These diseases are defined clinically through inflammatory disorder in gastrointestinal tract accompanied with diarrhea, abdominal pains, weight loss, nausea and pathological changes such as a loss of mucosal integrity and inflammatory cell infiltration (Xavier et al., 2007). In addition, the presence of white blood cells. at the intestinal mucosal tissue is characteristic feature of this disease (Wang et al., 2005). Colitis affects the distal colon portion and induces non-transmural inflammation, massive necrosis of mucosal and Colitis is a chronic disease that affects individuals at different ages and affects the quality of their life. So far, there are three different experimental models for colitis. However, animals in these models die quickly duo to the associates toxicity. The aim of this study is to optimize acetic acid induced acute colitis mouse model with olive oil co-administration. Male albino CD1 mice (n= 91) were randomly divided into 13 groups (n= 7/group). To induce colitis, mice were administrated with different volumes (100 or 200 μl) and concentrations (1%, 2% and 4%) of acetic acid by intrarectal (IR) injection either for three consecutive days or every other day for a total of 6 days. Olive oil (1 μL) was co-injected with acetic acid. One day later, mice were sacrificed and colon tissues were fixed in 10% formalin for histological examinations. The induction of colitis by acetic acid was dependent on both the volume and the concentration of acetic acid. Interestingly, co-administration of olive oil with different volumes and concentrations of acetic acid, although did not alter the severity of inflammation, it increased the animal survival rate and decreased the associated toxicity. Injection of co-administration of olive oil with acetic acid is a new modification to enhance the application of this model. Total phenolics content, flavonoids, anthocyanins, saponins, total antioxidant capacity (TAC) and 2,2-diphenyl-1picrylhydrazyl (DPPH) scavenging activity were assessed in their extracts. In vitro anticancer screening of the hydro-alcohol extracts was also assessed using human hepatocellular carcinoma (HepG-2) and breast adenocarcinoma (MCF-7) cell lines. The plant species revealed different metabolomic profiling. C. oxyphyllus showed the highest phenolic and flavonoids contents compared to other plant extracts. While, among these plant extracts, T. polium showed the highest level of TAC, saponins and anthocyanins contents. C. oxyphyllus showed the highest inhibition concentration 50% (IC50) against HepG-2 (18.8 μg/ml) and MCF-7 (4.1 μg/ml). The high-performance liquid chromatography ‎analysis of C. oxyphyllus extract revealed the presence of high content of benzoic acid and vanillic acid (phenolics) along with hesperidin (flavonoids). Several studies suggested that individuals with external locus of control (E-LOC) have more symptoms of depression. Other studies indicated to the effective role of psychological status on the immune system. The association between E-LOC and immunity in cancer patients it is not clear. Study the relation between Rotter internal-external locus of control (I-E LOC) and God health locus of control (GHLC) and interleukin-6 (IL-6) in breast cancer patients. The study included hundred breast cancer patients and fifty healthy controls. Patients and participators responded to I-E LOC and GHLC questionnaires. Peripheral blood samples were collected to measure gene expression of IL6 by using real time PCR. Breast cancer patients showed higher GHLC which was associated with higher level of IL-6 as compared to control subjects. By contrast, a significant inverse correlation between GHLC and IL-6 was found. However, there was no relationship between I-E LOC and IL-6. The results suggest no significant effect of LOC belief according to life on immune response against cancer, but high GHLC belief correlate with decreased IL-6. Further studies on the relations between other types of LOC and immune response in breast cancer is recommended. Alabssawy","doi":"10.21608/jcbr.2019.34587","DOIUrl":"https://doi.org/10.21608/jcbr.2019.34587","url":null,"abstract":"Crude Tetrodotoxin (TTX) extracts of liver, skin and muscles which isolated from Puffer fish, Lagocephalus sceleratus (L. sceleratus) were tested for their antimicrobial activity besides TTX levels (µg/g) using LC–MS–MS (Liquid Chromatograph / mass spectrometry) in cold and warm season and TTX identification by HPLC (High Performance Liquid Chromatograph), TLC (Thin Layer Chromatograph) and UV spectrum (Ultra Violet spectrophotometer). The results obtained showed interesting antibacterial and antifungal activity against Crude extracts of TTX isolated from liver, skin and muscles of L. sceleratus. The crude TTX of liver extract showed quite promising broad-spectrum antibacterial activity due to its capacity to inhibit the growth of almost bacterial tested strains. It has shown the maximum activity against Escherichia coli as 22.24 ± 0.96 in gram negative bacteria and Bacillus subtilis as 17.51 ± 0.51 in gram positive bacteria and minimum activity against Vibiro cholerae as 9.22 ± 0.65. The maximum antifungal activity was observed to crude TTX of liver extract against Aspergillus fumigatus as 19.21 ± 0.59 and minimum activity against Trichophyton rubrum as 5.33 ± 0.52 in crude TTX of skin extract. Changes in the levels of TTX in the livers, skins and muscles as a function of season and sex were determined. The highest TTX levels in livers, skins and muscles of female fish were 30.33, 3.38 and 2.26 µg/g, respectively in winter. The TTX level in the muscle of male fish in summer was 0.66µg/g but was otherwise below the toxic limit. Consequently, it can be dangerous to consume puffer fish, including the edible muscle, from Mediterranean Sea.
