The present study investigates the possible protective effect of NSO on CP-induced gastrointestinal toxicity. NSO administration (2</span> <span>ml/kg bwt, orally), prior to and following, a single dose CP treatment (6</span> <span><span>mg/kg bwt. ip), significantly attenuated the CP-induced decrease in brush border membrane (BBM) enzyme activities in intestinal homogenates and BBM vesicles<span> (BBMV). NSO administration also mitigated CP induced alterations in the activities of carbohydrate metabolism enzymes and in the enzymatic and non-enzymatic antioxidant parameters in the intestine. The results suggest that NSO by empowering the endogenous antioxidant system improves intestinal redox and metabolic status and restores BBM integrity in CP treated rats. Histopathological studies supported the biochemical findings. {"title":"Metaplastic phenotype in human primary bronchiolar epithelial cells after repeated exposure to native mainstream smoke at the air-liquid interface","authors":"Michaela Aufderheide , Shigeaki Ito , Shinkichi Ishikawa , Makito Emura","doi":"10.1016/j.etp.2017.01.015","DOIUrl":"10.1016/j.etp.2017.01.015","url":null,"abstract":"<div><p><span>3D constructs composed of primary normal differentiated human bronchiolar epithelial (NHBE) cells as mono- or co-culture in combination with normal human lung fibroblasts were exposed repeatedly at the air-liquid interface with non-lethal concentrations of mainstream cigarette smoke (4 cigarettes a day, 5</span> <span>days/week, 13 times repetition in total) to build up a permanent burden on the cells. Samples were taken after 4, 8 and 13 times of repeated smoke exposure and the cultures were analyzed by histopathological methods In comparison with the clean air exposure (process control) and incubator control cells the cigarette smoke exposed cultures showed a reduction of cilia bearing as well as mucus<span> producing cells. In both mono- as well as co-cultures, hyperplasia<span><span><span> was induced showing different histological cell types (undifferentiated secretory and squamous cell types). At the end of the exposure phase, we observed the development of non-hyperplastic areas strongly positive to CK13 antibody, commonly seen in squamous cells as a marker for non-cornified </span>squamous epithelium, thus suggesting a transition of the normal bronchial epithelial cells towards </span>metaplastic cells. The control cultures (clean air exposed and incubator cells) showed no comparable phenotypic changes. Hypoplastic kidney (HPK) rats have only about 20% of the normal number of nephrons and show progressive CKD. This study used an immunohistological method to assess glomerular and interstitial pathogenesis in male HPK rats aged 35–210</span> <span>days. CD68<span> positive-macrophages were found to infiltrate into glomeruli in HPK rats aged 35 and 70</span></span> <!-->days and to infiltrate into interstitial tissue in rats aged 140 and 210<!--> <!-->days. HPK rats aged 35 and 70<!--> <span><span><span>days showed glomerular hypertrophy, loss of normal linear </span>immunostaining of podocine, and increased expression of PDGFr-β, TGF-β, collagens, and </span>fibronectin<span>, with all of these alterations gradually deteriorating with age. α-SMA-positive myofibroblasts were rarely detected in glomerular tufts, whereas α-SMA-positive glomerular parietal epithelium (GPE) cells were frequently observed along Bowman’s capsular walls. The numbers of PDGFr-β-positive fibroblasts in interstitial tissue were increased in rats aged 35</span></span> <span>days and older, whereas interstitial fibrosis<span>, characterized by the increased expression of tubular PDGF-BB, the appearance of myofibroblasts doubly positive for PDGFr-β and α-SMA, and increased expression of collagens and fibronectin, were observed in rats aged 70 and older. The mass was detected at 50</span> <span>days of age, it grown quickly and the animal was humanely sacrificed eight days later. The tumor was located in the left cervical region, in the vicinity of the left submandibular and sublingual glands<span>. It was soft and reddish and had several dens with a bloody content. The tumor was PAS negative and exhibited immunostaining for ER-α. The histopathologic and immunohistochemical data are suggestive of a high-grade carcinoma from mammary gland.
