Oil Red O staining combined with quantitative lipid droplet analysis was conducted to examine the effects of EA on intracellular lipid droplet accumulation. Intracellular total cholesterol(TC) and triglyceride(TG) levels were quantified spectrophotometrically to assess EA&apos;s effects on intracellular lipid levels. Western blot analyzed protein expression of PPARγ, LXRα, ABCA1, and ABCG1 to delineate EA&apos;s pro-foamogenic mechanism.</p><p><strong>Results: </strong>EA dose-dependently suppressed MOVAS viability(P&lt;0.01). EA-treated groups exhibited significant increases in lipid droplet area/number and TC/TG content versus controls(P&lt;0.01). Stratified systematic sampling method was used to select 10 survey sites from 150 surveillance sites based on the Chinese Nutrition and Health Surveillance 2010-2012. demographic information, physical status, lifestyle were collected through questionnaire, physical measurements and laboratory tests. The TyG index was calculated using baseline fasting serum triglyceride and blood glucose, and was divided into quartiles(Q1-Q4). Multiple-adjusted logistic regression was used to test the association between the TyG index and the risk of hypertension, after adjusting for basic demographic information, physical characteristics, lifestyle behaviors, and intake of oil and salt. The restricted cubic spline model was applied to analyze the dose-response relationship between the TyG index and hypertension. Stratified analysis and interaction tests were conducted to evaluate the interaction in different subgroups.</p><p><strong>Results: </strong>A total of 2516 subjects with normal blood pressure at baseline was included, with mean TyG of 8.53±0.67 on baseline. And the rate of new onset hypertension was 57.95%. Significantly positive correlation trend was observed between the TyG index and hypertension(β=1.51, P&lt;0.05). Consistently, high levels of TyG index were significantly associated with increased risk of new-onset hypertension when TyG index was grouped into quartiles. Compared with Q1, the risks were 1.28 times(95%CI 1.01-1.62, P&lt;0.04), 1.39 times(95%CI 1.09-1.76, P=0.01), and 2.11 times(95%CI 1.64-2.71, P&lt;0.01)in Q2-Q4, respectively. Restricted cubic spline analysis suggested a positive linear relationship between the TyG index and the incidence of hypertension(P_(overall)&lt;0.01, P_(nonlinear) = 0.54). The polyphenols and multi-elements in the fruit juices were analyzed by nutrient density(ND) and index of nutrient quality(INQ).</p><p><strong>Results: </strong>The total anthocyanins content in fruit juice was none detected-1664 mg/L, with the highest content in blue honeysuckle juice. The total polyphenols content in fruit juice was 281-2948 mg/L, with the highest content in chestnut rose juice. Potassium had the highest content of macroelements in fruit juice, ranging from 288 to 1996 mg/L and manganese had the highest content of trace elements, ranging from 0.2 to 17.7 mg/L. The ND of potassium, calcium, sodium, magnesium, iron, zinc, copper, and selenium were relatively low, each of them less than 10%. The ND of manganese, chromium, and molybdenum are relatively high, ranging from 0.4% to 39.3%, 0.7% to 35.3% and 0% to 13.2%, respectively. The INQ of total anthocyanins in fruit juice ranged from 0 to 167.63. The INQ of total anthocyanins in fruit juice was greater than 1 except sea buckthorn juice and chestnut rose juice. The INQ of potassium and iron in fruit juices were less than 6, calcium and zinc were less than 2, magnesium and copper were less than 4, and sodium was less than 3. The INQ of manganese, chromium, and molybdenum were higher than that of other multi-elements, ranging from 0.21 to 24.00, 0.27 to 15.76 and 0 to 7.18, respectively. The INQ of selenium in all fruit juice was less than 1.</p><p><strong>Conclusion: </strong>The not from concentrated fruit juices(excluding sea buckthorn juice, chestnut rose juice and cranberry juice(Canada)) are good source of anthocyanins. Blueberry juice, Heilongjiang cranberry juice(China), blackberry juice(Germany), red raspberry juice, black wolfberry juice, Liaoning chokeberry juice(China), and sea buckthorn juice are supplements rich in manganese, chromium, and molybdenum for human body. Protein, dietary fiber, vitamin A, vitamin E, vitamin C, calcium, iron, potassium, magnesium and zinc were considered recommended nutrients, while sodium and fat were considered restrictive nutrients. The nutrient-rich foods(NRF 10.2) model was used for analysis and evaluation. Additionally, total carbohydrates and energy were included for energy contribution analysis.</p><p><strong>Results: </strong>Based on the nutrient data, the median percentages of energy contribution from the three major macronutrients were: carbohydrates 6.8%, fat 50.2% and protein 33.7%. The median NRF 10.2 index was 48.9, with most NRF 10.2 values greater than 0, indicating nutritional value. According to the classification, the medians NRF 10.2 index were ranked as follows: aquatic products&gt;mixed dishes&gt;livestock&gt;poultry&gt;snacks&gt;staple foods. Statistically significant differences in NRF 10.2 index were observed among different categories(H=15.01, P=0.010).</p><p><strong>Conclusion: </strong>The energy contribution of protein and fat from Jiangsu&apos;s special dishes is relatively high, while that of carbohydrates is low. Principal component analysis was conducted on the fatty acid composition, and the proportions of different functional fatty acids were compared. The polyunsaturated fatty acid ratio, atherogenic index(IA) and thrombogenic index(IT) of various chocolate products were calculated, and a health evaluation was carried out.</p><p><strong>Results: </strong>The fat content in chocolates and their products average ranged from 2.90 to 36.5 g/100 g, total fatty acids average ranged from 2.18 to 30.4 g/100 g, dark chocolate contains the highest fatty and total fatty acid content among them. Stearic acid and palmitic acid were the two main saturated fatty acids(SFA) in chocolate sample, and the average saturated fatty acid content ranging from 1.29 to 19.4 g/100 g, representing the highest proportion(59.2% to 75.3%). The samples contained abundant monounsaturated fatty acids(MUFA). The total content of monounsaturated fatty acids averaged 0.743 to 9.76 g/100 g, second only to saturated fatty acids, accounting for 21.4% to 34.2% of the total, mainly oleic acid, which ranged from 0.727 to 9.63 g/100 g. Polyunsaturated fatty acid(PUFA) average ranged from 0.130 to 1.48 g/100 g, accounting for 3.0% to 6.0% of the total. Linoleic acid was the main polyunsaturated fatty acids in the samples, which ranged from 0.122 to 1.12 g/100 g. The average n-6PUFA/n-3PUFA ratio in the samples was 10. The average IA of the samples was 0.4, and IT was 3.2. No glucose or fructose was detected in the sample. Using convenient sampling method, 350 child healthcare workers with experience in infant and young child feeding counselling from Beijing community health service centers were selected. Following presentations detailing the design and usage of the complementary food simulators through slides and videos, along with demonstration of the actual simulators, electronic questionnaires were distributed to gather their feedback.</p><p><strong>Results: </strong>Approximately 90% of the healthcare workers rated the simulator as good in terms of its usefulness, acceptability, interactivity, willingness to use, and overall evaluation. Among the 329 workers expressing willingness to utilize the simulators, nearly 90% believed that it could enhance the effectiveness of training and feeding counselling. {"title":"[Expression of HSP27, JAK2 and STAT3 apoptosis factors in silicotic fibrosis rat model].","authors":"Yi He, Xin Chang, Hanhan Wu, Wenli Li, Lu Liu, Wenxin Guo, Ying Liu, Liyan Zhu, Yanan Xiong, Haijing Deng","doi":"10.19813/j.cnki.weishengyanjiu.2025.04.020","DOIUrl":"https://doi.org/10.19813/j.cnki.weishengyanjiu.2025.04.020","url":null,"abstract":"<p><strong>Objective: </strong>To explore the relationship between heat shock protein 27(HSP27) and Janus kinase 2(JAK2), Recombinant signal transducer and activator of transcription 3(STAT3) related factors, To further explore the mechanism of silicosis fibrosis and potential therapeutic targets.</p><p><strong>Methods: </strong>The gene expression profile microarray(GSE110147) of a patient with pulmonary fibrosis was obtained from the gene Expression Omnibus(GEO) database, and the HSP27 high expression was set and the genes related to apoptosis pathway were screened out. The STRING online database was used to construct the key gene target map, and the protein interaction network(PPI) was constructed by Cytoscape. The correlation between HSP27 and JAK2, HSP27 and STAT3 was analyzed. The silicosis model was established by one-time instillation of silica(SiO_2) suspension through oropharyngeal endotracheal intubation. Twenty SPF healthy adult Wistar male rats aged 8 weeks were randomly divided into 4 groups, with 5 rats in each group. Silicosis model group for 6 weeks(feeding for 6 weeks) and silicosis model group for 8 weeks(feeding for 8 weeks): oropharyngeal intratracheally instilled 50 mg/mL SiO_2 suspension 1.0 mL per animal. Model control group for 6 weeks(feeding for 6 weeks) and model control group for 8 weeks(feeding for 8 weeks): saline 1.0 mL/animal was instilled into oropharynx and trachea. The pathological changes of lung tissue were observed. The silicon nodule samples were subjected to immunofluorescence assay, and the quantitative analysis of HSP27 and JAK2 apoptosis protein was performed by Western blot.</p><p><strong>Results: </strong>The correlation coefficient between HSP27 and STAT3, JAK2 was r=-0.815, P&lt;0.01, and the correlation coefficient between HSP27 and STAT3 was r=-0.817, P&lt;0.01, HSP27 and JAK2, STAT3 had a negative correlation. After the rats were exposed to dust, the silicosis model groups of 6 weeks and 8 weeks had significantly increased silicotic nodules in the lung. The result of immunofluorescence staining showed that HSP27, JAK2 and STAT3 were co-expressed in the fibrotic area. Compared with the model control group of 6 weeks and 8 weeks, the expression of HSP27, JAK2 and STAT3 in the fibrotic area of the silicosis model group of 6 weeks and 8 weeks increased, and the difference was statistically significant(With the extension of modeling time, the expression of HSP27 protein in the silicosis model 8 weeks group was higher than that in the silicosis model 6 weeks group(P&lt;0.05). The expression of JAK2 protein in the 8-week silicosis model group was lower than that in the 6-week silicosis model group(P=0.32), and the expression of STAT3 protein was lower than that in the 6-week silicosis model group(P&lt;0.05).</p><p><strong>Conclusion: </strong>HSP27, JAK2 and STAT3 are highly expressed in the silicotic nodules of the rat model of silicosis. According to east-west-central regional stratification, Jiangsu, Fujian(eastern region), Shanxi(central region), Chongqing and Guangxi Zhuang Autonomous Region(western region) were included into the study, with 2 follow-up sites of each province. Dietary intake, physical status, psychological behaviors, physical activities, lifestyle, nutrition-related diseases, muscle and cognitive function of the elderly aged 65 and above were collected through questionnaire, physical measurements, dietary surveys and laboratory tests. The project was implemented through a scientific research design, standardized survey procedures and full-process quality control, relying on Centers for Disease Control and Prevention System. A total of 10 659 participants were enrolled. Participants with complete dietary, biochemical, and anthropometric data were included. Blood pressure was measured three times using a mercury sphygmomanometer. Fasting venous blood samples were collected to quantify nine inflammatory biomarkers: hypersensitive C-reactive protein(hs-CRP, immunoturbidimetric assay), interleukin-1beta(IL-1beta), IL-2, IL-4, IL-6, IL-8, IL-10, IL-18, and tumor necrosis factor-α(TNF-α, ELISA). Multivariable logistic regression models adjusted for age, sex, income, residence, education, dietary approaches to stop hypertension(DASH) score, physical activity, body mass index, smoking, alcohol use, and diabetes status were used to evaluate associations. Restricted cubic spline curves explored dose-response relationships.</p><p><strong>Results: </strong>Among 1960 participants, 806(41.1%) had hypertension. After full adjustment, log-transformed inflammatory biomarkers showed significant positive associations with hypertension prevalence: hs-CRP(OR=1.50, 95% CI 1.22-1.86), IL-18(OR=1.98, 95%CI 1.48-2.65), TNF-α(OR=1.84, 95% CI 1.21-2.81), and IL-8(OR=1.23, 95%CI 1.02-1.49). Restricted cubic spline analysis revealed a nonlinear relationship for hs-CRP(P for nonlinear=0.007): hypertension possibility increased rapidly when ln(hs-CRP+1)&lt;0.56, plateaued at 0.56-1.302, and rose again above 1.302. Linear trends were observed for lnIL-18, lnIL-8, and lnTNF-α.</p><p><strong>Conclusion: </strong>Elevated serum hs-CRP, IL-18, IL-8, and TNF-α levels are independently associated with hypertension prevalence in Chinese adults, with a nonlinear dose-response pattern for hs-CRP. {"title":"[Determination of nine markers of tobacco exposure in urine by supported liquid extraction-liquid chromatography-mass spectrometry].","authors":"Yongjie Yang, Guannian Liu, Yifu Lu, Changming Ding, Xiaoming Shi","doi":"10.19813/j.cnki.weishengyanjiu.2025.04.018","DOIUrl":"https://doi.org/10.19813/j.cnki.weishengyanjiu.2025.04.018","url":null,"abstract":"<p><strong>Objective: </strong>To establish a liquid chromatography-mass spectrometry method for the simultaneous determination of nine tobacco exposure biomarkers in human urine, including: nicotine, cotinine, trans-3'-hydroxycotinine, nicotine nitrogen oxide, cotinine nitrogen oxide, nornicotinine, norcotinine, anatabine, and anabasine.</p><p><strong>Methods: </strong>Urine samples were hydrolyzed by adding β-glucuronidase/arylsulfatase(1000 units/sample) and shaking at 37 ℃ overnight in the dark. Deuterium-labeled internal standards were added, and the samples were extracted using a 96-well supported liquid extraction plate. The plate was eluted with 1.8 mL of isopropanol-dichloromethane solution(5∶95, V/V) and dried under a stream of nitrogen. The residues were reconstituted in pure water for analysis. A Gemini® 3 μm NX-C18 110A(2.0 mm×150 mm, 1.9 μm) column was used for separation, with gradient elution using 0.1%(V/V) ammonia solution and methanol as the mobile phase. The method utilized liquid chromatography-tandem mass spectrometry(LC-MS/MS) with a heated electrospray ionization source, employing parallel reaction monitoring and positive ion mode scanning.</p><p><strong>Results: </strong>The method exhibited correlation coefficients(r) for the standard curves of the nine tobacco exposure markers ranging from 0.9984 to 0.9999. The detection limits were between 0.02 and 1.40 ng/mL, and the quantitation limits were between 0.06 and 4.60 ng/mL. The matrix effect ranged from 85.9% to 109%.
