Blautia sp. AUH-JLD56 (KF374935) is a strictly anaerobic strain previously isolated by our research group from human feces. Under anaerobic conditions, this strain converts arctigenin to 3′-demethylarctigenin (3′-DMAG), reaching a maximum conversion concentration of 3.6 mM. To improve the oxygen tolerance of this wild-type strain, we performed long-term oxygen tolerance domestication and successfully obtained an oxygen-tolerant mutant. Phenotypic analysis showed that the growth of the oxygen-tolerant mutant under aerobic conditions (OD600 nm = 2.37) was slightly lower than that of the wild-type under strictly anaerobic conditions (OD600 nm = 2.82). Compared with the wild-type, the mutant exhibited an accelerated aerobic growth rate and enabled stable conversion of arctigenin. Notably, under aerobic conditions, the mutant achieved a maximum conversion concentration of 8.2 mM, which is significantly higher than the 3.6 mM obtained with the wild-type under anaerobic conditions. This study realizes, for the first time, efficient aerobic bioconversion of arctigenin to 3′-DMAG using an oxygen-tolerant derivative of a strict anaerobe, thereby overcoming the oxygen-dependent limitation of such strains. Our approach provides a new strategy and technical reference for the oxygen tolerance domestication and industrial application of other intestinal strict anaerobes with specific enzymatic functions.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-05-19","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148305619","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Herein, we propose a facile and highly efficient strategy to successfully prepare a defect-rich, highly active nitrogen-doped porous carbon-based electrocatalyst, U-Fe-N-C (Urea-assisted synthesized iron-nitrogen-carbon material), via a high-temperature co-pyrolysis treatment of heme in the presence of urea. The study demonstrates that urea not only acts as an excellent nitrogen source during pyrolysis, introducing abundant topological defects and heteroatom doping sites, but also prompts the carbon substrate to form a hierarchical sponge-like porous structure with a high specific surface area. This unique microenvironment effectively prevents the agglomeration of iron species at high temperatures, achieving efficient anchoring and high dispersion of catalytic active centers. Electrochemical tests indicate that under optimal synthesis conditions (precursor mass ratio of 1:3, calcination at 900 °C), U-Fe-N-C exhibits outstanding oxygen reduction reaction (ORR) catalytic activity (with a half-wave potential reaching 0.731 V vs. RHE) and possesses long-term durability far exceeding that of commercial Pt/C. Furthermore, liquid rechargeable zinc-air batteries assembled with U-Fe-N-C as the air cathode demonstrate exceptional stability, achieving up to 270 h of charge-discharge cycling without attenuation. This study not only provides profound insights into the mechanisms of pore formation and assistance but also offers a novel perspective for the rational design and scalable synthesis of high-performance metal-nitrogen-carbon (M-N-C) electrocatalysts.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-05-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147967090","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In this study, raisin polyphenols were purified with D101 macroporous resin, and the adsorption process followed the pseudo-first-order kinetic model and the Langmuir isotherm model, indicating monolayer, exothermic adsorption. UHPLC-MS/MS analysis of the purified raisin polyphenols (PRP) identified 10 phenolic compounds, with ferulic acid, quercetin, and isoquercetin as the major constituents. The inhibitory mechanism of PRP against porcine pancreatic α-amylase (PPA) was systematically investigated using inhibition kinetics, fluorescence spectroscopy, circular dichroism (CD) spectroscopy, molecular docking, and molecular dynamics (MD) simulations. PRP exhibited strong inhibitory activity (IC50=3.15±0.16 mg/mL) and acted as a mixed-type inhibitor, whereas the major monomers showed distinct inhibition behaviors. Fluorescence and CD analyses indicated that PRP and its monomers altered the microenvironment around tryptophan and tyrosine residues, induced secondary-structure rearrangement in PPA, and reduced enzymatic activity. MD simulations confirmed the stability of the polyphenol-PPA complexes. Binding free-energy calculations showed that quercetin had the most favorable overall binding affinity and complex stability among the tested monomers, driven mainly by hydrogen bonding and hydrophobic interactions. These findings clarify the α-amylase inhibitory mechanisms of PRP and its key constituents and provide a theoretical basis for developing natural hypoglycemic functional ingredients and plant-derived α-amylase inhibitors.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-04-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148305704","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Herein, we report a nonanuclear Pd-based coordination cage of Pd6-TMPP(Ni), constructed from Ni-chelated TMPP(Ni) as the metalloligand and Pd2+ ions (H2TMPP = meso-tetrakis (6-methylpyridin-3-yl) porphyrin). Pd6-TMPP(Ni) integrates dual ROS-generation for cancer therapy, viz., hydroxyl radical (•OH) production and photoinduced singlet oxygen (1O2) generation. In vitro cytotoxicity assays against cancer cell lines NCI-H82 (lung cancer), A549 (lung cancer), KYSE-510 (esophageal cancer), and Te-1 (esophageal cancer), reveal its potent dark toxicity (IC₅₀: 1.867–2.107 μmol L−1) and phototoxicity (IC₅₀: 0.835–1.528 μmol L−1), which is attributed to enhanced intracellular ROS accumulation. This work develops a versatile therapeutic platform that harnesses Ni-induced •OH for chemodynamic therapy (CDT) and porphyrin-generated ¹O₂ for photodynamic therapy (PDT), thereby mitigating the oxygen dependence of conventional PDT.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-04-21","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147877304","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

RJ proteins were analyzed for ACE inhibition potential using in silico tools, and suitable enzymes were selected for peptide release. Hydrolysis conditions were optimized using response surface methodology (RSM), and the resulting peptides were fractionated and purified. Mass spectrometry identified 57 peptides, with seven selected for synthesis based on scoring. IDFDF, DVNFR, and SFHRL showed the highest ACE inhibition, with IC50 values of 16.9 μM, 42.5 μM, and 242.6 μM, respectively. Lineweaver-Burk plots revealed IDFDF as a competitive inhibitor, DVNFR as a non-competitive inhibitor, and SFHRL as a mixed inhibitor. Molecular docking indicated that peptide-ACE interactions were primarily mediated through hydrogen bonds and Zn(II) coordination. These findings support the high-value utilization of RJ and the development of food-derived ACE inhibitory peptides.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-03-18","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.preprints.org/frontend/manuscript/9c084148c32c61c46d72faf8d23c9eff/download_pub","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147877772","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, such models are at risk of unintended replication, raising concerns regarding originality and intellectual property. While training-time mitigation strategies can address this issue, they typically require retraining or curated datasets, limiting their practicality for largescale systems. Inference-time methods provide a more lightweight alternative but often involve a trade-off between fidelity and memorization risk. This work introduces Repulsive Guidance (RG), a systematic inference-time mitigation strategy that reduces memorization without disrupting the intended conditional guidance from the text prompt. RG operates by enforcing divergence between dual diffusion trajectories through a repulsive term applied only during early denoising steps, without reversing the conditional guidance from the prompt. Experiments on MusicBench with the TANGO model demonstrate that RG offers a complementary mitigation strategy, providing new insights into balancing fidelity and memorization risk.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-03-12","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.preprints.org/frontend/manuscript/87e135ace53b4a1e938dcc6dcf868dba/download_pub","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147887746","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Utiliz-ing cooperative game theory, the research models strategic collaboration between apart-ment residents (demand side) and utility operators (plant side) to maximize energy effi-ciency and economic returns. The VPP structure is analysed over a 15-year life cycle, in-corporating net present value (NPV), payback period (PBP), and government subsidy im-pacts. A cooperative game framework is applied using the Shapley value to ensure fair profit allocation based on each party’s contribution. Results indicate improved self-sufficiency, peak load reduction, and mutual financial benefits. Scenario analyses show that government subsidies to the plant side significantly increase the likelihood of successful cooperation, while declining DER costs enhance the VPP’s economic viability. The findings demonstrate that apartments configured as VPPs achieve strong economic viability (39% ROI, 10.5-year payback) and operational performance (70% self-sufficiency, 40% peak reduction) when grid arbitrage is enabled and moderate government subsidies (35% PV, 45% BESS) are provided. This research provides a replicable model for urban en-ergy planning and policy development, promoting sustainable energy transitions through shared DER infrastructure and cooperative stakeholder engagement.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-03-03","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.preprints.org/frontend/manuscript/85d541f8884b9fd2023038c1ab65f11f/download_pub","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147921456","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Pancreatic lipase (PL) is an important target for obesity management as it plays a key role in lipid absorption. However, the clinical application of synthetic PL inhibitors is limited by adverse effects. Previous studies showed that natural products from traditional Chinese medicines (TCMs) is a promising alternative for developing safe PL inhibitors. In this study, we integrated molecular docking, molecular dynamics (MD) simulations, MM/PBSA binding free energy calculations and in vitro enzymatic assays to systematically screen potential PL inhibitors from TCM constituents. Six compounds with docking scores ranging from −9.9 to −9.0 kcal/mol. MD simulations confirmed the structural stability of six ligand−PL complexes via RMSD, radius of gyration (Rg), hydrogen bond and conformational analysis. MM/PBSA calculations revealed the binding free energies of the six compounds to PL ranged from −21.38±0.40 to −13.33±0.58 kcal/mol. In vitro validation showed five of the six compounds exhibited PL inhibitory activity, among which Hydroxygenkwanin (HYD) was the most potent competitive inhibitor (IC₅₀= 0.128±0.009 mM), followed by Atractylenolide I (ATR-I) (IC50 = 0.584±0.031 mM) and Peiminine (PEI) (IC50 = 0.748±0.042 mM). This study validates the efficiency of the integrated in silico and in vitro strategy for screening natural PL inhibitors and provide valuable reference for the development of novel anti-obesity agents or functional food ingredients.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-02-26","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.preprints.org/frontend/manuscript/aaaf40ed4d1745d57c59b8beb89f53e7/download_pub","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147381606","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Nickel oxide deposited silica (SiO2@NiO) prepared by depositing nickel oxide onto silica using liquid phase deposition method was employed as solid-phase extraction (SPE) adsorbent for the extraction of benzo[a]pyrene (BaP) in edible oil followed by high performance liquid chromatography-diode array detector (HPLC-DAD) analysis. The edible oil was diluted with n-hexane and then directly loaded to SiO2@NiO for SPE. The n-hexane was also used to clean the fat-soluble interference in the edible oil, while BaP was selectively captured due to the electron donor-acceptor interaction with SiO2@NiO. The extraction conditions such as amount of sorbent, volume of washing solvent, type and volume of desorption solvent were optimized. The method demonstrated good linearity over the range of 6-1875 ng/g with the limit of detection of 1.3 ng/g, the spiked recoveries in the range of 97.4-105.1 %, and the relative standard deviation (RSD) less than 3.0 %. This study investigates FVW governance by assessing awareness levels and examining FVW management practices among formal and informal fruit and vegetable retailers in Region F of the City of Johannesburg Metropolitan Municipality (COJMM). A quantitative, descriptive design was employed. Data were collected using structured questionnaires that assessed demographic details, awareness of FVW, and current FVW management practices, and analysed using descriptive statistics in SPSS version 30.0. The findings revealed fragmented governance across retail sectors, characterized by limited awareness of municipal waste management by-laws, consistent dependence on disposal-centred practices, and a lack of adoption of FVW valorization strategies. Formal retailers displayed higher awareness, with access to FVW minimization training, but still mainly relied on disposal, whereas informal retailers displayed significant gaps in awareness and FVW training. The study concludes that unsustainable FVW management is mainly influenced by structural governance limitations, emphasizing the need for inclusive and integrated approaches to improve urban FVW governance in LMIC.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-02-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.preprints.org/frontend/manuscript/be71783cb33ec4631ac23f5bb8f6d3dd/download_pub","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147881473","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
