TCGA verileri kullanılarak pankreas kanserinde lncRNA H19 ve UCA1 ifadeleri değerlendirilmiştir. Bu lncRNA’ların ve invazyon genlerinin ifadeleri üzerine etkileri qRT-PZR ile belirlenmiştir. \u0000Bulgular: PANC-1 hücrelerinde borik asitin IC50 dozu 14248,8 µM, MIA PaCa-2 de 15707,5 µM olarak hesaplanmıştır. TCGA verileri H19 ve UCA1 ifadelerinin pankreas kanserinde yüksek olduğunu göstermiştir. Borik asit muamelesi ile H19 ve UCA1 lncRNA seviyeleri azalmıştır. Ayrıca borik asit iki hücre hattında CDH1 ve TIMP1 ifadesini arttırmıştır. CDH2 ifadesi ise baskılanmıştır. Borik asit PANC-1 hücrelerinde TIMP2, MIA PaCa-2 hücrelerinde TIMP3 ifadesini arttırmıştır. İnvazyon testinde borik asit her iki hücrede invazyonu anlamlı derecede baskılanmıştır. \u0000Sonuç: Borik asit pankreas kanseri hücrelerinde ifadesinin yüksek olduğu görülen H19 ve UCA1 ifadelerini baskılamıştır. Bozkurt, Hülya Gökmen","doi":"10.26453/otjhs.1161706","DOIUrl":"https://doi.org/10.26453/otjhs.1161706","url":null,"abstract":"Objective: The study aimed to evaluate the effect of cycloplegia on the ocular biometric parameters and intraocular lens (IOL) power calculated by Sanders–Retzlaff–Kraff/theoretical (SRK/T) formula in the cataractous eyes. \u0000Materials and Methods: This cross-sectional study included 68 senile cataractous eyes of 68 patients scheduled to undergo cataract surgery. Measurements of anterior chamber depth (ACD), axial length (AL), white to white (WtW) diameter, keratometry (K1, K2, Kmean), central corneal thickness (CCT), and pupil size (PS) were obtained with AL-Scan (Nidek Co., Ltd, Gamagori, Japan) before and after cycloplegia. The SRK/T formula was used to calculate IOL power with target refraction of 0 D. Cycloplegia induced by cyclopentolate hydrochloride %1. \u0000Results: The mean age of the patients was 70.60±4.07 years (range 65 to 80). A significant increase was observed in ACD after cycloplegia (p< 0.001). There was no statistically significant difference between pre-dilation and post-dilation AL, WtW, CCT, K1, K2, Kmean, and IOL power readings (p> 0.05). Thiol groups which have high antioxidant capacity, turn to disulpide (DS) groups with biochemical reactions that neutralize different oxidant compounds. Thiol/Disulpide homeostasis (TDH) has significant effects on cell mechanisms, transcription and apoptosis. We present the role of prognosis of dynamic TDH in patients operated for NSCLC. \u0000Methods: The patients who operated for early stage with NSCLC prospectively analysedTDH tests were measured by the automated spectrophotometric method. \u0000Results: 77 patients patients, 61 volunteer controls participated in this study. Nanoparticle size and distribution were evaluated by Dynamic light scattering (DLS). Cytotoxicities of NPs were evaluated in MDA-MB-231 and MCF-7 breast cancer cells and HUVEC using MTT test and their morphological images were compared. \u0000Results: While the average size of CP-NPs was around 2-3 nm, Cis-NPs was 7-8 nm. It was observed that both NPs groups were homogeneously dispersed. According to the cytotoxicity results, both CP-NPs and Cis-NPs were more cytotoxic on MCF-7 breast cancer cells. In addition, CP-NPs and Cis-NPs showed significant cytotoxicity on MCF-7, MDA-MB-231 breast cancer cells, while low cytotoxicity was detected in normal HUVEC cells. The study was completed with the data obtained from the records of 58 cases with a mean age of 69.93±6.24 years. Visual Analog Scale, Hospital for Special Surgery Knee Score Hospital Anxiety and Depression Scale were used. \u0000Result: When the measurement scores were compared according to gender, no statistically significant difference was found in pain(p:0.49) and physical function(p:0.20). When anxiety-depression levels were compared, it was found that females got higher scores than males(p:0.00). The data were collected using the Questionnaire Form, the UCLA Loneliness Scale (Version 3), the Multidimensional Scale of Perceived Social Support (MSPSS), and the Scale for Readiness of Pregnant Women to Hygienic Care of the Newborn (SRPWHCN). Descriptive statistics, ANOVA test, independent sample t-test, and Pearson correlation analysis were used to evaluate the research data. \u0000Results: The mean age of the pregnant women was 25.75±4.64. {"title":"Secondary Bacterial Agents and Antibiotic Resistance Profiles in Respiratory Tract Specimens of Patients with COVID-19 Pneumonia","authors":"Oya Akkaya, Ayşenur Uğur, Habibe Övet, Feyza Alp, Mustafa Önder Gönen","doi":"10.26453/otjhs.1094238","DOIUrl":"https://doi.org/10.26453/otjhs.1094238","url":null,"abstract":"ABSTRACT \u0000Objective: The aim of the study is to determine which bacteria are the causative agents and the antibiotic resistance rates in the respiratory tract secretions of patients who have diagnosed with COVID-19 and to compare these results with before the pandemic. \u0000Method: This study is a retrospective study comparing bacteria grown in bronchoalveolar lavage (BAL) fluid and endotracheal aspirate(ETA) fluids coming from adult clinical services and intensive care units and their antibiotic resistance profiles between January 2019 and December 2019 before the pandemic, with the results between January 2020 and December 2020 during the pandemic period. \u0000Results: A total of 900 culture results were examined in 2019; Acinetobacter baumannii was detected in 36%, Klebsiella pneumoniae in 23%, Pseudomonas aeruginosa in 14%, and Staphylococcus aureus in 8%. In 2020, during the pandemic, 660 culture results were examined, and same bacteria was detected in 43%, 23%,16% and 5%, respectively. K.pneumoniae's 3rd generation cephalosporin resistance, A. baumannii's resistance to tigecycline, gentamycin, tobramycin and P. aeruginosa's resistance to ceftazidime were found to have increased significantly during the pandemic compared to the pre-pandemic period. \u0000Conclusion: In hospitalized patients with COVID-19, secondary bacterial agents are mostly multidrug-resistant Gram-negative bacteria. A Descriptive Information Form, the Domestic Violence Scale and Beck Depression Inventory were used as the data collection instruments. In the statistical analysis, frequencies, percentages, means, ANOVA, t-test, correlation analysis and regression analysis were utilized. \u0000Results: The mean domestic violence and depression scores of the women were found to be low. There was a positive and significant relationship between domestic violence and depression. Factors such as the age of the woman and her spouse, status of being subjected to violence, employment status, educational level and form of marriage were determined as factors that were effective on domestic violence and depression. \u0000Conclusion: Domestic violence is a significant factor leading women to experience depression. Then the interactions of each of the receptor and drug molecules at the binding site were examined by molecular docking studies, which is a computer-aided drug design method, and their binding affinities were evaluated. \u0000Results: As a result of the analyses, it was determined that the drug named Hydroxychloroquine has the highest and the drug called Dexamethasone has the lowest binding affinity for all three receptors.
