which accounted 20,7% of initial value. Further application of selective TRPA1 channel agonist (AITC) resulted in restoration of sensitivity to capsaicin TRPV1 channels ( resensitization TRPV1 channels). Noradrenalin and salbutamol were used to investigate peptidoglycane impact on the myometrial β-adrenergic receptors. It was shown that inhibited by these substances myometrial contractility increased to the initial level after peptidoglycane application. The same effect we observed under the cAMP level elevation by forscolin. Peptidoglycan’ s ability to strengthen contractions was inhibited by the 8-brom-cAMP and papaverine application. Stimulation of Gs-protein by the cholera toxin didn’t influence on the peptidoglycane effect while the blocking of Gi/o-protein by the pertussis toxin caused stopping it’s manifestation. Following polymorphisms have been studied: G894→T of the eNOS exon 7 and Т-786→С of the eNOS promotor, А1266→G of the Eln exon 16, Т-381→C of the NPPB promotor, ІD of the ACE, Arg60→His of the LMP2, Met235→Thr of the AGT, A1166→C of the ATR1, C-1562→T of the MMP9, C-1306→T of the MMP2, and С-8→G of the PSMA6. It was shown that children with genotypes G/T by eNOS (G894→T), G/G by Eln (А1266→G), C/C by NPPB (Т-381→C) and І/D by ACE genes have lower EDVD (Р<0,05) than patients with others allelic variants of these genes, and this does not depend on duration of the disease, level of glicated hemoglobin and initial diameter of a humeral (brachial) artery. There is established a high number of links between indicators of rheoencephalogram (REG) and electroencephalogram (EEG) in the left cerebral hemisphere than in the right. In frontomastoidal allocation 19 correlations and in occipitomastoidal - 59 links. We suppose that poor circulation in vertebroplasty-basilar system leads to the defeat of the brain stem, which, with afferent pathways of the reticular formation, connects the thalamus with the cortex. In the reticular formation there is an inhibition of ascending activators influences, which eland to decreasing of the cortex is tonus. You can talk about the functional immaturity of the system of nonspecific activation by the reticular formation of the brain stem. Children with violation of motor activity had significantly more negative and positive significant and high correlation among the existing indicators of electric brain activity and cerebral hemodynamics, in our opinion, is due to the development of interconnection compensation that is carried out by adjustment of the functional systems and the formation of new forms of adaptive responses in conditions of disontogenetik. Feature correlation pattern of the EEG, of children with disorders of motor activity, is associated with a significantly great number of high and significant correlations between measures of electrical brain activity in the δ- and q- rhythms, especially in the temporal areas of the cerebral cortex. According to visual analysis of EEG there is revealed a common manifestation of changes of bioelectric brain activity in children with disorders of motor activity. This is manifested in the development of paroxysmal activity of action potentials of θ- and δ-rhythms with the focus of activity in the anterior areas of the cerebral cortex; the formation of a mosaic representation of the θ-rhythms in temporal areas; the presence of hypersynchronous a-paroxysms in the posterior areas of the cerebral cortex. The given facts testify to activation of mechanisms of limbic-neocortical systems and synchronizing influences of the reticular formation of the stem and diencephalic structures. We inspected 15 practically healthy persons in which the content of myeloperoxidase was 74,5±16,3 ng/ml, 13 patients with a stable stenocardia (the conent of myeloperoxidase was 218,4±30,9 ng/ml) and 60 patients with acute infarct of myocardium (the content of myeloperoxidase was 606,0±59,3 ng/ml at the beginning of therapy) in the dynamics of treatment. In patients accepting standard treatment (without application of Korvitin), the content of myeloperoxidase was unchanged during 7 days of treatment. It patients administered korvitin, the action of this preparation depended on the initial level of myeloperoxidase. In autumn experiments,\u0000introduction of melatonin to young rats caused an increase of\u0000alkaline phosphatase by 43,9% and hialuronidase activity by\u000015,4% and decreased the content of glycosaminoglycans by\u000046,7%. The concentration of total lipids and triglycerides was\u0000decreased. In spring, the introduction of melatonin to young\u0000rats inhibited the physiological remodeling of bone tissue,\u0000reduced the activity of alkaline phosphatase and increased\u0000the activity of acid on the 78% and tartrate resistant acid\u0000phosphatases by 72% regardless of the oxygen consumption\u0000level. The concentration of glycosaminoglycans and free amino\u0000acids increased. Genotyping for FLG (rs11204981) was performed in the following populations:\u0000patients with asthma (n = 99); ages 5-18 years (8 ± 2.1), and control group (n = 98); ages 5–18 years\u0000(12 ± 2.1) by using Real-time PCR. Level of mRNA expression was estimated by using reverse transcription\u0000and following real-time PCR. It was found out that 5.05 % of patients and 2.02 % of control group had\u0000minor allele (AA; P>0.05), 27.27 % and 36.36 % of patients and control group, respectively, had heterozygous\u0000allele (GA; P>0.05) and 67.68 % and 61.62 % had major allele (GG) (P>0.05). Variants with the\u0000AA-genotype of the FLG rs11204981 were found to be 2.5 times more frequently among patients than in\u0000control group. We also found out that the level of mRNA FLG expression in GG-genotype is 22.8 ± 11.67\u0000(P>0.05 compared to AA-genotype), 92.95 ± 35.3 in GA genotype (P<0.05compared to GG-genotype) and\u000021.8 ± 13.4 in AA genotype (P>0.05 compared to GA-genotype). Thus, heterozygous variant has significantly\u0000higher expression of filaggrin in buccal epithelium.
