Current photoprotective techniques have limitations due\u0000to the amount of topical application, insufficient physical protection, and adverse responses to topical\u0000agents. A newer option that offers a promising defense against solar radiation is systemic agents, the\u0000well-researched Polypodium leucotomos is significant in this category, and several studies have found\u0000to be effective ―oral sunscreenǁ. Polypodium leucotomos (PL) extract has gained significant attention\u0000as a potential therapeutic agent in the field of dermatology, particularly in the photoprotection and\u0000management of various dermatological conditions. This abstract provides a comprehensive overview\u0000of the mechanism, safety, and efficacy of Polypodium leucotomos extract in relation to photoprotection\u0000and dermatological conditions. Our primary aim is to review Polypodium leucotomos' photoprotective\u0000results from studies that are currently available, and our secondary goal is to describe Polypodium\u0000leucotomos' applications in different dermatological conditions. Articles discussing Polypodium\u0000leucotomos uses in photoprotection and dermatologic conditions are searched and downloaded from\u0000publicly available databases such as PubMed, JSTOR, PLOS, Science Direct, DOAJ, Google Scholar,\u0000and Web of Science and evaluate the outcomes of the Polypodium leucotomos effect. Firstly, we\u0000delve into the mechanisms underlying the photoprotective properties of PL extract. Extensive research\u0000has demonstrated that PL extract exerts its protective effects through multiple pathways, including\u0000antioxidant activity, DNA repair stimulation, immunomodulation, and inhibition of inflammatory\u0000mediators. These mechanisms collectively contribute to the prevention of photodamage,\u0000such as sunburn, DNA damage, and photoaging. Furthermore, we explore the safety profile\u0000of PL extract, emphasizing its remarkable tolerability and minimal adverse effects reported in clinical\u0000studies. The absence of systemic absorption and its compatibility with other treatments make PL\u0000extract a promising option for long-term use in photoprotection and dermatological care. A promising\u0000alternative to traditional photoprotection may be Polypodium leucotomos. Additional thorough\u0000clinical trials are necessary to ascertain its role and effectiveness more accurately. The efficacy of PL\u0000extract in managing various dermatological conditions is also discussed. Multiple studies have\u0000demonstrated its beneficial effects in conditions such as vitiligo, melasma, psoriasis, and atopic dermatitis.\u0000The anti-inflammatory and immunomodulatory properties of PL extract have shown promise\u0000in reducing disease severity, improving skin symptoms, and enhancing the quality of life for patients.\u0000Additionally, the abstract highlights the potential future directions and areas of research for PL extract.\u0000Investigating its combination therapy wi

Validated\u0000qualitative and quantitative reversed-phase HPLC analyses of the P. amarus extract were performed\u0000for the determination of lignan and polyphenol contents. Human THP-1-derived macrophages were prepared\u0000by treatment of THP-1 cells with phorbol 12-myristate 13-acetate (PMA). The inhibition of\u0000cytokines released by the extract, lignans and polyphenols in the cells was investigated using ELISA assay.\u0000\u0000\u0000\u0000Chemical profiling of P. amarus was carried out by LC-MS/MS analysis. At a dose of 50 μM, COR exhibited a maximum inhibition of\u000081.11% on TNF-α secretion, while GER showed 72.56% inhibition on IL-1β secretion. COR demonstrated\u0000the strongest inhibition of TNF-α secretion, exhibiting an IC50 value of 9.06 μM, which was\u0000comparable to that of dexamethasone (7.07 μM). Meanwhile, GER was the most potent against IL-\u00001β secretion, exhibiting an IC50 value of 20.09 μM. In the case of TNF-α secretion, the order of potency\u0000observed among the active compounds, with regard to IC50 value, was COR > GER > HYPO >\u0000PHY > NIR > GA > EA >ISO > PHYLT. For IL-1β secretion, the order of potency was GER > NIR\u0000> COR > GA > EA > PHY > HYPO > PHYLT > 1SO.\u0000\u0000\u0000\u0000The polyphenol contents of P. amarus, especially COR and GER, contributed significantly\u0000to the suppression of cytokines secretion, and they have the potential to be developed into\u0000agents for the treatment of pathologic inflammation.\u0000

Huangqi Decoction (HQD) is a Traditional Chinese Medicine (TCM)\u0000effective for treating liver conditions, including cirrhosis. Therefore, both the active ingredients and\u0000the pharmacological actions of HQD deserve further exploration. The active components and pharmacological\u0000actions of HQD in preventing and treating liver cirrhosis were investigated using network\u0000pharmacology. The actions of the principal active ingredient, Mairin, were investigated empirically.\u0000\u0000\u0000\u0000Using network pharmacology, the critical components of HQD were identified from multiple\u0000databases, and UPLC screening and targets were investigated using Swiss Target Prediction.\u0000Targets associated with liver cirrhosis were identified using the GeneCards database. GO and KEGG\u0000enrichment analysis of targets that overlapped between HQD and cirrhosis were analyzed in DAVID,\u0000and a “component-target-pathway” network for HQD was created in Cytoscape 3.7.2. The biological\u0000functions of the key active component, Mairin, were investigated using in silico docking, cell experiments,\u0000and evaluation in a carbon-tetrachloride (CCl4)-induced mouse model of liver cirrhosis.\u0000CCK-8 and F-actin assays were used to measure cell viability and hepatic stellate cell (HSC) activation,\u0000respectively; fibrosis was measured by histological and immunohistochemical evaluations, and\u0000the levels of the cirrhosis-related protein α-SMA and predicted essential target proteins in the PI3KAKT,\u0000NFκB-IκBα, and NLRP3-IL18 pathways were determined by western blotting.\u0000\u0000\u0000\u0000Fourteen active HQD components, 72 targets, and 10 pathways common to HQD and cirrhosis\u0000were identified. Network analysis indicated the association of Mairin with most targets and\u0000with inflammation through the PI3K/Akt, NF-κB, and NLRP3 pathways. Dose-dependent reductions\u0000in the activation and proliferation of LX-2 cells after Mairin treatment were observed. Mairin reversed\u0000the histopathological changes in the livers of cirrhosis model mice. Mairin also significantly\u0000reduced the α-SMA, NF-κB, IκBα, NLRP3, and IL-18 protein levels while increasing those of p-\u0000PI3K and p-Akt, suggesting that Mairin mitigates liver cirrhosis through modulation of the PI3KAKT,\u0000NFκB-IκBα, and NLRP3-IL18 pathways.\u0000\u0000\u0000\u0000Using a comprehensive investigative process involving network pharmacology, bioinformatics,\u0000and experimental verification, it was found that Mairin, an active component of HQD,\u0000may be useful for developing specific treatments for preventing and treating liver cirrhosis.\u0000
