Quyen","doi":"10.15625/0866-7160/V40N2.11070","DOIUrl":"https://doi.org/10.15625/0866-7160/V40N2.11070","url":null,"abstract":"The green alga Chlamydomonas reinhardtii is a model organism for studying mechanism of the body in response to changes in environmental conditions, photosynthesis as well as chloroplast biogenesis and can be modified to produce high value proteins. In this study, we present results on the selection of cultural conditions for the growth of cell wall - deficient mutant C. reinhardtii (503 strain- control) and recombinant 503-2 strain expressing VP28 protein of White Spot Syndrome Virus (WSSV) in 250 ml flasks. Suitable conditions for the growth of both control and recombinant strains were TAP medium, temperature of 25 C, pH = 7, light intensity of 2 klux, C source - Trisbase, N source - nitrate and salinity of 50 - 100 mM. In these conditions, maximum cell density of the control and recombinant strains was 3.20 ± 0.08 x 106 and 2.96 ± 0.17 x 106 cells/ mL after 3 and 7 days of cultivation, respectively. The presence of vp28 gene in the recombinant microalgal strain was stably maintained at all levels of culture. Currently, the number of individuals in Peanut worm populations are declining considerably due to over-exploitation and habitat degradation. However, information on individual density and biomass of Peanut worm populations in Vietnam is still lacking, as is information on environmental factors affecting them. This study was carried out at various sites in the lower reaches of mangrove forest of the Gianh river, Quang Binh province. We calculated individual and hole densities per month by counting the number of individuals and holes according to each site area. We found significant differences (p ≤ 0.037) among sampled sites in hole density (from 1.43 to 2.54 holes/m2, average of 1,96 holes/m2), individual number (from 45 to 81 individuals, average of 62 individuals/site area), and biomass (from 20.27 to 51.16 g/m2, average of 34.57 g/m2). However, individual density (from 0.51 to 0.98 individuals/m2, average of 0.68 individuals/m2) was not significantly different among sites. Water temperature, pH, and salinity have the significant effect on individual and hole densities, individual number, and biomass of peanut worms (p ≤ 0.001; excepting water temperature affects the biomass of peanut worms). It is used mainly as a starting material in the biosynthesis of pharmaceuticals as well as employed in the production of crop protection agents, polymers, cosmetics and food additives. Four Gram-negative, moderately halophilic, pyruvate and poly (3-hydroxybutyrate)- producing strains were isolated from soil samples collected from mangrove forest at Giao Thuy, Nam Dinh province. Preliminary screening results showed that among fours strain, the ND7 strain was able to produce the highest bioplastic-poly (3-hydroxybutyrate) (PHB) and extracellular secretion of pyruvate, respectively, reaching 50.23% dry weight and 6.87 g/L after 60 hours of cultivation. When cultured in high alkaliphilic medium (pH 9.4), the pyruvate secretion increased significantly (21.02 g/L after 60 hours of cultivation). Based on the morphological, bio-chemical characteristics and nucleotide sequence analysis of partial 16S rRNA gene, the ND7 strain was iden-tified as Halomonas maura. One of the potential sources for screening bacteria is plant endophytic bacteria. In this study, we isolated and screened endophytic bacteria for transformation of ginsenoside Rb1 of Ngoc Linh ginseng collected at village number 2, Tra Linh commune, Nam Tra My district, Quang Nam province. There are 45 strains isolated form rhizobium (24 strains), petioles (8 strains) and leaves (13 strains). After screening, there are 27 strains positive with β-glucosidase test, an enzyme which catalyses the hydrolysis of terminal non-reducing residues in β-glucosides-aglycone linkage of ginsenoside Rb. In this study, a modified procedure for catalase from bovine liver and its properties were reported. Bovine liver catalase was purified to electrophoretic homogeneity as a single protein band around 60 kDa on SDS-PAGE by acetone fractionation, followed by ion-exchange chromatography on DEAE-Sepharose and CM-Sepharose columns. The specific activity of the purified catalase was 79,277 units per mg of protein (U/mg) with 1.87% recovery and purification fold was roughly 60 times. The catalase was a homo-tetramer with a molecular mass of about 240.987 kDa as determined by native gel electrophoresis. The purified enzyme showed the highest activity at pH 7, 37°C, and remained active over a broad range of pH from 5 to 10 and range of temperature from 4°C to 40°C. Its activity was inactivated by incubating in 60°C for 30 min. The activity of the enzyme was induced by Ca2+ and inhibited by Na+, Ni2+, Cu2+, Zn2+, Fe2+, Fe3+ and NaN3. In order to diversify antigen sources for vaccine production to against PRRSV, co-expressing LTB and PRRSV GP5- an important structure protein GP5 of PRRSV were expressed in tobacco. Genes encoded for GP5 of PRRSV isolated from pigs infected PRRSV in Vietnam in 2010 and was constructed in plant expression vector under the control of seed specific phaseolin promoter and constitutive promoter 35S. The genes were then transferred into the C9-1 tobacco using Agrobacterium. The stable insertion of LTB-GP5genes in genomeoftransgenic tobacco was confirmed by PCR and RT-PCR. Western hybridization indicated that fused protein LTB-GP5 was successful expressed in tobacco seeds. Analysis of transgenic tobaco seeds revealed that under regulation of promoter beta-phaseolin, the level expression of GP5 was significant improved as quantified by ELISA (2.2 % of total soluble protein). These fatty acids play an important role on human health. In this article, we presented results of safety assessment and pharmacological effect of Algal Oil Omega 3-6 (AOO-3-6) which made from oil of S. mangrovei TB17 on the experimental animals after 90 days of use. The results of acute toxicity study showed that by oral administration of the AOO-3-6 on white mice at the highest dose of 15.27 g total of DHA, EPA, DPA / kg of whole body of mice/ day (the dosage was 113.63 times higher than the mice dose), the LD50 was not determined. It means that AOO-3-6 capsules was no signs toxicity. At doses of 78.4 mg and 352.8 mg of total of DHA, EPA, DPA/ kg of whole bode of rat/ day (effec-tive dose znd 4.5 times higher than the expected dose) for 90 continuous days on the mice didn’t affect the normal increment of body weight, hematological indices, biochemical indicators; function of liver and kid-ney; histopathology of liver, kidney, spleen of white rats. At the same time, at doses of 134.4 and 403.2 mg total of DHA, EPA and DPA/kg of body weight/day improved the ability to learn, remember on the mice. In addition, it also allowed building its reflexes sustainability in finding food to be faster with shorter reflectance time. Fosb is a tree native of Vietnam, this medicinal leaf has a high viscosity that has been used to heal wounds, heal the sores, stop bleeding... Polysaccharide has attracted great attentions for its benefits to human health. Polysaccharide from natural sources have diverse anti-inflammatory, anticoagulant and wound healing activities. Polysaccharide is not only valuable in medicine, also widely used in foodstuffs such as gel thickening or emulsifying agents, emulsifiers, fillers. The polysaccharide content in P. carruthersii var atropurpureum leaves was (11.63 ± 0.375)% in dry weight. The appropriate polysaccharide extraction conditions were determined: water, material/solvent ratio (1g/25ml), extracted temperature of 60°C, extraction time 15 hours. In this study, the abundance and life cycles (lysogeny and lytic) of viruses associated with contrasted coral health states (healthy and bleached) were studied and compared. In addition, the density of bacteria and Symbiodium were also be determined by epifluorescence microscopy. The results showed that viral abundance increased 1.2 times and bacterial abundance decreased 0.6 times in healthy corals compared to bleached ones. The number of Symbiodinium significantly reduced 5.6 times in bleached corals compared to healthy corals. One of the most important results in this study was the coexistence of the both lysogenic and lytic viruses in healthy coral. The fraction of lysogenic bacteria was 2 times higher in healthy corals than in bleached ones. Healthy corals carried 3.5-fold more viral production than the diseased ones. Overall, the results showed that, in healthy state, corals might be a favorable environment for viral infection, promoting the development of both temperate and virulent phages. Lam is an important food crop in the world as well as in Vietnam. Despite its many benefits, the production of sweet potato is restricted in many areas of the world by diseases, weed, and, particularly, pests. As an alternative, genetic transformation provides the means for complementing conventional breeding to improve sweet potato to resistant to pest. In this study, shoot tip explants of KB1 sweet potato variety were infected with A. tumefaciens C58 carying pBI101/cry3Ca1 con-struct. The selection were occured on callus producing medium (SM) containing 0.5 g/L picloram, 50 mg/L kanamycin and 500 mg/L cefotaxime. Survival embryogenic calli were then transferred to embryo producing medium (EG2) supplemented with 1.0 mg/L ABA and 1.0 mg/L GA3 after 2–3 weeks. Putative transgenic shoots regenerated on medium (RM) adding 0.5 mg/L kinetin and 1.0 mg/L BAP were rooted on root producing medium (RR). The tentative transgenic lines were proved positively by PCR and finalized by Southern hybridization, and biotest in laboratory. Conclusionly, we obtained 62 tentative transgenic sweet potato lines resistant to kanamycin. Among these lines, five putative transgenic lines were proved positively by Southern hybridization and have one copies of the cry3Ca1 gene.
