The aim of this research was to study the ability of S. aureus to form biofilms. Additionally, we examined the genes involved in cell resistance and sensitivity to antibiotics. Samples were collected from December 2020 to May 2021 from Simmental and black-and-white cows. The study was carried out on a total number of 643 cows, of which 278 (23 %) were in the subclinical mastitis stage. Finally, 64 S. aureus isolates were isolated and identified. The highest level of phenotypic resistance was observed to antibiotics of the tetracycline (tetracycline - 48.4 %, doxycycline - 32.8 %) and <math><mi>β</mi></math> -lactam (ampicillin - 45.3 %, penicillin - 45.3 %) groups. The genes encoding antibiotic resistance were characterized with the polymerase chain reaction method: blaZ in 30 isolates, mecA in 1 isolate, ermC in 15 isolates, aph (3) in 2 isolates, tetK in 19 isolates, tetM in 9 isolates. The tested S. aureus isolates had the ability to form biofilms in 76.6 % ( <math><mrow><mn>49</mn> <mo>/</mo> <mn>64</mn></mrow> </math> ) of cases. Of these, 69.4 % were resistant to at least one antibiotic. The obtained results have shown that S. aureus, identified in cows with subclinical mastitis, was resistant mainly to tetracycline and <math><mi>β</mi></math> -lactam antibiotics. In addition, S. aureus isolates expressed resistance genes to the above drugs and had the ability to form biofilm. Subsequently, the detected variants were correlated with important wool traits. The traits included greasy fleece weight (GFW, g), staple length (SL, cm), prickle factor (PF, %), medullated fiber (MF, %), fiber diameter (FD, <math><mrow><mi>µ</mi></mrow> </math> m), crimp percentage (CR, %) and the standard deviation of FD (SD <math><msub><mi></mi> <mi>fd</mi></msub> </math> , <math><mrow><mi>µ</mi></mrow> </math> m), as well as the subjectively assessed traits of kemp score (KS), handle grade (HG), greasy color grade (GCG), bulk grade (BG), luster grade (LG) and staple structure (SST). Animals were genotyped by polymerase chain reaction (PCR) - single strand conformation polymorphism (SSCP). Five SSCP banding patterns representing three different nucleotide variants (A, B and C) were detected. DNA sequencing confirmed three single nucleotide polymorphisms (SNPs). Animal age significantly affected GFW ( <math><mrow><mi>P</mi> <mo>=</mo> <mn>0.007</mn></mrow> </math> ), SD <math><msub><mi></mi> <mi>fd</mi></msub> </math> ( <math><mrow><mi>P</mi> <mo>=</mo> <mn>0.006</mn></mrow> </math> ), SL ( <math><mrow><mi>P</mi> <mo>=</mo> <mn>0.002</mn></mrow> </math> ), CR ( <math><mrow><mi>P</mi> <mo>=</mo> <mn>0.006</mn></mrow> </math> ), KS ( <math><mrow><mi>P</mi> <mo>=</mo> <mn>0.001</mn></mrow> </math> ), LG ( <math><mrow><mi>P</mi> <mo>=</mo> <mn>0.006</mn></mrow> </math> ) and SST ( <math><mrow><mi>P</mi> <mo>=</mo> <mn>0.013</mn></mrow> </math> ). Nevertheless, little is known about the association of the polymorphisms of these genes with litter size (LS). In this study, the selected single-nucleotide polymorphisms (SNPs) were genotyped using a Sequenom MassARRAY® platform, and the distribution of different genotypes of the SNPs in the seven sheep breeds (Small-tailed Han, Hu, Cele Black, Suffolk, Tan, Prairie Tibetan, and Sunite sheep) were analyzed. The reliability of the estimated allele frequency for all seven SNPs was at least 0.9545. Given the association of the TGIF1 g.37866222C  <math><mo>></mo></math>  T polymorphism with LS in Small-tailed Han sheep ( <math><mrow><mi>p</mi> <mo><</mo> <mn>0.05</mn></mrow> </math> ), fecundity differences might be caused by the change in amino acid from proline (Pro) to serine (Ser), which has an impact on secondary, tertiary protein structures with concomitant TGIF1 functionality changes. Animals were evaluated monthly from the pre-mating period (non-pregnant), during pregnancy, and from 30 to 105 d of lactation. The effects of single or twin births and the kid's sex were also considered. Serum E <math><msub><mi></mi> <mn>2</mn></msub> </math> , P <math><msub><mi></mi> <mn>4</mn></msub> </math> , and cortisol concentrations were measured using immunoenzymatic assay kits. The highest concentrations of E <math><msub><mi></mi> <mn>2</mn></msub> </math> and P <math><msub><mi></mi> <mn>4</mn></msub> </math> ( <math><mrow><mi>P</mi> <mo><</mo> <mn>0.0001</mn></mrow> </math> ) were found during pregnancy and their lowest values ( <math><mrow><mi>P</mi> <mo><</mo> <mn>0.0001</mn></mrow> </math> ) in the non-pregnant period. E <math><msub><mi></mi> <mn>2</mn></msub> </math> was negatively correlated with P <math><msub><mi></mi> <mn>4</mn></msub> </math> ( <math><mrow><mi>r</mi> <mo>=</mo> <mo>-</mo> <mn>0.41</mn></mrow> </math> ; <math><mrow><mi>P</mi> <mo><</mo> <mn>0.01</mn></mrow> </math> ) during lactation. The mothers with twin kids showed the highest concentration of P <math><msub><mi></mi> <mn>4</mn></msub> </math> ( <math><mrow><mi>P</mi> <mo><</mo> <mn>0.04</mn></mrow> </math> ) at <math><mo>></mo></math>  95-115 d of gestation and the lowest of E <math><msub><mi></mi> <mn>2</mn></msub> </math> ( <math><mrow><mi>P</mi> <mo><</mo> <mn>0.04</mn></mrow> </math> ) at <math><mo>></mo></math>  50-70 d of lactation. Pregnant goats carrying male kid(s) presented the highest E <math><msub><mi></mi> <mn>2</mn></msub> </math> concentrations ( <math><mrow><mi>P</mi> <mo><</mo> <mn>0.02</mn></mrow> </math> ) at <math><mo>></mo></math>  130-150 d of gestation. Different physiological conditions induced a temporal relationship with the endocrine profile in Nicastrese goats. Understanding the effects of single or twin fetuses on the gestation and lactation will also be helpful to improve the managemental approach for the health of mothers and their kids.
","PeriodicalId":55481,"journal":{"name":"Archiv Fur Tierzucht-Archives of Animal Breeding","volume":"64 2","pages":"467-474"},"PeriodicalIF":0.0,"publicationDate":"2021-12-09","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8672451/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"39852989","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":3,"RegionCategory":"农林科学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
