The identification of predictive and interpretable prognostic biomarkers is critical for advancing precision medicine in NB. We proposed an integrative network-based machine learning method for biomarker discovery, which employed a network smoothed t-statistic support vector machine to select prognostic related biomarkers, and then we performed network analysis on these biomarkers to find hub genes. Later, we conducted a comprehensive analysis to integrate bulk and single-cell RNA sequencing data to character the tumor microenvironment of prognostic state and correlated them to the discovered hub genes. This analysis identified 528 prognostic biomarkers associated with NB. Network-based analysis further refined this set to 11 hub prognostic biomarkers for NB: AURKA, BLM, BRCA1, BRCA2, CCNA2, CHEK1, E2F1, MAD2L1, PLK1, RAD51, and RFC3. Among these genes, high RFC3 expression was significantly associated with poor prognosis, highlighting its potential as a novel prognostic biomarker in NB. Additionally, our findings revealed that these biomarkers are correlated to chemotherapy drugs, such as vincristine and cyclophosphamide. Furthermore, drug sensitivity analyses identified several candidate drugs, such as dactinomycin, bortezomib, docetaxel, and sepantronium bromide, that may hold therapeutic potential for NB treatment. This study offers novel insights to underlying NB prognosis and therapeutic targets and provides a foundation for developing personalized treatment strategies to improve clinical outcomes.
","PeriodicalId":520221,"journal":{"name":"Pediatric discovery","volume":"3 2","pages":"e70009"},"PeriodicalIF":0.0,"publicationDate":"2025-05-27","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12258108/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"144645133","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Epidemiological and clinical data were collected and analyzed using statistical methods to understand the disease characteristics. The onset and progression of SARS-CoV-2 infection in children were significantly associated with age distribution, basic illness, vaccination status, exposure history, and family clustering. The most common clinical symptoms were fever (91.7%) and cough (81.6%). Laboratory findings indicated elevated neutrophil count, neutrophil-to-lymphocyte ratio, platelet-to-lymphocyte ratio, prothrombin time, and procalcitonin, creatine kinase, D-dimer, and IL-6 levels, along with decreased lymphocyte count, platelet count, and lymphocyte-to-monocyte ratio. Lung computed tomography (CT) showed imaging changes strongly linked to severe infection. After receiving anti-inflammatory, symptomatic, supportive, and antipathogen therapies, 74% of the children displayed clinical symptomatic relief, 26% of the children were cured and discharged, and there were no fatalities. In Zhuhai, children infected with SARS-CoV-2 commonly exhibit family-based transmission, with fever and cough as the predominant symptoms. Factors, such as young age, basic illness, specific laboratory markers, and patchy exudative changes on lung CT scans, served as critical indicators of severe infection. Early detection and monitoring of these factors, along with timely vaccination against novel coronavirus, can mitigate disease severity and prevent progression.
","PeriodicalId":520221,"journal":{"name":"Pediatric discovery","volume":"3 2","pages":"e70001"},"PeriodicalIF":0.0,"publicationDate":"2025-05-19","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12258080/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"144645137","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Yet, research into its renal protective effects, particularly for cisplatin-induced AKI, is markedly limited. Our study explores the therapeutic potential of PU against AKI triggered by cisplatin administration. Utilizing an AKI model induced by a substantial dose of cisplatin, we assessed renal function through histological examination (hematoxylin and eosin staining) and biochemical markers, including blood urea nitrogen (BUN) and serum creatinine (Scr). Our results demonstrated that PU may mitigate cisplatin's structural damage to renal tissue. Post-treatment with PU, a significant reduction in pro-inflammatory cytokines interleukin-1β (IL-1β), IL-6, and tumor necrosis factor-α (TNF-α) was observed through quantitative polymerase chain reaction (PCR) analysis. Additionally, the expressions of apoptotic and anti-apoptotic proteins B cell leukemia/lymphoma two (Bcl-2), Bcl2-Associated X protein (Bax), and cleaved-caspase3 evaluated via Western blot and immunofluorescence indicated that PU could inhibit apoptosis in response to cisplatin injury. Oxidative stress markers, such as malondialdehyde (MDA) and superoxide dismutase (SOD), showed decreased MDA levels and elevated SOD activity following PU treatment, suggesting a reduction in oxidative damage. Furthermore, cisplatin-induced downregulation of NRF2 and NQO1 was countered by PU, highlighting its restorative effect on antioxidant pathways as confirmed by Western blot and immunofluorescence. Collectively, our findings endorse PU's protective capability against cisplatin-induced AKI, endorsing its therapeutic potential to counteract cisplatin's nephrotoxicity.
","PeriodicalId":520221,"journal":{"name":"Pediatric discovery","volume":"3 4","pages":"e70000"},"PeriodicalIF":0.0,"publicationDate":"2025-05-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12753014/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145879850","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Although it remains known that maternal stress can negatively impact maternal health, the downstream effects of stress on fetal and infant development in the long-term are unclear. This leaves a gap in understanding epigenetic factors that impact growth and development, highlighting the need to address potential developmental delays in early childhood. Review of the literature indicates debate over the effect of maternal stress on cognitive development and whether maternal stress is a mediator or causal factor in fetal development. Although evidence suggests that maternal stress disrupts the functional connectivity of the amygdala, information on the sub-regions of the brain impacted by prenatal stress exposure remains unknown. Studies focusing on environmental disasters and war have also indicated adverse impacts on fetal development, additionally emphasizing a greater need for representation in future research. Timing of fetal exposure to stressors also impacts different domains of development. The Georgia OASIS platform was utilized to investigate variations in reported pregnancies, reported fetal mortality rate, and Perinatal Periods of Risk (PPOR) during the years 2013-2023. Increases in fetal mortality were anticipated to be associated with decreases in the rates of maternal, newborn, and infant care. Although evidence exists to support this hypothesis, causation cannot be established; however, the poor state of maternal healthcare in Georgia warrants further investigation of the barriers to healthcare to improve fetal and infant health outcomes statewide.
","PeriodicalId":520221,"journal":{"name":"Pediatric discovery","volume":"3 3","pages":"e70004"},"PeriodicalIF":0.0,"publicationDate":"2025-05-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12483304/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145208838","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The aim of this study is to explore its clinical presentations and compare the diagnostic accuracy of transthoracic echocardiography (TTE) with computed tomographic angiography (CTA) on DOLV. We took TTE as a first-line examination modality, considering CTA and surgical results as the gold standard. Ten suspected patients with DOLV were identified at the Children's Hospital of Chongqing Medical University from March 2009 to November 2023. The clinical presentations, TTE, CTA, and follow-up data of the children were analyzed retrospectively. All 10 cases (100%) underwent TTE examination, and 7 patients (70%) received CTA examination. Six patients (60%) ultimately underwent surgery and were confirmed to have DOLV. DOLV were initially diagnosed using TTE with a diagnostic accuracy rate of 85.71% after comparisons with CTA and surgery results. Ventricular septal defect (100%) and atrial septal defect (80%) were the common associated abnormality. The clinical manifestation of DOLV was atypical, and TTE has a relatively high diagnostic accuracy for DOLV in pediatrics, making it a very valuable tool for early detection.
","PeriodicalId":520221,"journal":{"name":"Pediatric discovery","volume":"3 4","pages":"e70005"},"PeriodicalIF":0.0,"publicationDate":"2025-05-08","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12753015/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"145879760","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The rise in screen time among children under 5 years old has raised concerns about its association with motor development including gross and fine motor skills. Conflicting evidence on the association of screen time requires more investigation and the planning of targeted interventions. This systematic review aims to explore the relationship between screen time and motor development in children aged 0-7 years, considering various influencing factors like screen type, exposure duration, and context. Following preferred reporting items for systematic reviews and meta-analyses (PRISMA) and assessing the methodological quality of systematic reviews (AMSTAR) guidelines, a literature search was conducted in May 2024 in databases including PubMed, Cochrane Central Register of Controlled Trials (CENTRAL), and ScienceDirect. Eligible studies were observational or experimental, involved children aged 0-7 years, and assessed the outcomes between screen time and motor development. The quality of the included studies was assessed through Joanna Briggs Institute's (JBI) critical appraisal checklist. Out of 1490 records initially identified, 24 studies met the inclusion criteria for this review. Among these, 17 studies reported a significant negative correlation between screen time and motor development in children, while 5 studies found no statistically significant association. Two studies presented mixed findings, indicating both negative and positive associations between screen time and motor development. Excessive screen time in early childhood is mainly linked to negative effects on motor development. The association varies with screen content and environment, highlighting the need for balanced screen time and early interventions.
","PeriodicalId":520221,"journal":{"name":"Pediatric discovery","volume":"3 2","pages":"e70002"},"PeriodicalIF":0.0,"publicationDate":"2025-04-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12258109/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"144645130","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
