In the present study, we examined a method for warming and dilution of vitrified blastocysts that would provide simple and highly stable results for practical porcine embryo transfer in the field. Although we have already succeeded in obtaining piglets using this simple method whereby blastocysts are vitrified using micro volume air cooling (MVAC) then warmed and diluted in a syringe to remove the cryoprotectant, the rate of miscarriage was as high as 40%. Using the conventional method employing microscopy in the laboratory, the pre-warming temperature and volume of the porcine warming and dilution solution (PWDS) for vitrified embryos were 45℃ and 3 m l , respectively. In order to facilitate the transfer of porcine embryos in the field, it is necessary to develop a simplified method based on opti-mization of the dilution conditions. For this purpose, changes in the temperature of PWDS under various conditions were investigated. An individual vitrification device without embryos was taken out from LN 2 followed by insertion into a syringe filled with 1.5 m l , 2 m l or 3 m l of PWDS. These syringes were held for 3 min in room air at 15℃ or 27℃, or in water at 38℃, then the PWDS temperature in each case was recorded. A significant decrease in temperature was observed for all volumes in room air temperature, being approximately 8 - 10℃ at 15℃ and 4 - 6℃ at 27℃. On the other hand, there was very little decline in the water temperature at 38℃; all volumes of PWDS showed a drop of approximately 0.1℃. Based on these results, pregnancy was examined after transfer of vitrified embryos diluted with 1.5 or 3 m l of PWDS warmed at 39℃. Between 12 and 16 blastocysts were transplanted surgically into the uterus of 5 recipients each assigned to each PWDS volume group, and 4 of the recipients each became pregnant and delivered a total of 19 and 17 piglets respectively. The farrowing rates of group of 1.5 and 3 m l were 80% and 60%, and the piglet production rates relative to the number of transplanted embryos were 23.6% and 27. 5%, respectively. Fertility and piglet production were equivalent, with no significant difference between the two volumes of PWDS. However, when the PWDS was kept warm, the ratio of pregnancy loss improved to 0% (1.5 m l ) and 25.0% (3 m l ), compared with 40% with no warming. These results suggest that when warming and diluting blastocysts in the syringe with an individual vitrification device inserted, optimal results are obtained by keeping the PWDS at 39℃. In addition, good results can be obtained when the volume of PWDS is 1.5 m l or 3 m l .","PeriodicalId":274137,"journal":{"name":"Nihon Yoton Gakkaishi","volume":"70 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2020-12-22","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"114476589","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Blastocysts and expanded blastocysts were collected from Duroc and were vitrified using the micro volume air cooling (MVAC) method. Here, 35 Landrace and Duroc Gilts (8 - 10 months-old) were used as recipients. We investigated three types of devices, including commercially available deep implant device (Device A group), deep implant device prototype (Device B group), and modified bovine embryo transfer device (Device C group). In Test 1, the position of each device’s tip was confirmed by laparotomy under general anaesthesia following insertion into the recipient to confirm the transplantation site, and then the vitrified embryos were transferred. Each test group had 5 recipients. The number of transferred embryos per recipient was 14 - 16, corresponding as the average number of ovulated embryos per litter. In Test 2, each device was inserted into a non-anesthetized standing recipient, and then the vitrified embryos were transferred. Each test group used had 5 recipients. The number of transferred embryos per recipient was 15 - 17 embryos. In Test 1 of the A group, one recipient’s uterus was accidentally penetrated by insertion of the device, and as a result, embryo transfer was not performed. In other recipients, the device tip positions were confirmed at the uterine body or near the uterine horn junction. Two out of four recipients were pregnant and successfully delivered, and a total of eight piglets were born in the A group. In the Device B group, the tips of the devices were positioned near the uterine horn junction in all the recipients. Three out of five recipients were pregnant and successfully delivered, producing a total of 13 piglets. In the Device C group, the tips of the devices were positioned near the cervical outlet of the uterine body in all the recipients. None of the five recipients were pregnant. The result of Test 2 in the Device A group indicated that three out of five recipients were pregnant, but two miscarried. The remaining recipient delivered five piglets. In the Device B group, three out of five recipients became pregnant and delivered a total of 16 piglets. In the Device C group, three out of five recipients became pregnant and delivered a total of 14 piglets. These results show that non-surgical transfer of porcine vitrified embryos into the uterine body of a standing recipient without anaesthesia administration had the same efficiency as surgical embryo transfer in terms of piglet production rate. A total of 48 crossbred pigs (Large White × Landrace × Duroc, six barrows, and six gilts) were housed at a density of four pigs (two barrows and two gilts of similar weight) per pigpen. The pigs were divided into four groups each of which MoR was added to feed at levels of 0% (Control group), 3% (MoR3% group), 6% (MoR6% group) and 12% (MoR12% group). The pigs were fed with experimental diets when the average weight of the pigs in each group reached approximately 50 kg and was terminated when the average weights reached approximately 110 kg. Feed and drinking water were provided ad libitum . For the growth performance examined for twelve pigs in each group, the MoR 12% group showed a significantly higher feed intake than the control group (P < 0.05). No significant differences were detected in other growth performance. For the frequency of water intake investigated for eight pigs in each group, the MoR 12% group showed a significantly higher frequency of water intake than the control group (P < 0.05). For the carcass characteristics investigated for the following numbers of pigs (control: nine pigs, MoR3% : eleven pigs, MoR 6%, 12% : twelve pigs), carcass length in gilt was significantly shorter in the 6% group than the 3% group (P < 0.05), and that in barrow was significantly shorter in the 6% group than the control group (P < 0.05). For meat quality examined for three pigs (two barrows, one gilt) in each control, MoR3% and 6% group, no difference was observed. Our results suggested that MoR ≤ 6% is appropriate when feeding MoR to growing pigs without increasing the frequency of water intake, considering the growth performance, carcass characteristics and meat quality.","PeriodicalId":274137,"journal":{"name":"Nihon Yoton Gakkaishi","volume":"23 18 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2020-09-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"126211842","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The aim of this study was to compare pork pH values measured by using ISFET electrode without temperature compensation with conventional glass electrode. Porcine longissimus muscles harvested from 32 carcasses were subjected to pH measurement at 4 day after slaughter. ISFET pH values were significantly higher than glass electrode pH value of 5 . 66. The least squares means of ISFET pH was 6 . 11 at the surface of pork sample and 6 . 14 at the inner part of muscle. Although there were some statistical significant correlations between ISFET and glass electrode pH values, correlation coefficients between them were too low for the estimation of glass electrode pH value by ISFET pH value. These results clearly demonstrated that pH values measured by ISFET electrodes without temperature compensation become higher than those of glass electrode. Thus, appropriate cor-rections of values, e.g. temperature compensation and estimation using regression, would be necessary when using ISFET electrode for pork pH measurement. J.
