To dissect the genetic basis of grain vitamin E content in rice, we performed a genomewide association study (GWAS) on a diverse panel of 533 rice accessions. A major locus, VEQ2, was identified on chromosome 2, where the CC genotype significantly increased αtocopherol content compared with the TT genotype. We developed a kompetitive allelespecific PCR (KASP) marker for VEQ2 and validated its effect in an F₂ population derived from Baikezaohe (TT) and AKITAKOMACHI (CC), confirming semidominant inheritance. Through markerassisted backcrossing, we introgressed VEQ2 into an elite indica background, generating nearisogenic lines. The CC introgression lines exhibited α-tocopherol and α-tocotrienol levels that were approximately 1.86- and 5.28-fold those of the control, respectively, accompanied by reduced β-tocopherol, whereas the TT lines displayed the opposite metabolic profile, with no significant penalties in agronomic traits. Analysis of 4,726 accessions revealed strong subspecies differentiation: TT was predominant in indica (84.2%), whereas CC was nearly fixed in japonica (99.5%). Collectively, these findings demonstrate that VEQ2 serves as a key genetic switch modulating the rice vitamin E metabolic profile and represents a promising target for biofortification of indica rice without compromising yield.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-08-25","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148861245","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The high soluble oxalate content of star fruit has become a major obstacle to the further expansion of its consumer market. Elucidating the origin and accumulation mechanisms of oxalate from the perspective of star fruit physiology is particularly important, as it provides the theoretical foundation for modern molecular breeding and the development of advanced cultivation and management practices. In this study, we quantified three oxalate-related organic acids in star fruit at two developmental stages and performed an integrated analysis combining targeted metabolomic and transcriptomic data. Our results showed that during the fruit expansion stage, when oxalate accumulation is relatively high, genes encoding several key enzymes involved in the glycolate oxidative degradation pathway and the L-ascorbate oxidative degradation pathway were significantly up-regulated. Our findings suggest a potential physiological role in regulating cell expansion and conversion between glycolate and amino acids for oxalate in star fruit, thereby addressing the knowledge gap regarding the mechanism underlying the progressive increase in oxalate content during fruit growth.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-08-19","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148861244","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Here, a highly sensitive and specific surface-enhanced Raman scattering lateral flow immunochromatographic assay (SERS-LFIA) was developed for simultaneous determination of T-2 toxin and DON on a single test line. Silver-coated petal-like gap-enhanced Raman tags encoded with 4-aminothiophenol or 4-nitrobenzenethiol were conjugated with antibodies against T-2 toxin or DON, respectively. A mixture of T-2 toxin-ovalbumin and DON-ovalbumin coating antigens was immobilized on the test line. After a 15 min assay, Raman signals at 1136 cm−1 and 1333 cm−1 were measured for quantitative detection. The assay produced IC50 values of 0.35 ng mL−1 for T-2 toxin and 0.29 ng mL−1 for DON, with limits of detection of 0.0033 and 0.0067 pg mL−1, respectively. No mutual interference or significant cross-reactivity with aflatoxin B1, ochratoxin A, zearalenone, patulin, or fumonisin B1 was observed. Recoveries from spiked corn and wheat samples ranged from 89.7% to 114.6%, with relative standard deviations below 9.09%. However, whether these benefits can be transmitted from mother to offspring remains unexplored. Purpose: This study investigated whether maternal LYT consumption confers cross-generational protection against metabolic and behavioral disorders induced by perinatal bisphenol A (BPA) exposure. Methods: A mouse model of perinatal BPA exposure (0.03% in diet) was established with or without maternal LYT supplementation (2.5% in diet). Metabolic parameters were assessed through biochemical assays and gene expression analysis (RT-PCR). Energy expenditure and spontaneous activity were monitored using a Comprehensive Lab Animal Monitoring System (CLAMS). Hippocampal proteomic profiling was performed using label-free quantitative proteomics. Results: LYT significantly reduced BPA absorption and enhanced its glucuronidation and excretion. Notably, LYT exhibited bidirectional metabolic regulation, alleviating gestational hyperglycemia in dams while restoring hypoglycemia in offspring, and normalizing underweight and hypolipidemia without disturbing normal physiology. Mechanistically, LYT modulated the SIRT6 (sirtuin 6)/FOXO1 and SIRT6/SREBP1 pathways to enhance gluconeogenesis and lipogenesis. Concurrently, LYT rectified BPA-induced hyperactivity and reduced energy expenditure. Proteomic analysis revealed that LYT partially restores BPA-induced dysregulation of cholesterol metabolism and glutamatergic/GABAergic synaptic pathways, rebalancing excitation-inhibition homeostasis. Conclusions: These findings establish LYT as a promising dietary intervention for counteracting environmental health risks across generations, acting through toxin clearance, bidirectional metabolic regulation, behavioral normalization, and neural excitation-inhibition balance restoration.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-07-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148672211","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Here, a representative nickel cobalt sulfide/layered double hydroxide het-erostructure has been deposited on carbon cloth to construct free-standing electrode for supercapacitor, marked as NCS@NCOH/CC. Benefited from the nanoporous struc-ture and ultrathin nanosheets, especially the enhanced electronic transfer kinetics due to heterostructure design, the electrode presents potential capacitive performance. Specifically, when increasing the current density from 1 to 20 A g-1, the DSC declines from 1339.1 to 1149.9 F g-1, corresponding to a retention of 85.9%, manifesting excep-tional rate performance. When charging and discharging under 10 A g-1 for 10,000 cy-cles, the DSC can maintain 71.3%, illustrating good cycle durability. Moreover, the fabricated NCS@NCOH/CC//AC/NF hybrid capacitor also demonstrates unique cyclic stability of 86.9% energy retention after 10,000 cycles under 5 A g-1. And an energy density of 20.52 Wh kg-1 at 1604.2 W kg-1 can be realized, corresponding to a tenfold increase compared with AC/NF//AC/NF symmetric capacitor. This work proves the feasibility of fabricating high performance transition metal sulfide@ hydroxide hetero-structure electrode by an electrodeposition method.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-07-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148672209","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The four genotypes were two landraces, EPACE 10 and Paulistinha, and two commercial cultivars, Novaera and BR 17 Gurguéia. Water deficit was imposed for eight days, until leaf water potential (Ψa) was close to -2.0 MPa, and then the plants were rehydrated until maturation. On the last day of stress, EPACE 10 and Paulistinha had higher Ψa values than the other two genotypes. When irrigated, landraces had higher grain protein content (GPC) than the commercial cultivars. Under drought, total GPC was reduced only in EPACE 10, while it remained unchanged in BR 17 Gurguéia. In this condition, EPACE 10 exhibited a notably higher grain amino acid content (GAC), with methionine levels peaking. In addition, SDS-PAGE was successfully used to discriminate the genotypes. Under irrigation, the commercial genotypes (BRS Novaera and BR 17 Gurguéia) yielded more than the two landraces. Still, BRS Novaera had lower methionine and cysteine contents. However, under drought, EPACE 10 and BR 17 Gurguéia had the highest yields. In contrast, EPACE 10 had the best amino acid profile, especially for methionine and cysteine, but with slightly lower lysine content. Therefore, EPACE 10 may serve as a potential genotype for improving food security in low-input agriculture, and BR 17 Gurguéia for irrigated farming. However, more studies are needed, especially in the field, to confirm these results.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-06-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://livimagens.sct.embrapa.br/amostras/00076280.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148305620","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Conventional methods are time-consuming, operationally complex, and costly, limiting their use for point-of-care testing. This study reports a flexible, aptamer-based capacitive biosensor that exploits alternating current electrokinetics for ultrafast detection of cortisol in small-volume samples. Aptamers are immobilized via Au-S self-assembly on gold interdigitated electrodes on a PET substrate, and ACEK-induced fluid motion and dielectrophoresis rapidly enrich cortisol at the electrode interface, producing measurable interfacial capacitance changes ΔC/C0. Experimental results demonstrate detection limits of 0.412 ng/mL in PBS and 0.337 ng/mL in artificial sweat, with response times within 1 minute and excellent linear response across 1-1000 ng/mL concentrations. Requiring only 10 μL of sample, the sensor exhibits good repeatability, specificity, and interference resistance, making it suitable for rapid cortisol level detection. To enhance detection stability, this study designed and integrated a microfluidic chip, enabling efficient sample delivery and stable detection. The system demonstrates strong interference resistance, revealing potential applications in health management and disease monitoring.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-06-05","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148305782","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Clinical manifestations of BVD suggest that inflammatory responses are involved in the pathogenesis of BVDV, whereas the underlying molecular mechanisms remain unclear. As a key signaling hub of innate immunity, inflammasomes activate Caspase-1, thereby facilitating the cleavage of Gasdermin D (GSDMD), promoting membrane pore formation and triggering pyroptosis, thereby contributing critically to host antiviral immunity and viral pathogenesis. This study used Madin-Darby bovine kidney (MDBK) cells as a model to compare infection responses between the cytopathic BVDV NADL strain and the non-cytopathic TC strain. The results showed that both strains upregulate the expression of IL-1β and IL-18 and promote the assembly of the NLRP3 inflammasome, indicating activation of upstream inflammasome signaling; however, only the NADL strain markedly induced Gasdermin D (GSDMD) cleavage, membrane pore formation, and pyroptosis. Further mechanistic studies revealed that core protein C (from the NADL strain) interacts with the inflammasome adaptor protein ASC and promotes NLRP3 inflammasome activation, thereby enhancing the secretion of inflammatory cytokines. In summary, this study provides the molecular mechanism by which cytopathic BVDV activates NLRP3 inflammasome-mediated pyroptosis in an ASC-dependent manner through the core protein C, providing new insights into the inflammation-related pathogenic process of BVDV and the differences in infection outcomes of different viral biotypes.","PeriodicalId":521283,"journal":{"name":"Preprints.org","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2026-05-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"148305557","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
