Firstly, the CNN model obtained by large-scale natural image training was used to migrate the characteristics of the renal occupancy lesions CT images, and then the fine-tuning of the full connection layer was used to realize the benign and malignant classification of the images. \u0000 \u0000 \u0000Results \u0000The evaluation indexes of the VGG19 model were lower than ResNet50 and Inception V3, and the training result showed obvious overfitting. In the fatigue load group, RAW264.7 cells were treated with 5 000 μe mechanical stretch strain, and then cultured in an osteoclast culture medium that was an α-MEM complete medium containing 40 ng/ml macrophage colony-stimulating factor and 40 ng/ml osteoclast differentiation factor. In the mechanical stimulation + ICA group, RAW264.7 cells were treated as the same procedure in the fatigue load group, and then cultured in an α-MEM complete medium containing 1×10-5 mol/L ICA simultaneously with a 1000 μe tensile strain on the substrate. The activity of tartrate-resistant acid phosphatase (TRAP) was detected using a TRAP assay kit. The mRNA expression of the osteoclast marker genes, i.e. TRAP, cathepsin K(CTSK) and matrix metalloproteinase 9 (MMP-9) was detected by real-time RT-PCR. The nuclear translocation of nuclear factor kappa B (NF-κB) was analyzed by Western Blot. \u0000 \u0000 \u0000Results \u0000Compared with the fatigue load group, the combination of mechanical stimulation (1 000 μe substrate stretching) and ICA (1×10-5 mol/L) could significantly inhibit the activity of TRAP in osteoclasts (P<0.01) and reduce osteoclastosis. In the drug-loaded nanocarriers, CpG was covalently bound to the surface of HA, and OVA was physically blended into DOTAP-PLGA nanocarriers. The nanoparticles were characterized by transmission electron microscopy and dynamic light scattering. The in vitro release of CpG and OVA in the nanoparticles was investigated. The uptake and distribution of nanoparticles in mouse BMDCs were studied by flow cytometry and laser scanning confocal microscopy. The maturation and cytokine expression of mouse BMDCs were evaluated by flow cytometry and enzyme-linked immunosorbent assay, respectively. \u0000 \u0000 \u0000Results \u0000The CpG-HA-OVA-PLGA nanoparticles loading CpG and OVA were prepared. The average particle size was (305.1±2.2) nm and the polydispersity index was 0.203. A core-shell structure of the nanoparticles modified by HA was clearly observed by transmission electron microscopy. Cellular experiment results showed that CpG-HA-OVA-PLGA nanoparticles could be efficiently uptaken by mouse BMDCs, and promote lysosomal release of CpG and cytoplasmic delivery of antigen OVA. Therefore, safe and effective narcotic analgesic for cancer patients is the key factor for successful completion of examination and treatment. Dexmedetomidine (Dex) is a novel α2-adrenergic receptor agonist. Comparing with traditional opioids, Dex is more effective and safer, and can provide sedation, anxiolytic and analgesia after administration. It has the highest incidence of cervical spondylosis and has been classified as a difficult disease by the World Health Organization. Cervical spondylotic radiculopathy mainly manifests as root symptoms and signs consistent with the diseased segment, and the symptoms of stiffness, pain and numbness, which seriously affect the physical and mental health and life quality of patients. In clinical practice, the symptoms of cervical spondylotic radiculopathy can be alleviated by physical therapy, acupuncture, etc. Therefore, searching for more effective treatments has become a hot topic of current researches. Manipulations have the advantages of simple operation, quickness, quick effect and high recognition degree, and can achieve therapeutic purpose by improving the dynamic and static balance of cervical spines, correcting joint dislocation and improving blood circulation. Among them, the curative effect of rotational manipulations is more remarkable. The researches on rotational manipulations by domestic and foreign scholars mainly focus on the in vitro and in vivo biomechanics of cervical spines, and the biomechanical researches on the rotational manipulation itself were different. At present, the mechanical research on the rotational manipulation is still in the development stage, and there is no clear conclusion on the similarities and differences of the mechanical parameters and their influence on the mechanical environment of the cervical spine. Immunological checkpoint molecules play a key role in tumor immune escape, and have attracted much attention in basic and clinical studies recently. Immune checkpoint molecules transmit inhibitory signal to anti-tumor immune cells and inhibit their activity through receptor/ligand interactions, thereby exerting anti-tumor function. Molecular expression of the immunological checkpoints of esophageal cancer is closely related to prognosis. At present, clinical trials of treating esophageal cancer using specific antibodies to block the immune checkpoint related signaling pathways are being widely carried out and show some initial effects. However, there are still some patients who are resistant to this treatment. As a result, it is urgent to find novel biomarkers that can accurately predict the therapeutic effects of anti-tumor immunotherapy for esophageal cancer. In this paper, the researches on esophageal cancer immunological checkpoints and its clinical application were reviewed from four aspects, including the immune checkpoint molecules and their antibodies, the relationship between the expression of esophageal cancer immunological checkpoints and the prognosis, the clinical trials of esophageal cancer immunotherapy and novel biomarkers for predicting the efficacy. The complete virus was used to infect human lung cancer cell lines A549, H1975 and human B-cell lymphoma cell line K562. The stable cell lines were obtained by puromycin selection. Fluorescence microscopy and quantitative PCR were used to confirm the RFP and Fluc expression. \u0000 \u0000 \u0000Results \u0000The lentiviral vector pHBLV-Fluc-RFP was successfully constructed. Cancer cell line A549, H1975 and K562 stably expressing Fluc and RFP was obtained. Subcutaneous administration of insulin forms a portal-peripheral concentration gradient of insulin above the liver, which is inconsistent with normal physiological conditions. Intraperitoneal insulin administration has been extensively investigated because that is closer to physiological state. In this paper, the pharmacokinetic and pharmacodynamic studies of insulin intraperitoneal administration were reviewed. Compared with the conventional subcutaneous delivery, intraperitoneal insulin administration can not only reduce the incidence of hypoglycemia, but also has the advantage of correcting abnormal lipid metabolism. This means that intraperitoneal administration of insulin has a positive effect on the prevention and treatment of diabetic complications. Then, the prepared oyster HA was used as the raw material for porous scaffold preparation by sponge-soaking and sintering, successively. The porosity and compressive strength of the scaffold were adjusted by controlling the soaking time and absorbed HA slurry of the sponges. \u0000 \u0000 \u0000Results \u0000Hydrothermal synthesis was an effective method for preparing oyster HA. When the volume of the sponge cube was 1 cm3, the material absorbed by one to three times sponge-soaking were 0.184 8 g, 0.318 1 g and 0.426 1 g, respectively, the corresponding porosity were 91.5%, 82.9% and 78.5%, and the compressive strength were 1.06 MPa, 3.99 MPa and 8.49 MPa. \u0000 \u0000 \u0000Conclusion \u0000The oyster shell powder can be effectively converted into HA under the hydrothermal reaction condition of 220 ℃ for 6 h. The preparation of HA porous bone repair material by sponge-soaking method can obtain ideal porosity and mechanical strength.
