{"title":"Molecular dynamics simulation studies and dynamic network analysis of Bacillus subtilis YsxC in GDP and GTP-Mg 2+ bound states","authors":"Shuchika Devaraj Biligere, Kavya Kallahalli Mohankumar, Upendra Nagarajachari, Krishnaveni Sannathammegowda","doi":"10.34117/bjdv10n5-044","DOIUrl":"https://doi.org/10.34117/bjdv10n5-044","url":null,"abstract":"Ribosomes are essential cellular organelles made up of rRNA and rproteins found in both prokaryotic and eukaryotic cells, and play an important role in protein synthesis. The functional ribosomes are synthesized through the process called ribosome biogenesis, which proceeds with the help of certain factors such as chaperones, ribosomal proteins, and GTPases. GTPases bind to the premature ribosomal subunit and function as a checkpoint, ensuring the proper assembly of other proteins. To aid this process, GTPase undergoes conformational changes, alternating between an active GTP-bound state and an inactive GDP bound state. YsxC is a GTPase that functions this way, to help in the maturation of 50S subunit. Although YsxC plays an important role in ribosome biogenesis, a detailed knowledge of how the GDP and GTPMg2+ states of YsxC assist in the switching process is yet to be realized. Therefore, a study on the GDP and GTP-Mg2+ bound states is done for YsxC GTPase of Bacillus subtilis by all-atom molecular dynamics simulation for a period of 500 ns. The simulations aided the analysis of the RMSD from which it was noticed that both the GDP and GTP-Mg2+ states of YsxC attained equilibration after 200ns. The analysis of the Rg for the systems showed that the GTP-Mg2+ system showed higher values, in comparison to the GDP system, indicating conformational changes in the two systems. Thus, a residue-wise calculation for the difference in the SASA (solvent accessible surface area) of the GTP-Mg2+ and GDP system is done, and it is noticed that the GTP-Mg2+ system, especially in the regions recognized as Switch I and Switch II had a higher difference in SASA value. In which Switch I’s Lys55 (positive residue) and Switch II’s Arg89 (positive residue) showed a distinct difference, indicating that Switch I has more accessibility to solvent. Giving rise to a supposition that the residue Lys55 attracts the negative rRNA and aids in the binding of YsxC with the premature 50S subunit in the GTP-Mg2+ bound state by associating with the rRNA. To fathom the role of the GTP-Mg2+ bound state in increasing the SASA value of Switch I region, a community network is constructed to extract the shortest path. More number of paths are observed between the selected residue in the GTP-Mg2+ bound state inferring that the increased SASA in the Lys55 is due to the stronger connection with the nucleotide. The process starts from the appropriate PVT data, exploitation data, selecting necessary parameters, designing wells, and predicting exploitation to find the best case for the Electric Submersible Pump exploitation plan. The results showed that if oil is only exploited by natural energy, it will not be effective, so it is possible to equip a Electric submersible pump system with 2 exploitation wells for the most efficient exploitation. O controle é realizado através de simulação, usando modelos dos elos obtidos com dados reais do sistema, através de estimação de parâmetros usando o algoritmo dos mínimos quadrados recursivo (MQR). A identificação dos elos é realizada em malha aberta, utilizando dados reais coletados do robo. são  projetados e implementados controladores adaptativos via imposição de pólos para o controle de posição dos dois elos. Os controladores são projetados em malha fechada com base nos modelos obtidos para os elos. a estratégia de controle a ser usada, baseia-se em um modelo explícito do sistema. Aims: There were two objectives in the study. First objective was to examine whether there was a relationship between a teacher’s sense of efficacy and the perceived occupational stress. Second objective was to understand whether a teacher’s years in service had an impact on their resilience. Method: A questionnaire with few demographic questions and three scales which assessed their sense of efficacy, occupational stress and resilience were presented via Qualitrics. Participants were attracted through a social media advert. Approval was granted to conduct the study under reference number UG7496 by the ethics panel of Cardiff School of Sport and Health Sciences. A quantitative research was conducted and the collected data was analysed using SPSS. Results: A total number of 113 responses were collected where 89 responses were used for analysis. Spearman’s correlation tests were carried out, and the computed results indicated a moderate negative relationship between teacher’s sense of efficacy and occupational stress, r=-0.56, n=89, p<.001. Further, a strong positive relationship was indicated between years in service and resilience of a teacher, r=0.70, n=89, p<.001. Conclusions: Both hypotheses which were tested were supported by the obtained results. O Conselho Nacional de Trânsito autoriza que órgãos da segurança pública ministrem o curso especializado de condutor de veículo de emergência para seus efetivos, ressaltando que empresas privadas ofereçam o curso, mas com certeza sem a expertise das peculiaridades atinentes ao serviço policial, ao patrulhamento ostensivo motorizado, que envolve muito mais conhecimento e decisões rápidas que simplesmente dirigir. O rol de disciplinas regulamentares e obrigatórias são semelhantes as aplicadas nos cursos de formação de policiais, o que facilita a compreensão em paralelo com as práticas de direção em situação de risco citada como obrigatória na legislação de trânsito. Entendimentos jurídicos que o policial militar que se envolve em sinistro de trânsito na condução de uma viatura policial está amparado pela excludente de ilicitude do estrito cumprimento do dever legal, assim livre de arcar com custos de ressarcimentos. Anurans are one of the most endangered groups of creatures on earth and perform a critical function in the ecology. The preservation of biodiversity, sustainable development and food security are all major goals of agroforestry. In dispersed agricultural environments, agricultural activities like paddy and rice fields may offer frogs a habitat and aid in their preservation. The long-term impacts of introducing frogs in rice yield and soil Phosphorus availability are largely underestimated and undervalued. Currently, the cultivation of rice paddy creates 11% of the anthropogenic CH4 (Methane) in the atmosphere, which can be regulated through the use of frogs. The frogs and toads are the bio-indicators of the environment. They consume bugs, weeds, and other harmful insects to keep their numbers in check. They also operate as a biological pest management and supply food for other creatures including birds, reptiles, and small mammals. The frog's waste increases the rice crops' access to nutrients and also has a fertilising effect. This study was conducted to characterize Bacillus velezensis DCJ 2, assess its plant growth promotion effect and investigate the effectiveness of different delivery methods of the bacterial inoculum for the growth promotion of Aglaonema ‘Maria’. In this context, DCJ 2, isolated from the rhizosphere soil of A. Maria was screened to investigate its potential as a plant growth-promoting bacterium. The pure culture of DCJ 2 maintained on nutrient agar was utilized for morphological, biochemical and physiological examinations. The molecular characterization of the strain was performed with different primer pairs targeting 16SrRNA, gyrA, pgsB and sugar kinase genes of DCJ 2. A pot experiment was carried out to investigate the plant growth promotion effect. It was conducted in the greenhouse as per the complete randomized block design with eight treatments and five replicates. . Healthy plants of A. ‘Maria’ established in field soil and solarized soil were treated with 100 ml of the suspension of the DCJ 2 at the rate of 10 8 CFU/ml either as a soil drench or foliar spray. The partial 16SrRNA, gyrA, pgsB and sugar kinase gene sequences of DCJ 2, BLAST searched in the NCBI database revealed that it has a high similarity to Bacillus velezensis. The pot experiment showed that the application of strain DCJ 2 as a soil drench to solarized soil (Treatment 6) yielded a significant increase in growth parameters at p=0.05. Based on molecular analysis, morphological, biochemical, and physiological reactions, DCJ 2 was identified as Bacillus velezensis. This study revealed that B. velezensis DCJ 2 has the potential to boost the growth of A. While the increasing presence of best-known fast food chain restaurants worldwide such as McDonald’s and KFC is a manifestation of globalization, the shrewd and strategic adaptations of their menu to satiate the local palates and tastes by these “Glocal Enterprises” is illustrative of glocalization. Conversely, the chief point of glocalization perspective is: “even the most local ways of carrying out business operations are more and more co-constituted by economic and social trends that are most global.” Language has a huge role to play in addressing the cultural and linguistic barriers and designing a successful global talent management and language learning solution that is complemented by local support. When used as an extension of the “Think Global, Act Local” grassroots movement, glocalization also refers to market advantage of local businesses versus global companies.  In the face of globalization, both the little guys and would-be entrepreneurs can emerge triumphant by tapping into their hometown markets and then scaling up.
