In this study, the expression of ZNF33B in ovary and early embryo was determined by immunohistochemistry and immunofluorescence techniques. Results showed that the expression of ZNF33B in the ovary was mainly in the cytoplasm of oocytes and granulosa luteal cells of ovarian corpus luteum and significantly reduced during follicular ovulation to luteal degeneration. The expression of ZNF33B in the early embryo transferred from the nucleus to the whole cell, suggesting that the expression of ZNF33B is spatiotemporally specific. Then, in combination with the single nucleotide polymorphism (SNP) database, the g.-61G > T mutant of the 5 ′ -untranslated region (5 ′ UTR) of the ZNF33B gene was screened out from 556 Changbaishan black cattle, and the frequency of the mutant gene was counted. The statistics of superovulation and superovulation traits confirmed significant differences between the two genotypes in the quantity and quality of oocytes obtained after superovulation. Alfalfa silage was used as standard forage for comparison. Virginia fanpetals samples were harvested in the bud-formation stage and alfalfa samples were harvested in the late bud stage. Virginia fanpetals silage had a crude protein (CP) content of 176 g kg -1 dry matter (DM), a neutral detergent fiber (NDF) content of 378 g kg -1 DM, and a lignin content of 42.8 g kg -1 DM. Virginia fanpetals silage had higher acidity (pH of 4.30) and was characterized by intense lactic acid fermentation compared with alfalfa silage (80 % vs. 51 % of the total acids). The digestibility coefficient of Virginia fanpetals silage was as follows: for DM it was 0.707, for organic matter (OM) it was 0.724, for CP it was 0.861, and for NDF it was 0.609. In comparison with alfalfa silage, Virginia fanpetals silage was characterized by higher apparent digestibility of nutrients, but a significant difference was noted only for CP. The voluntary intake of Virginia fanpetals silage was significantly higher than that of alfalfa silage (1427.4 vs. 954 g DM). The greatest differences in voluntary intake were observed 0–2 and 8–12 h after feeding. Virginia fanpetals silage had a chemical composition similar to that of alfalfa, but it was characterized by a more desirable fermentation pattern and higher digestibility, and it was more willingly consumed by rams. They can function as “miRNA sponges” in regulating the expression of their target genes. It was thought that circRNAs are involved in the development of the secondary hair follicle (SHF) in cashmere goats. In our previous investigation, a new circRNA named circRNA-0100 was identified from the SHF of cashmere goats, but its function is unknown. In this work, we found that circRNA-0100 exhibited significantly higher expression at anagen SHF bulge than its counterpart at telogen in cashmere goats. Based on the use of both overexpression and siRNA interference assays, our data indicated that circRNA-0100 promoted the differentiation of cashmere goat SHF stem cells (SHF-SCs) into hair follicle lineage, which was evaluated by analyzing the transcriptional level changes of six indicator genes in SHF-SCs of cashmere goats. Using the RNA pull-down technique, we showed that circRNA-0100 served as “molecular sponges” of miR-153-3p in SHF-SCs. Through the use of dual-luciferase reporter assays, our data indicated that circRNA-0100 positively regulated the transcriptional expression of the KLF5 gene via the miR-153-3p-mediated pathway. Ultimately, we showed that circRNA-0100 promoted the differentiation of SHF-SCs into hair lineage, which might be achieved via sequestering miR-153-3p to heighten the KLF5 expression in SHF-SCs of cashmere goats. Kamoun","doi":"10.5194/aab-65-79-2022","DOIUrl":"https://doi.org/10.5194/aab-65-79-2022","url":null,"abstract":"Abstract Olive mill waste is low-quality feed and rarely used in ruminant nutrition because of its high lignocellulose content, the existence of anti-nutritional factors such as total polyphenol and condensed tannin, and low protein contents. This in vitro research was conducted to valorize this waste (crude olive cake, extracted olive cake, and olive leaves) using an exogenous fibrolytic enzyme produced from Trichoderma longibrachiatum in ruminal nutrition. The enzymatic activity of this additive was 1161 units of endoglucanase per millilitre, 113 units of exoglucanase per millilitre, and 2267 units of xylanases per millilitre. This treatment was applied by spraying substrates with four doses: 0 (control), 1 (low), 2 (medium), and 4 µLg-1 µLg-1 (high) of dry matter olive mill waste in an air-conditioned room at 26  ∘C for 12 h before in vitro incubation. For the crude olive cake, this additive at high doses increased degradation of 14 % of cellulose and 8 % of hemicellulose compared with the control at 12 h before the in vitro incubation. Consequently, it increased dry matter solubility and reduced sugars at this period compared to the control. Upon ruminal incubation, the high dose of exogenous fibrolytic enzyme increased the gas production from the immediately soluble fraction and insoluble fraction, the rate of gas production for the insoluble fraction, the dry matter degradability by 26 %, the organic matter degradability by 24 %, the metabolizable energy value by 28 %, and the microbial crude protein production by 24 % compared with the control. For olive leaves, an exogenous fibrolytic enzyme at medium dosage can also hydrolyse the hemicellulose compound, release fewer sugars, and increase dry matter solubility compared with the control at 12 h before the in vitro incubation. Upon in vitro incubation, the medium dose increased the gas production from immediately soluble and insoluble fractions, the rate of gas production for the insoluble fraction, the dry matter degradability by 13 %, the organic matter degradability by 11 %, the metabolizable energy value by 12 %, and the microbial crude protein production by 12 % compared with the control. However, the highest dose altered the gas production from insoluble fractions and decreased microbial crude protein production by 6 % compared with the control. Under the same conditions, an exogenous fibrolytic enzyme applied to extracted olive cake did not produce any effect in the chemical composition and nutritional value. These results showed clearly that effectiveness of exogenous fibrolytic enzyme varied with incubated waste. Increasing the nutritional value of crude olive cake and olive leaves using an exogenous fibrolytic enzyme can encourage breeders to use this waste as feed at a low cost in animal nutrition. However, these conventional methods have the disadvantage of taking a lot of time and incurring considerable cost. The allele-specific PCR method performs PCR using two primers, and a single nucleotide polymorphism (SNP) genotype can be identified through electrophoresis, saving time and cost. Highly accurate results can be obtained by designing specific primers according to the allele of the SNP under study, utilizing primer binding to a complementary matching sequence. In this study, we established a genotyping system with the AS-PCR technique, using SNPs related to the improvement of the meat quality and meat mass of Korean cattle. Using the PRIMER1 program, we designed specific primers for SNPs located at the 3 <math><msup><mi></mi> <mo>'</mo></msup> </math> end, with one SNP marker in the <i>HSPB1</i> gene related to meat quantity and two SNP markers in the <i>ADH1C</i> and <i>FASN</i> genes related to meat quality in cattle. AS-PCR was performed on 10 Korean cattle using the primers designed with this system, and the genotypes could be identified by the size of the PCR product amplified as a result of electrophoresis. In the case of the <i>HSPB1</i> g.2352T  <math><mo>></mo></math>  C SNP, the T allele was amplified to 148 bp, and the C allele was amplified to 222 bp. The <i>ADH1C</i> c.-64T  <math><mo>></mo></math>  C SNP was amplified to 492 bp at the T allele and 330 bp at the C allele. The <i>FASN</i> g.17924G  <math><mo>></mo></math>  A SNP A allele was amplified to 377 bp and the G allele to 507 bp. The results for each SNP genotype were verified using direct sequencing, which showed that the genotypes identified by direct sequencing and the genotypes identified by the AS-PCR method matched exactly. The functional hypothesis of milk exosomal miRNAs and their potential bioavailability in milk to newborn mammals were investigated. Briefly, 37 exosomal miRNAs were upregulated compared to miRNAs found outside the exosomes. Among these miRNAs, ssc-miR-193a-3p expression was upregulated 1467.35 times, while ssc-miR-423-5p, ssc-miR-551a, ssc-miR-138, ssc-miR-1 and ssc-miR-124a were highly concentrated and upregulated 13.58-30.06 times. Moreover, these miRNAs appeared to be relevant for cell development and basic physiological processes of the immune system. Following the analysis of target gene prediction and related signalling pathways, 9262 target genes were mainly concentrated in three signalling pathways: metabolic pathways, pathways in cancer, and phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signalling pathways. Among 9262 target genes, more than 20 miRNAs were enriched in exosomes, such as methyl CpG binding protein 2 (MECP2) and glycogen synthase 1 (GYS1). After determining the miRNA localization-, distribution- and function-related metabolism, we found that these exosomes were specifically concentrated miRNA target genes and they were interrelated with cell development and basic cell functions, such as metabolism and immunity. If the environmental (microclimate) conditions and rearing management are not properly managed, stocking broilers at a high density may, however, result in stressful conditions that are harmful for the production, health and welfare of broilers. To ameliorate these unfavorable effects of overcrowding stress, dietary interventions have been conducted. Probiotics, prebiotics, synbiotics, plant-derived products, vitamins, propolis, amino acids, fatty acids, etc. have been supplemented in diets to deal with the harmful impact of stress induced by a high stocking density of broilers. The results could lay a theoretical basis for understanding the molecular mechanism that regulates hair follicle development. The skin of Aohan fine-wool sheep at different developmental stages (embryonic day 90, E90d, and 120, E120d, and postnatal day 1, B1d, and 30, B30d) were selected. Real-time quantitative polymerase chain reaction (RT-qPCR) and immunohistochemistry were used to study the levels of mRNA and proteins, respectively. The RT-qPCR results showed that the mRNA expression level of EPHA4 at B1d was significantly lower than at E120d ( <math><mrow><mi>p</mi> <mi><</mi> <mn>0.01</mn></mrow> </math> ). The expression of Ephrin A3 at E120d was significantly higher than that at E90d and B1d ( <math><mrow><mi>p</mi> <mi><</mi> <mn>0.01</mn></mrow> </math> ). Immunohistochemical detection results showed that the level and localisation of EPHA4 and Ephrin A3 proteins had spatial and temporal specificity. EPHA4 expression in dermal papilla cells might be important for inducing Aohan fine-hair follicle regeneration and for controlling the properties of the hair. Ephrin A3 might play an important role in the redifferentiation of secondary hair follicles and might also be involved in the inhibition of apoptosis-related gene expression in hair follicles. Leptin (LP, OB, obese) is an important hormone that synthesizes mostly from adipose tissue and regulates glucose metabolism and homeostasis. <i>DGAT1</i> and <i>Lep</i> genes are closely related to reproduction, growth, milk yield and composition in water buffalo breeds. This study aimed to identify genetic variation in the <i>DGAT1</i> and <i>Lep</i> gene regions in 150 water buffalo individuals from five different provinces of Turkey using DNA sequencing. A total of 38 nucleotide variations and indels have identified 761 bp long partial intron 2 and exon 3 and 5 <math><msup><mi></mi> <mo>'</mo></msup> </math> UTR regions of the <i>Lep</i> gene in Anatolian water buffalo populations; 422 bp long partial exon 7-9 and exon 8 regions of <i>DGAT1</i> gene were amplified and two mutations were defined in the point of 155 and 275 nucleotide that is three genotypes for S allele and Y allele of <i>DGAT1</i> gene in intron 7 in Anatolian buffalo populations, respectively. Local chickens' reproductive systems will disrupt as a result of the incubation period activity, and they will lay fewer eggs. This study aimed to determine the prolactin gene polymorphism in IPB-D1 hens and its relationship to egg production. The polymorphism of the exon 5 prolactin gene was examined on 112 samples of the IPB-D1 chicken DNA collection from the Division of Animal Genetics and Breeding, Faculty of Animal Sciences, IPB University. By performing the phenol-chloroform method, the genomic DNA was obtained. A polymerase chain reaction (PCR) product with a size of 557 bp was produced as a result of the DNA amplification. Three single-nucleotide sequences were discovered. Three single-nucleotide polymorphisms (SNPs), g.7835A  <math><mi>></mi></math>  G, g.7886A  <math><mi>></mi></math>  T, and g.8052T  <math><mi>></mi></math>  C, were found in exon 5 of the PRL gene. Each mutation was polymorphic and in Hardy-Weinberg equilibrium.
